Pumping test of a water table aquifer is carried out to estimate the aquifer parameters, the obtained data were analysed through the solution of both Neuman(1975) and Jacob(1946) methods through AQTESOLV and Spreadsheet programs, the results of each methods are compared to evaluate the applicability and accuracy of the solution theoretically and practically. In the paper an example is presented, which is conducted for a constant rate pumping test from Ohio, in Fairborn(near Dayton), and it supplied by S.E. Norris(U.S. Geological Survey, Columbus, Ohio). The main objective of this study is to introduce both program and the way of the applications, and compare the results and the hand on of both programs in the field.

Developing a practical means of reducing alloimmunization in chronically transfused patients would be of significant clinical benefit. Immunocamouflaging red blood cells (RBCs) by membrane grafting of methoxypoly(ethylene glycol) (mPEG) may reduce the risk of allo-immunization. The results of this study showed that antibody recognition of non-ABO antigens was significantly reduced in an mPEG-dose- and polymer size-dependent manner, with higher molecular weight mPEGs providing better immunoprotection. Furthermore, in vivo immunogenicity was significantly reduced in mice serially transfused with mPEG-modified xenogeneic (sheep; sRBCs), allogeneic (C57Bl/6), or syngeneic (Balb/c) RBCs. Following a primary transfu- sion of sRBCs, mice receiving mPEG-sRBCs showed a >90% reduction in anti-sRBC IgG antibody levels. After two transfusions, mice receiving mPEG-sRBCs showed reductions of >80% in anti-sRBC IgG levels. Importantly, mPEG-modified autologous cells did not induce neoantigens or an immune (IgG or IgM) response. These data suggest that the global immunocamouflage of RBCs by polymer grafting may provide a safe and cost-effective means of reducing the risk of alloimmunization.

Alcohol use disorder(AUD) is a global health concern associated with several *** health problems related to AUD, such as cognitive deficits, have been linked to *** use has been associated with changes in neuroimmune activity, although current literature has yielded mixed results. For example, markers of gliosis, including translocator protein 18-kDa(TSPO), proinflammatory cytokines, glutamate(Glu), and myo-inositol(mI), are disrupted in the alcohol-dependent brain. Further, neuroinflammatory-related phenomena including membrane turnover, blood brain barrier(BBB) permeability, and adenosine release have also shown alterations in AUD. However, current literature remains inconclusive about the directionality of these changes. Both in vivo and in vitro studies have provided insight on the relationship between alcohol use and neuroinflammatory processes, suggesting considerable treatment potential for alcohol use disorder and its inflammatory comorbidities. Here, we review current neuroimaging literature assessing the impacts of alcohol use on neuroimmune activity in the brain.

Bone remodeling is a tightly regulated resorption and formation of bone matrix for physiological processes or to maintain bone function. Bone remodeling involves the synchronized differentiation and activity of bone-related cell types including bone matrix-depositing osteoblasts, bone matrix-resorbing osteoclasts, collagen/extracellular matrix-producing chondrocytes, and the progenitors of these cell types. T and B cells are adaptive immune cells that can influence bone remodeling by directly regulating the function of bone-related cells under normal and pathophysiological conditions. The specific mechanisms through which T cells control remodeling are not well defined. Here, we review the impact and influence of T cells and their products on the differentiation and function of bone cells during bone remodeling. Synthesizing new connections and highlighting potential mechanisms may promote additional avenues of study to elucidate the full role that immune cells play in regulating bone homeostasis.

Immune checkpoint inhibitors(ICIs) are monoclonal antibodies that target downregulators of the anti-cancer immune response: Cytotoxic T-lymphocyte antigen-4, programmed cell death protein-1, and its ligand programmed death-ligand *** have revolutionized the treatment of a variety of malignancies. However,many immune-related adverse events have also been described which mainly occurs as the immune system becomes less suppressed, affecting various organs including the gastrointestinal tract and causing diarrhea and colitis. The incidence of immune-mediated colitis(IMC) ranges from 1%-25% depending on the type of ICI and if used in combination. Endoscopically and histologically there is a significant overlap between IMC and inflammatory bowel disease,however more neutrophilic inflammation without chronic inflammation is usually present in IMC. Corticosteroids are recommended for grade 2 or more severe colitis while holding the immunotherapy. About one third to two thirds of patients are steroid refractory and benefit from infliximab. Recently vedolizumab has been found to be efficacious in steroid and infliximab refractory cases. While in grade 4 colitis, the immunotherapy is permanently discontinued, the decision is controversial in grade 3 colitis.

In Slovakia, a direct disposal of spent nuclear fuel in a deep geological repository within the country after a certain period of interim storage is a preferred option. This paper briefly describes near field model of radionuclide migration developed in GoldSim simulation code environment and analyses the calculated results on time-dependent release rates of safety relevant radionuclides. Given the fact that GoldSimalso enables to perform probabilistic simulations using the Monte Carlo method, a probabilistic approach was chosen to assess the influence of selected near field parameter uncertainties related to radionuclide migration on the radionuclide release rates from the bentonite buffer to the surrounding host rock. Based on the results, release rates of nuclides which exceed their solubility limits are effectively lowered and many of nuclides are significantly sorbed on the buffer material. It can be seen that the variance of the total release rate in the case of solubility uncertainty is almost two orders of magnitude within a long period of time.

In the wake of ever-increasing environmental pollution, human skin in the modern urban world is exposed to increased levels of harmful environmental pollutants. Many studies have shown that these pollutants can weaken the epidermal skin barrier and thus facilitate the penetration of these substances into the skin. An important goal of modern skin care against harmful environmental influences should therefore be to protect and strengthen the epidermal barrier and to repair occurring damage quickly and efficiently. With this in mind, in the present study we investigated what damage cigarette smoke causes to the epidermal barrier and 1) whether the regular application of a O/W emulsion (Day Cream) can protectively strengthen the epidermal barrier against environmental damage and 2) whether a cigarette smoke-induced disruption of the epidermal barrier is restored faster and better by the regular application of a another O/W emulsion (Night Cream) than in product-untreated skin. The two products are slightly different in plant-oil, active ingredient composition and texture. Firstly, the study has shown that the Lipbarvis? method is suitable for measuring the effect of cigarette smoke, in contrast to conventional biophysical measurement methods (transepidermal water loss, skin hydration). Secondly, both products were able to improve skin barrier function in the corresponding test scenario. This was demonstrated for both test products by a significantly reduced TEWL, significantly increased skin hydration and significantly improved length of the lipid lamellae in the intercellular space, as well as the protective effect of the day cream on the epidermal skin barrier and the regenerative properties of the tested night cream. .
