TAU is a microtubule-associated protein that promotes microtubule assembly and stability in the *** is missorted and aggregated in an array of diseases known as *** are essential for neuronal function and regulated via a complex set of post-translational modifications,changes of which affect microtubule stability and dynamics,microtubule interaction with other proteins and cellular structures,and mediate recruitment of microtubule-severing *** impairment of microtubule dynamics causes neuronal dysfunction,we hypothesize cognitive impairment in human disease to be impacted by impairment of microtubule *** therefore aimed to study the effects of a disease-causing mutation of TAU(P301L)on the levels and localization of microtubule post-translational modifications indicative of microtubule stability and dynamics,to assess whether P301L-TAU causes stability-changing modifications to *** investigate TAU localization,phosphorylation,and effects on tubulin post-translational modifications,we expressed wild-type or P301L-TAU in human MAPT-ko induced pluripotent stem cell-derived neurons(i Neurons)and studied TAU in neurons in the hippocampus of mice transgenic for human P301L-TAU(p R5 mice).Human neurons expressing the longest TAU isoform(2N4R)with the P301L mutation showed increased TAU phosphorylation at the AT8,but not the p-Ser-262 epitope,and increased polyglutamylation and acetylation of microtubules compared with endogenous TAU-expressing neurons.P301L-TAU showed pronounced somatodendritic presence,but also successful axonal enrichment and a similar axodendritic distribution comparable to exogenously expressed 2N4R-wildtype-TAU.P301L-TAU-expressing hippocampal neurons in transgenic mice showed prominent missorting and tauopathy-typical AT8-phosphorylation of TAU and increased polyglutamylation,but reduced acetylation,of microtubules compared with non-transgenic *** sum,P301L-TAU results in changes in micr

Collagen is an endogenousfluorophore that accounts for about 70%of all proteins of human skin,so it can be an optical marker for structural abnormalities in tissues registered by laserfluorescent diagnostics in *** the examples of such abnormalities as scars,scleroderma and basal cell carcinoma,this study shows the differences between coefficients offluorescent contrast k_(f)(λ)of abnormalities from the ones for healthy tissues atfluorescent excitation wavelength 360380 *** is shown that scars and dysplasia are characterized by reduced values of k_(f)(λ)for *** to high turbidity and phase heterogeneousness as well as variation of parameters of blood microcirculation and concentrations of other related chromophores,there is no mathematical model that precisely calculates the concentration of collagen in tissues only with the use of the value offluorescent signal ***,probably,the best marker of the pathological process is a comprehensive representation of k_(f)(λ)for all endogenousfluorophores,i.e.,for all used visible *** this case identification of abnormal tissues is quite possible by detecting some deviations of coefficients k_(f)(λ)for the optically identical and symmetrical regions of the human body.

NECAB family of neuronal calcium-binding proteins in health and disease

The N-terminal EF-hand calcium-binding proteins 1–3(NECAB1–3) constitute a family of predominantly neuronal proteins characterized by the presence of at least one EF-hand calcium-binding domain and a functionally less well characterized C-terminal antibiotic biosynthesis monooxygenase domain. All three family members were initially discovered due to their interactions with other proteins. NECAB1 associates with synaptotagmin-1, a critical neuronal protein involved in membrane trafficking and synaptic vesicle exocytosis. NECAB2 interacts with predominantly striatal G-protein-coupled receptors, while NECAB3 partners with amyloid-β A4 precursor protein-binding family A members 2 and 3, key regulators of amyloid-β production. This demonstrates the capacity of the family for interactions with various classes of proteins. NECAB proteins exhibit distinct subcellular localizations: NECAB1 is found in the nucleus and cytosol, NECAB2 resides in endosomes and the plasma membrane, and NECAB3 is present in the endoplasmic reticulum and Golgi apparatus. The antibiotic biosynthesis monooxygenase domain, an evolutionarily ancient component, is akin to atypical heme oxygenases in prokaryotes but is not wellcharacterized in vertebrates. Prokaryotic antibiotic biosynthesis monooxygenase domains typically form dimers, suggesting that calcium-mediated conformational changes in NECAB proteins may induce antibiotic biosynthesis monooxygenase domain dimerization, potentially activating some enzymatic properties. However, the substrate for this enzymatic activity remains uncertain. Alternatively, calcium-mediated conformational changes might influence protein interactions or the subcellular localization of NECAB proteins by controlling the availability of protein–protein interaction domains situated between the EF hands and the antibiotic biosynthesis monooxygenase domain. Behavior of an indigenously fabricated transferred arc plasma furnace for smelting studies

The utilization of industrial solid waste for metal recovery requires high-temperature tools due to the presence of silica and alumina, which is reducible at high temperature. In a plasma arc furnace, transferred arc plasma furnace（TAP） can meet all requirements, but the disadvantage of this technology is the high cost. For performing experiments in the laboratory, the TAP was fabricated indigenously in a laboratory based.on the different inputs provided in the literature for the furnace design and fabrication. The observed parameters such as arc length, energy consumption, graphite electrode consumption, noise level as well as lining erosion were characterized for this fabricated furnace. The nitrogen plasma increased by around 200 k（200 ℃） melt temperature and noise levels decreased by ~10 d B compared to a normal *** plasma offered 100 k（100 ℃） higher melt temperature with ~5 d B higher sound level than nitrogen plasma. Nitrogen plasma arc melting showed lower electrode and energy consumption than normal arc melting, whereas hyd.ogen plasma showed lower energy consumption and higher electrode consumption in comparison to nitrogen plasma. The higher plasma arc temperature resulted in a shorter meltdown time than normal arc with smoother arcing. Hyd.ogen plasma permitted more heats, reduced meltdown time, and lower energy consumption, but with increased graphite consumption and crucible wear. The present study showed that the fabricated arc plasma is better than the normal arc furnace with respect to temperature generation, energy consumption, and environmental friendliness. Therefore, it could be used effectively for smelting-reduction studies.

Entropy generation analysis of natural convective radiative second grade nanofluid flow between parallel plates in a porous medium

The present article explores the entropy generation of radiating viscoelastic second grade nanofluid in a porous channel confined between two parallel plates. The boundaries of the plates are maintained at distinct temperatures and concentrations while the fluid is being sucked and injected periodically through upper and lower plates. The buoyancy forces, thermophoresis and Brownian motion are also considered due to the temperature and concentration differences across the channel. The system of governing partial differential equations has been transferred into a system of ordinary differential equations(odEs) by appropriate similarity relations, and a shooting method with the fourth-order Runge-kutta scheme is used for the solutions. The results are analyzed in detail for dimensionless velocity components. The temperature, concentration distributions, the entropy generation number, and the Bejan number corresponding to various fluid and geometric parameters are shown graphically. The skin friction, heat and mass transfer rates are presented in the form of tables. It is noticed that the temperature profile of the fluid is enhanced with the Brownian motion, whereas the concentration profile of the fluid is decreased with the thermophoresis parameter, and the entropy and Bejan numbers exhibit the opposite trend for the suction and injection ratio.
