Proteomic composition of the acrostyle＂ Novel approaches to identify cuticular proteins involved in virus-insect interactions

The acrostyle is a distinct anatomical region present on the cuticle at the inner face of the common food/salivary canal at the tip of aphid maxillary stylets. This conserved structure is of particular interest as it harbors the protein receptors of at least l plant virus, Cauliflower mosaic virus＇, and presumably has other roles in plant-insect interactions. Previously we reported immunolabeling of a highly conserved motif of cuticular proteins from the CPR family （named for the presence of a Rebers and Riddiford consensus） within the acrostyle. Here we report the development of novel tools to further study the proteomic composition of this region and to identify proteins involved in insect- virus interactions. Using a series of antibodies against cuticular proteins from the RR-2 subfamily, we identified additional peptides present within the acrostyle. Our results demonstrated that the acrostyle is a complex structure containing multiple domains of cuticular proteins accessible for interaction. In addition, an array of overlapping peptides,which covers the diversity of the majority of the RR-2 subfamily, was ~tevelopect as a generic tool to characterize cuticular protein/pathogen interactions. Upon probing this array with Cucumber mosaic virus particles, consensus peptide sequences from hybridizing peptides were identified. Use of these novel tools has extended our knowledge of the proteomic composition of insect maxillary stylets and identified sequences that could be involved in virus binding, thus contributing to further elucidation of the various properties and functions of the acrostyle.

The synthesis of ultrathin rhenium disulfide(ReS_(2))nanoribbons within single-walled carbon nanotubes(SWNTs)has been *** decacarbonyl complex is encapsulated into the SWNTs to provide a source of confined rhenium atoms,which readily react with iodine to form discrete nm-sized clusters of rhenium iodide[Re_(6)l_(14)]^(2-) embedded in the *** final step of the synthesis is accomplished by admitting hydrogen sulfide gas into nano test tubes,yielding twisted nanoribbons of rhenium disulfide encapsulated in carbon nanotubes,ReS_(2)@*** width,structure,and composition of rhenium disulfide nanoribbons are strictly controlled by the extreme confinement of the host-SWNT.A holistic analytical approach combining complementary imaging and analysis methods is used at each synthetic step to elucidate the structure and composition of the guest material and reveal the role of the SWNT contributing towards the electronic interactions with encapsulated inorganic *** ReS_(2) nanoribbons are expected to retain the electronic properties of the bulk material,such as direct bandgap,the low dimensional form of this material can be of interest for use in nanoscale electronic devices.

Treatment of a mouse model of collagen antibody-induced arthritis with human adipose-derived secretions

The use of adipose-derived cells as a treatment for a variety of diseases is becoming increasingly common. These therapies include the use of cultured mesenchymal stem cells (MSCs) and freshly isolated stromal vascular fraction (SVF) alone, or in conjunction with other cells such as adipocytes. There is a substantial amount of literature published on the therapeutic properties of MSCs and their secretions as the main driver of their therapeutic effect. However, there is little data available on the therapeutic potential of secretions from SVF, either with or without adipocytes. We investigated the ability of secretions from human adipose SVF alone and the SVF co-cultured with adipocytes as a proxy for cell therapy, to ameliorate an inflammatory disorder. This ethics approved study involved the treatment of collagen antibody-induced arthritis (CAIA) in mice with secretions from SVF, SVF co-cultured with adipocytes, or a vehicle control via both intravenous (IV) and intramuscular (IM) routes. Treatment outcome was assessed by paw volume, ankle size and clinical arthritis score measurements. Serum samples were obtained following euthanasia and analysed for a panel of 32 mouse cytokines and growth factors. The dose and timing regime used for the IM administration of both human secretion mixtures did not significantly ameliorate arthritis in this model. The IV administration of SVF adipocyte co-culture secretions reduced the paw volume, and significantly reduced the ankle size and clinical arthritis score when compared to the IV vehicle control mice. This was a superior therapeutic effect than treatment with SVF secretions. Furthermore, treatment with SVF adipocyte coculture secretions resulted in a significant reduction in serum levels of key cytokines, IL-2 and VEGF, involved in the pathogenesis of rheumatoid arthritis. Therefore, the SVF cocultured with adipocytes is an attractive therapeutic for inflammatory conditions.
