Modular polyketide synthases(PKSs)are a multidomain megasynthase class of biosynthetic enzymes that have great promise for the development of new compounds,from new pharmaceuticals to high value commodity and specialty *** colinear biosynthetic logic has been viewed as a promising platform for synthetic biology for *** to this colinearity,domain swapping has long been used as a strategy to introduce molecular ***,domain swapping often fails because it perturbs critical protein-protein interactions within the *** our increased level of structural elucidation of PKSs,using judicious targeted mutations of individual residues is a more precise way to introduce molecular diversity with less potential for global disruption of the protein *** we review examples of targeted point mutagenesis to one or a few residues harbored within the PKS that alter domain specificity or selectivity,affect protein stability and interdomain communication,and promote more complex catalytic reactivity.

Metabolic engineering efforts toward rewiring metabolism of cells to produce new compounds often require the utilization of non-native enzymatic machinery that is capable of producing a broad range of chemical *** encompass one of the largest classes of chemically diverse natural *** thousands of known polyketides,modular polyketide synthases(PKSs)share a particularly attractive biosynthetic logic for generating chemical *** engineering of modular PKSs could open access to the deliberate production of both existing and novel *** this review,we discuss PKS engineering efforts applied at both the protein and cellular level for the generation of a diverse range of chemical structures,and we examine future applications of PKSs in the production of medicines,fuels and other industrially relevant chemicals.

Metabolic homeostasis requires dynamic catabolic and anabolic processes. Autophagy, an intracellular lysosomal degradative pathway, can rewire cellular metabolism linking catabolic to anabolic processes and thus sustain homeostasis. This is especially relevant in the liver, a key metabolic organ thatgoverns body energy metabolism. Autophagy’s role in hepatic energy regulation has just begun to emerge and autophagy seems to have a much broader impact than what has been appreciated in the field. Though classically known for selective or bulk degradation of cellular components or energy-dense macromolecules, emerging evidence indicates autophagy selectively regulates various signaling proteins to directly impact the expression levels of metabolic enzymes or their upstream regulators. Hence, we review three specific mechanisms by which autophagy can regulate metabolism: A) nutrient regeneration, b) quality control of organelles, and C) signaling protein regulation. The plasticity of the autophagic function is unraveling a new therapeutic approach. Thus, we will also discuss the potential translation of promising preclinical data on autophagy modulation into therapeutic strategies that can be used in the clinic to treat common metabolic disorders.

Plant phenotyping is typically a time-consuming and expensive endeavor,requiring large groups of researchers to meticulously measure biologically relevant plant traits,and is the main bottleneck in understanding plant adaptation and the genetic architecture underlying complex traits at population *** this work,we address these challenges by leveraging few-shot learning with convolutional neural networks to segment the leaf body and visible venation of 2,906 Populus trichocarpa leaf images obtained in the *** contrast to previous methods,our approach(a)does not require experimental or image preprocessing,(b)uses the raw RGb images at full resolution,and(c)requires very few samples for training(e.g.,just 8 images for vein segmentation).Traits relating to leaf morphology and vein topology are extracted from the resulting segmentations using traditional open-source image-processing tools,validated using real-world physical measurements,and used to conduct a genome-wide association study to identify genes controlling the *** this way,the current work is designed to provide the plant phenotyping community with(a)methods for fast and accurate image-based feature extraction that require minimal training data and(b)a new population-scale dataset,including 68 different leaf phenotypes,for domain scientists and machine learning *** of the few-shot learning code,data,and results are made publicly available.

Understanding the maintenance and shift in reproductive strategies is a fundamental question in evolutionary research. Although many efforts have been made to compare different reproductive strategies, the association between reproductive strategies and lineage divergence is largely unknown. To explore the impact of different reproductive strategies on lineage divergence, we investigated the evolution of clonality in Saxifraga sect. Irregulares+Heterisia. by integrating several lines of evidence, we found that the loss of clonality in Irregulares+Heterisia was associated with a progressive increase in diversification rate and intraspecific morphological diversity but with a reduction in species distribution range. Our findings provide insights into the ecological and evolutionary effects of different reproductive strategies,suggesting the necessity of integrating clonality into ecological and evolutional research.

The dispersion and influence of soluble and particulate trace elements(TEs) present in the materials from abandoned mines is an imminent problem on terrestrial and aquatic *** goal of this research is to provide a broader view of such transport,assessing particle size distribution and TE interactions(e.g.,scavenging) with mine tailings and locally derived sediments transported by water in arid *** grains from wash sediment were collected from a dry ephemeral wash in Nelson,Nevada where the Techatticup Mine and Mill worked between 1850 and *** were evaluated to determine the association and behavior of certain metals and metalloids including aluminum,arsenic,barium,cadmium,copper,chromium,iron,manganese,lead,selenium,vanadium,and zinc to rock varnish laminated *** show that some metals and metalloids were more concentrated on larger particles whereas others indicated the opposite *** example,As was greatest on silt fractions and least on coarse fractions,while Se was detected only on ***,Cu,Fe,Mn,Pb,and Zn concentrations all increased with decreasing particle size(silt> sand> coarse),whereas Al,ba,and V showed the opposite trend(silt b,As,and Se were lower in mine tailing sediment treated with wash than those obtained for source *** in treated wash sediment samples,the high EFs of ba,Cr,V,and Mn suggested a possible greater affinity of these metals to the Al,Fe,and Mn oxides from the RV *** data found in this study show that transport of contaminated sediments in arid regions could lead to the enrichment of some TEs in downgradient areas from source materials.
