Epistatic effect between ACACA and FABP2 gene on abdominal fat traits in broilers

Epistasis is generally defined as the interaction between two or more genes or their mRNA or protein products to influence a single trait. Experimental evidence suggested that epistasis could be important in the determination of the genetic architecture of complex traits in domestic animals. Acetyl-coenzyme A carboxylase alpha （ACACA） and fatty acid binding protein 2 （FABP2） are both key factors of lipogenesis and transport. They may play a crucial role in the weight variability of abdominal adipose tissue in the growing chicken. In this study, the polymorphisms of c.2292G〉A in A CA CA and c.-561A〉C in FABP2 were detected among individuals from two broiler lines which were divergently selected for abdominal fat content. Epistasis between the two SNPs on abdominal fat weight （AFW） and ab- dominal fat percentage （AFP） was analyzed. The additive x additive epistatic components between these two SNPs were found significant or suggestively significant on both AFW and AFP in lean lines of the 9th and 10th generation; whereas, it was not significantly associated with either AFW or AFP in fat lines. At the same time, there were not any other significant epistatic components found in both generations or in both lines. Significant epistatic effects between these two SNPs found only in the lean lines could partly be due to the fact that the abdominal fat traits in these two experimental lines have been greatly modified by strong artificial selection. The results suggested that the epistasis mode may be different between the lean and fat chicken lines. Our results could be helpful in further understanding the genetic interaction between candidate genes contributing to phenotypic variation of abdominal fat content in broilers.

Fattiness is an important parameter to estimate meat quality, which has high heritability. In this experiment, F2 chickens derived from Broilers crossing to Silky were used to study the effect of extracellular fatty acid binding protein (EX-FABP) gene on abdominal fat accumulation. Five exons and partial introns of the gene were amplified by six pairs of primers, and then single nucleotide polymorphisms (SNPs) were detected by single strand conformation polymorphism (SSCP) and subsequently confirmed by sequencing. There were four nucleotides variations found, T-C at 659, C-T at 924, CA-TG at 968 and C-T at 1985 respectively. The result of least square analysis suggests that the chickens with HH genotype defined by the third pair of primers have a lower abdominal fat weight and abdominal fat percentage than the chickens with other genotypes (GG, GI, GH and II). Duck prion gene was cloned and sequenced. Similar to mammalian prion protein (PrP), duck prion is encoded by a single exon of a single copy in genome, which was confirmed by Southern blot analysis. All of the structural features of mammalian PrP were also identified in the duck PrP. Compared with mammalian PrP, it exhibited a 30% of general similarity. When compared with chicken PrP, it showed a higher homology of 97%. A phylogenetic tree was constructed to trace evolution of prion gene in animals.

Myostatin, a new member of the TGF-p superfamily, is predominantly expressed in skeletal muscle cells and functions as a negative regulator of skeletal muscle growth in animals. Recently, we have reported three single nucleotide polymorphisms (SNPs) in the chicken my-ostatin gene. Herein, we investigate the association of those SNPs with the production traits in a F2 chicken line derived from Broilers crossing to Silky with the least square analysis. The results show that the BB and AA genotypes are strongly associated with abdominal fat weight (AFW), abdominal fat percentage (AFP), and birth weight (BW) (P < 0.05). Breast muscle percentage (BMP) of the AA type is higher than that of the AB type. The breast muscle weight and breast muscle percentages of F2 individuals have significant difference between CC and DD genotypes (P< 0.05). Breast muscle weight (BMW) of EF birds is higher than that of EE birds (P< 0.05). In this report, we present the first genetic evidence to show that chicken myostatin not only plays an important role in controlling skeletal muscle growth and differentiation, but also may be involved in regulation of adipose growth in chicken.

Study on Characteristics of a Strain of Penicillium *** of Heterotrophic Ammonium Oxidation

An ammonia-oxidizing strain of Penicillium sp., M25-22 which was isolated from chicken feces compost, was studied to make clear its characteristics of ammonium oxidation and provide references for its application in aerobic composting. In media containing （NH4）2SO4 at a concentration of 0.5 g 100 mL^-1, mycelium weight and utilization ratio of ammonium nitrogen increased markedly during the 1st 5 d, concentration of nitrate increased markedly from the 3rd to the 5th d and concentration of nitrite remained at a low level. The strain showed the ability to oxidize ammonium nitrogen growing in media containing glucose, sucrose, starch, or cellulose as the sole carbon source and to oxidize the negative trivalent nitrogen of ammonium sulfate, peptone, acetamide, urea, or L-aspartate. Slowly available carbon or nitrogen sources such as starch, cellulose, and peptone were beneficial to ammonium oxidation. In media containing sucrose as the sole carbon source and ammonium sulfate as the sole nitrogen source, nitrate formation was the most pronounced with sucrose present at a concentration of 12 g L^-1 and ammonium nitrogen present at a concentration of 2.438 mg mL% at the initial pH level of 7.5 and at 30℃.

Landscape Change and its effect on the environment of Daqing City

In this paper, we will study the temporal-spatial landscape change from 1978 to 2001 in Daqing City, based on the mul- ti spectral scanner (MSS) and thematic mapper (TM) images of remote sensing (RS) of 1978, 1988, 1992, 1996 and 2001. In order to explain the effects of the component change on the whole landscape, we make a statistic analysis about the transfer probability matrix of the component types of the landscape by using the ARCGIS platform through the overlay analysis of different landscape components. Based on the transfer probability matrix, we calculate the in-transfer ratio, the out-transfer ratio and the contributor ratio of the special transfer process of the main landscape components. The results illustrate that the cultivated-land, grassland, swamp, and alkaline land are the main contributors resulting in the landscape change: the out-transfer ratios of grassland and swamp are much higher than the in-transfer ones; the in-transfer ratio of alkaline land is much higher than the out-transfer one; the in-transfer ratio of the land-lot is always higher than its out-transfer with the highest rate per year, although there is a small difference among the in-transfers and the out-transfers per year. The main landscape changing processes are the salination of cultivated-land, salination or desertification of grassland, salination and degeneration of swamp. Results in this paper can provide scientific basis for investigating the landscape change mechanism of resources-based city.

Fibroblast growth factor (FGF)-21 signals through both FGF receptor-1 and 2

Fibroblast growth factor (FGF)-21 is a member of the FGF superfamily based on sequence homology. However, unlike most members of this family it does not show any mitogenic activity in all cell types tested. The objective of this study is to identify and characterize receptors for this molecule. Sequencing of the cDNA clones from 3T3-L1 adipocytes indicates that the only isoforms for FGFR-1 and 2 expressed in 3T3-L1 cells are 1IIIc and 2IIIc, respectively, suggesting that FGF-21 regulates glucose metabolism in 3T3-L1 adipocytes through FGFR-1IIIc and FGFR-2IIIc.

Study on Tibetan Chicken embryonic adaptability to chronic hypoxia by revealing differential gene expression in heart tissue
