AIM： TO investigate the role of Lactobacillus crispatus （L. crispatus） strain China Center for Type Culture Col- lection （CCTCC） M206119 in intestinal inflammation.METHODS： Forty 8-wk-old Balb/c mice （20± 2 g） were divided into four groups of 10 mice each. Three groups that had received dextran sulfate sodium （DSS） were administered normal saline, sulfasalazine or CCTCC M206119 strain, and the fourth group received none of these. We assessed the severity of colitis using a disease activity index, measured the colon length and weight, collected stools and mesenteric lymph nodes for bacterial microflora analysis. One centimeter of the proximal colon, middle colon and distal colon were collected and fixed in 10% buffered formalin, dehydrated in ethanol, and embedded in paraffin. Interleukin （IL）- 1β, IL-6 and tumor necrosis factor （TNF）-α expression was detected using reverse transcription polymerase chain reaction. Protective factors zonula occludens （ZO）-1 and β-defensin 2 were detected by immunoblot-ting. The features of CCTCC M206119 strain were identified based on morphology, biochemical profile, and 16S RNA sequencing.RESULTS： DSS-colitis animals treated with CCTCC M206119 had markedly more severe disease, with greater weight loss, diarrhea, fecal bleeding, and shortened colon length. In addition, the CCTCC-M206119- treated group had comparatively higher histologi- cal scores and more neutrophil infiltration than the controls. Expression of protective factors ZO-1 and β-defensin 2 was downregulated due to destruction of the mucosal barrier after CCTCC M206119 strain treatment. An in vitro assay demonstrated that CCTCC M206119 strain increased the nuclear translocation of nuclear factor-κB in epithelial cells. Intestinal proinflam- matory or anti-inflammatory cytokine responses were evaluated. As one of the three payloads for the Advanced Space-based Solar Observatory(ASO-S)mission,the Lyman-alpha(Lyα)Solar Telescope(LST)is composed of three instruments:a Solar Corona Imager(SCI),a LyαSolar Disk Imager(SDI)and a full-disk White-light Solar Telescope(WST).When working in-orbit,LST will simultaneously perform high-resolution imaging observations of all regions from the solar disk to the inner corona up to 2.5 R⊙(R⊙stands for the mean solar radius)with a spatial resolution of 4.8′′and 1.2′′for coronal and disk observations,respectively,and a temporal resolution of 30–120 s and 1–120 s for coronal and disk observations,*** maximum exposure time can be up to20 s due to precise pointing and image stabilization *** the three telescopes of LST,SCI is a dual-waveband coronagraph simultaneously and independently observing the inner corona in the HI Lyα(121.6±10 nm)line and white light(WL)(700±40 nm)wavebands by using a narrowband Lyαbeam splitter and has a field of view(FOV)from 1.1 to 2.5 R⊙.The stray-light suppression level can attain<10^-6 B⊙(B⊙is the mean brightness of the solar disk)at 1.1 R⊙and≤5×10^-8 B⊙at 2.5 R⊙.SDI and WST are solar disk imagers working in the Lyαline and 360.0 nm wavebands,respectively,which adopt an off-axis two-mirror reflective structure with an FOV up to 1.2 R⊙,covering the inner coronal edge area and relating to coronal *** present the up-to-date design for the LST payload.

慢性充血性心衰患者肝功能与N端脑利钠肽前体相关性研究

目的探讨慢性充血性心力衰竭(chronic congestive heart failure,CHF)患者肝功能指标与N端脑利钠肽前体水平(N-terminal pro-brain natriuretic peptide,NT-proBNP)变化的相关性及临床意义。方法测定242例慢性心力衰竭患者(心衰组)和68例健康对照者(对照组)肝功能、血清NT-proBNP水平和左室射血分数(left ventricular ejection fraction,LVEF),心力衰竭患者心功能按NYHA心功能分级分为心衰Ⅱ级、Ⅲ/Ⅳ级组,分析肝功能、NT-proBNP水平与不同心衰程度的关系。结果心衰组总胆红素(total bilirubin,TBil)、直接胆红素(direct bililubin,DBil)、间接胆红素(indirect bilirubin,IBIL)、NT-proBNP水平明显高于对照组,心衰III/IV组TBil、DBil、IBil、NT-proBNP水平较II级组明显升高,在心衰组,TBil、DBil、IBil与NT-proBNP和NYHA心功能分级成正相关,与LVEF值成负相关。心衰组治疗后,TBil、DBil、IBil与NT-proBNP较治疗前明显降低。结论血清胆红素水平随心力衰竭严重程度的增加而升高,并与NT-proBNP水平和心功能状态相关,对慢性心力衰竭的诊断和病情判断有一定的临床价值。

目的研究非酒精性脂肪肝(NAFLD)形成和发展过程中不同阶段的蛋白质谱变化,寻找相关功能蛋白质分子,为治疗新靶点的研究提供依据。方法通过高脂饮食饲养,形成非酒精性脂肪肝病的SD大鼠模型,并观察不同时间肝脏的病理特征;对不同实验阶段大鼠肝脏总蛋白进行蛋白质组学分析,找出差异蛋白质并分析其在非酒精性脂肪肝形成过程中的作用。结果病理学改变:高脂组SD大鼠肝细胞脂肪变性程度、LEE指数较普通组明显(P<0.05);有效地鉴定出4种NAFLD形成过程中的差异蛋白质,分别为甲硫氨酸腺苷转移酶1、谷胱甘肽S-转移酶、还原型辅酶Ⅰ脱氢酶和醛脱氢酶A1。其中前三种蛋白质表达量随着高脂饮食时间增加而逐渐减少,而醛脱氨酶则逐渐增多。结论用高脂饲料喂养的SD大鼠可成功模拟代谢综合症患者NAFLD的肝脏病理学改变;高脂饮食小鼠肝内蛋白质组学随着肝内脂肪的聚集,其抗氧化能力逐渐减弱,清除内源醛基化产物的能力逐渐增加,物质代谢向合成代谢方向发展。

Calciphylaxis is a rare disease characterized histologically by microvessel calcification and microthrombosis,with high mortality and no proven ***,we reported a severe uremic calciphylaxis patient with progressive skin ischemia,large areas of painful malodorous ulcers,and mummified *** of the worsening symptoms and signs refractory to conventional therapies,treatment with human amnion-derived mesenchymal stem cells(hAMSCs)was *** release inspections of hAMSCs,efficacy,and safety assessment,including cytokine secretory ability,immunocompetence,tumorigenicity,and genetics analysis in vitro,were *** further performed acute and long-term hAMSC toxicity evaluations in C57BL/6 mice and rats,abnormal immune response tests in C57BL/6 mice,and tumorigenicity tests in neonatal Balbc-nu nude *** the preclinical research,the patient was treated with hAMSCs by intravenous and local intramuscular injection and external supernatant application to the *** followed up to 15 months,the blood-based markers of bone and mineral metabolism improved,with skin soft tissue regeneration and a more favorable profile of peripheral blood mononuclear *** biopsy after 1-month treatment showed vascular regeneration with mature noncalcified vessels within the dermis,and 20 months later,the re-epithelialization restored the integrity of the damaged *** infusion or local treatment-related adverse events ***,this novel long-term intravenous combined with local treatment with hAMSCs warrants further investigation as a potential regenerative treatment for uremic calciphylaxis due to effects of inhibiting vascular calcification,stimulating angiogenesis and myogenesis,anti-inflammatory and immune modulation,multidifferentiation,re-epithelialization,and restoration of integrity.
