Aberrant protein aggregation leads to various human diseases,but little is known about the physical chemical properties of these aggregated proteins in ***,we developed a boron-dipyrromethene(BODIPY)-based HaloTag probe,whose conjugation to HaloTag-fused proteins allows us to study protein aggregates using both fluorescence intensity and *** of BODIPY fluorophore reveals key structural features to attain the dual *** optimized probe exhibits increased fluorescence intensity and elongated fluorescence lifetime in protein *** lifetime imaging using this probe indicates that protein aggregates afford different viscosity in the forms of soluble oligomers and insoluble aggregates in live *** strategy presented in this work can be extended to enable a wide class of HaloTag probes that can be used to study a variety of physical properties of protein aggregates,thus helping unravel the pathogenic mechanism and develop therapeutic strategy.

For some years it has been possible to control the particle size of fly ashes, by-products of thermal power stations. Incorporating these very fine particles （obtained by grinding and/or pneumatic selection） improves the physical-mechanical characteristics of mortars and concretes. In this study, we measured the lime consumption of the various fractions （granulometric and densimetric） and identified by X-ray diffraction the neoformed phases by the pozzolanic reaction, to show that it is not sufficient to simply define the pozzolanicity of products based on lime consumption since it does not take into account the nature of the phases formed. The size of the particles used in the test samples also has a determining effect on the quantity of lime consumed. Before comparing results, it is necessary to ensure that the size of the particles （of the global ash and its constituents） be the same. We aimed to develop a whole-genome sequencing(WGS)-based copy number variant(cNV)calling algorithm with the potential of replacing chromosomal microarray assay(cMA)for clinical ***-cNV is thus developed for cNV detection from WGS *** performance of this cNV calling algorithm was evaluated in a blinded manner on 31 samples and compared to the 112 cNVs reported by clinically validated cMAs for these 31 *** result showed that JAX-cNV recalled 100%of these ***,JAX-cNV identified an average of 30 cNVs per individual,representing an approximately seven-fold increase compared to calls of clinically validated *** validation of 24 randomly selected cNVs showed one false positive,i.e.,a false discovery rate(FDR)of 4.17%.A robustness test on lowercoverage data revealed a 100%sensitivity for cNVs larger than 300 kb(the current threshold for college of American Pathologists)down to 10×*** cNVs larger than 50 kb,sensitivities were 100%for coverages deeper than 20×,97%for 15×,and 95%for 10×.We developed a WGS-based cNV pipeline,including this newly developed cNV caller JAX-cNV,and found it capable of detecting cMA-reported cNVs at a sensitivity of 100%with about a FDR of 4%.We propose that JAX-cNV could be further examined in a multi-institutional study to justify the transition of first-tier genetic testing from cMAs to ***-cNV is available at https://***/TheJacksonLaboratory/JAX-cNV.

收获是花生生产关键环节,用工量占整个生产过程60%以上,机械化收获是一个国家花生生产水平的重要体现。中国和美国同是世界重要的花生生产与出口大国,但因花生生产水平特别是收获机械化水平差距悬殊,从而导致花生出口的国际竞争力不同。美国两段式机械化收获方式及其先进的花生收获机械化技术,使花生生产高产优质高效、国际市场竞争优势明显,而中国多种花生机械化收获方式并存,收获机械化水平相对较低,从而制约了花生生产效益和出口竞争力。基于大量文献的系统分析、中国花生生产实地调研以及美国花生生产和收获机械化的实地考察与综合分析,综述了中国和美国花生生产及其收获机械化发展现状,分别阐述了中国和美国最新花生起收机、捡拾收获机等机器的类型、总体结构型式和主要性能参数等;分析了花生收获机械关键装置的结构原理与特点;综合分析了美国花生收获机械化发展历程以及快速发展的主要动因,中国花生收获机械化发展的主要制约因素;提出了中国花生收获机械化发展动态。对深入了解中国和美国花生收获机械化技术现况及其发展历程,进行中国花生收获机械化发展总结,在借鉴美国成功经验基础上因地制宜地采取措施,加速发展我国花生收获机械化,具有一定的参考价值。

For the mammalian brain to process and decipher the rich panoply of sounds that abound in the world, nature has evolved an elegant collection of neural circuits dedicated to this task. Indeed, the complexity, variety and number of neural pathways devoted to computing auditory information is unique among sensory modalities （Kaas, 2008）. Amyotrophic lateral sclerosis（ALS）is a devastating neurodegenerative disorder characterized by loss of upper and lower motor *** mechanisms contribute to the disease initiation and progression,including mitochondrial dysfunction which has been proposed to be a central determinant in ALS ***,while mitochondrial defects have been mainly described in ALS-linked SOD1 mutants,it is now well established that mitochondria become also dysfunctional in other ALS *** such context,the mitochondrial quality control system allows to restore normal functioning of mitochondria and to prevent cell death,by both eliminating and replacing damaged mitochondrial components or by degrading the entire organelle through *** evidence shows that ALS-related genes interfere with the mitochondrial quality control *** review highlights how ineffective mitochondrial quality control may render motor neurons defenseless towards the accumulating mitochondrial damage in ALS.
