Efficacy of gecko polysaccharide on suppressed immune response induced by cyclophosphamide in mice

OBJECTIVE:To evaluate the efficacy of gecko polysaccharide on the cyclophosphamide-induced suppressed immune response in ***:Polysaccharides were extracted from fresh gecko for the first time using an orthogonal method and protein was removed using Sevag reagent(chloroform∶N-butanol,5∶1,v/v).The gecko polysaccharide(GPCE)content was determined by the phenol-concentrated sulfuric acid *** immunocompromised mouse model was established by intraperitoneally injecting cyclophosphamide at 100 mg/kg into 48 *** effects of GPCE on immune regulation in mice were assessed by a thymus-spleen index,serum hemolysin levels,and the proliferation of splenic *** cell CD4+,CD8+,and the CD4+/CD8+ ratio were evaluated by flow cytometry and the tumor necrosis factor-α(TNF-α)levels were measured by ***:The optimal extraction process for gecko polysaccharide included a 1∶20 ratio of material to liquid(v/v),an extraction temperature of 60℃ and a time of 2 *** polysaccharide content of the extract was 65.74%.GPCE was analyzed by HPLC and primarily contained glucose and small amounts of mannose,rha,and *** with the model,the thymus index,the spleen index were indices for GPCE increased with dose,whereas the high and medium groups exhibited significant differences(P0.05);The high GPCE dose significantly increased the level of serum hemolysin(P<0.01).CONCLUSIONS:Gecko polysaccharide significantly improved the suppressed immune response induced by cyclophosphamide in mice and promoted the secretion of tumor necrosis *** mechanism of gecko polysaccharide as an a

目的探讨靶向PLK1基因在肝癌基因治疗中的可行性。方法采用PLK1小干扰核糖核酸分子(siRNA)转染人肝癌HepG2细胞,分别以荧光实时定量PCR和Western blot检测PLK1基因和蛋白的表达水平,观察PLK1 siRNA转染对肝癌细胞体外增殖的影响。并于转染不同时间后收集细胞,分别采用琼脂糖凝胶电泳和TUNEL方法检测肝癌细胞的凋亡情况。结果 PLK1基因明显抑制癌细胞体外生长(P<0.05)。肝癌HepG2细胞经siRNA转染处理后,PLK1 mRNA和蛋白表达水平明显下降(P<0.05)。DNA电泳出现明显的梯度图谱;转染组癌细胞凋亡指数明显增加。结论 PLK1 siRNA转染可明显抑制肝癌细胞增殖,其机制可能与诱导细胞凋亡有关。本结果为肝癌以PLK1基因治疗提供了一条新的思路。

Effect of heat treatment on microstructure and mechanical properties of selective laser-melted PH13-8Mo stainless steel

PH13-8Mo stainless steel powder with high sphericity and well fluidity was prepared by the plasma rotating electrode *** formed parts with ultra-high purity were manufactured by selective laser *** tensile and impact mechanical properties of the printed parts under three heat treatment regimes were compared to those without heat *** microstructure,grain orientation and phase composition were characterized by electron-backscatter diffraction,X-ray diffraction and transmission electron *** characterization results reveal that the addition of heat treatment makes the grains enlarge,and after different heat treatments,the average grain size from 1.51μm in the printed state increases to 2.78,3.09 and 2.06μm,*** formed parts are mainly composed of martensite and retained ***,the NiAl and M23C precipitates form,which are the major strengthening phases of PH13-8Mo stainless *** optimal heat treatment process is 925℃×1 h water cooling(WC)+0℃×2 h air cooling(AC)+540℃×4 h *** this heat treatment process,PH13-8Mo formed parts have the optimal comprehensive mechanical properties:the tensile strength,yield strength and impact energy KU2 are 1492 MPa,1432 MPa and 63 J,*** strengthening and toughening via heat treatment are mainly attributed to dislocation strengthening,the formation of NiAl and M23Co precipitates and the change of volume fraction of the retained austenite.

Piezoelectric accelerometers excel in vibration *** the emerging trend of fully organic electronic microsystems,polymeric piezoelectric accelerometers can be used as vital front-end components to capture dynamic signals,such as vocal vibrations in wearable speaking assistants for those with speaking ***,high-performance polymeric piezoelectric accelerometers suitable for such applications are *** organic compounds such as PVDF have inferior properties to their inorganic counterparts such as ***,most existing polymeric piezoelectric accelerometers have very unbalanced performance *** often sacrifice resonance frequency and bandwidth for a flat-band sensitivity comparable to those of PZT-based accelerometers,leading to increased noise density and limited application *** this study,a new polymeric piezoelectric accelerometer design to overcome the material limitations of PVDF is *** new design aims to simultaneously achieve high sensitivity,broad bandwidth,and low *** samples were manufactured and characterized,demonstrating an average sensitivity of 29.45 pC/g within a±10 g input range,a 5%flat band of 160 Hz,and an in-band noise density of 1.4μg/√*** results surpass those of many PZT-based piezoelectric accelerometers,showing the feasibility of achieving comprehensively high performance in polymeric piezoelectric accelerometers to increase their potential in novel applications such as organic microsystems.

The DArk Matter Particle Explorer(DAMPE)is a space high-energy particle andγ-ray detector whose major scientific goals are the indirect detection of dark matter particles,the origin of cosmic rays and highenergyγ-ray *** its successful launch in December 2015,the DAMPE has been operated smoothly in orbit for more than 8 *** direct measurements of the boron-to-carbon and boron-to-oxygen flux ratios,and the proton+Helium spectrum up to 316 TeV have been obtained,revealing new spectral features with very high *** search results ofγ-ray spectral line and fractionally charged particles indicate a good potential of DAMPE for indirect dark matter detection and new physical discovery in *** DAMPE measurements are expected to significantly advance our understanding of the fundamental problems in astroparticle physics.

目的探讨乌司他丁（Ulinastatin）在精准肝切除围手术期对肝功能的保护作用。方法选取行精准肝切除手术的原发性肝癌患者56例，将患者分为实验组（U组）和对照组（C组）各28例。C组给予常规药物治疗，U组在C组基础上予以乌司他丁治疗。检测两组患者围手术期肝脏功能及炎症因子变化。结果术前1d两组各检测指标差异无统计学意义（P〉0．05），术后两组各检测指标水平不同程度上升，U组术后1、3、5、7d ALT水平分别为（231±29；140±21；56±13；42±6）U／L，均低于C组（267±29；158±23；69±18；53±8）U／L；AST水平分别为（175±23；94±25；47±16；36±8）U／L，均低于C组（191±17；139±16；64±15；46±11）U／L；TBIL水平分别为（30±6；39±9；31±9：21±6）μmol／L，均低于C组（34±6；46±7；35±8；26±7）μmol／L；hs-CRP水平分别为（52±22；112±23；71±16；42±13）mg／L，均低于C组（69±23；129±25；72±15；49±15）mg／L；IL-1β水平分别为（7．8±0．9；11．1±1．5；8．9±1．6；5．6±0．9）pg／ml，均低于C组（9．2±1．3；13．0±2．1；11．14-1．4；7．3±0．8）pg／ml；IL-6水平分别为（187±30；76±25；46±15；26±8）pg／ml，均低于C组（260±36：92±16：53±11；30±8）pg／ml；TNF-α水平分别为（17±4；12．0±2．4；8．6±2．4；6．7±2．0）pg／ml，均低于c组（25±4；18．7±3．3；15．0±3．1；9．6±1．7）pg／ml，两组患者各检测指标在术后不同时间点对比，差异均有统计学意义（均P〈0．05）。结论乌司他丁对精准肝切除围手术期患者的肝功能具有保护作用．

Effect of tubastatin A on NLRP3 inflammasome activation in macrophages under hypoxia/ reoxygenation conditions

BACKGROUND:There are currently no effective drugs to mitigate the ischemia/reperfusion injury caused by fluid resuscitation after hemorrhagic shock(HS).The aim of this study was to explore the potential of the histone deacetylase 6(HDAC6)-specific inhibitor tubastatin A(TubA)to suppress nucleotide-binding oligomerization domain-like receptor protein 3(NLRP3)inflammasome activation in macrophages under hypoxia/reoxygenation(H/R)***:The viability of RAW264.7 cells subjected to H/R after treatment with different concentrations of TubA was assessed using a cell-counting kit-8(CCK8)***,2.5μmol/L TubA was used with RAW264.7 cells under H/R ***264.7 cells were divided into three groups,namely the control,H/R,and TubA *** levels of reactive oxygen species(ROS)in the cells were detected using fluorescence *** protein expression of HDAC6,heat shock protein 90(Hsp90),inducible nitric oxide synthase(iNOS),NLRP3,gasdermin-D(GSDMD),Caspase-1,GSDMD-N,and Caspase-1 p20 was detected by western *** levels of interleukin-1β(IL-1β)and IL-18 in the supernatants were detected using enzyme-linked immunosorbent assay(ELISA).RESULTS:HDAC6,Hsp90,and iNOS expression levels were significantly higher(P<0.01)in the H/R group than in the control group,but lower in the TubA group than in the H/R group(P<0.05).When comparing the H/R group to the control group,ROS levels were significantly higher(P<0.01),but significantly reduced in the TubA group(P<0.05).The H/R group had higher NLRP3,GSDMD,Caspase-1,GSDMD-N,and Caspase-1 p20 expression levels than the control group(P<0.05),however,the TubA group had significantly lower expression levels than the H/R group(P<0.05).IL-1βand IL-18 levels in the supernatants were significantly higher in the H/R group compared to the control group(P<0.01),but significantly lower in the TubA group compared to the H/R group(P<0.01).CONCLUSION:TubA inhibited the expression of HDAC6,Hsp90,and iNOS in macrophages
