Implications of Klotho in vascular health and disease

Cardiovascular disease(CVD) is a prevalent condition in general population and the first cause of death overall. Klotho, a pleiotropic protein related to longevity that acts as a co-receptor of the fibroblast growth factor 23, has been proposed as a key regulator of the development of CVD. In the few clinical studies made, it has been observed a relationship between low levels of soluble Klotho and the occurrence and severity of CVD, as well as a reduction of cardiovascular risk when they are high. Also, different polymorphisms of human Klotho gene have been related to the incidence of cardiovascular events. moreover, several experimental studies indicate that this protein acts in the maintenance of vascular homeostasis. Klotho improves endothelial dysfunction through promotion of nO production and mediates antiinflammatory and anti-aging effects such as suppression of adhesion molecules expression, attenuation of nuclear factor-kappa B or inhibition of Wnt signaling. Furthermore,this protein is related to the attenuation of vascular calcification as well as prevention of cardiac hypertrophy. The expression of this protein in the vascular wall implies a new scenario for the treatment of vascular disorders. The purpose of this review is to provide an overview of the relationship between the Klotho protein and CVD, in addition to its role in the maintenance of functional vascular integrity.

In vivo testing of mucus-permeating nanoparticles for oral insulin delivery using Caenorhabditis elegans as a model under hyperglycemic conditions

The aim was to evaluate the potential of mucus-permeating nanoparticles for the oral administration of *** nanocarriers,based on the coating of zein nanoparticles with a polymer conjugate containing PEG,displayed a size of 260 nm with a negative surface charge and an insulin payload of 77 mg/*** intestinal pig mucus,the diffusivity of these nanoparticles(PPA-nPs)was found to be 20-fold higher than bare nanoparticles(nPs).These results were in line with the biodistribution study in rats,in which nPs remained trapped in the mucus,whereas PPA-nPs were able to cross this layer and reach the epithelium *** therapeutic effcacy was evaluated in Caenorhabditis elegans grown under high glucose *** this model,worms treated with insulin-loaded in PPA-nPs displayed a longer lifespan than those treated with insulin free or nanoencapsulated in *** finding was associated with a signifcant reduction in the formation of reactive oxygen species(ROS)as well as an important decrease in the glucose and fat content in *** effects would be related with the mucus-permeating ability of PPA-nPs that would facilitate the passage through the intestinal peritrophic-like dense layer of worms(similar to mucus)and,thus,the absorption of insulin.

Analysis of the Sensory and Hedonic Impacts of Sweet and Bitter Tastes in Perinatally Underfed Rats

newborn rats discriminate tastes and generate gustofacial response (GFR) because the neuronal substrate is already operating. The oral application of sucrose or quinine hydrochloride produces a specific GFR. We analyzed the effects of perinatal undernutrition on the GFR development of rats at two cue concentrations. In the undernourished group, pregnant dams received different percentages of a balance diet. After birth, prenatally underfed pups continue the undernourishment by remaining for 12 h with a foster dam, and for 12 h with a nipple-ligated mother. Cues were presented as a single droplet of sucrose, sodium chloride, or quinine at low or high concentrations onto the lips at postnatal days (PDs) 1 and 3, and mouth-opening (mOF) and lip-licking frequencies (LLF) were noted. On PD 1 the undernourished group showed smaller mOF increases in response to low salt and quinine stimuli than the controls but no differences at high concentrations. On PD 3, both low and high concentrations of the sucrose and quinine cues significantly increased the mOF in the underfed compared to the control group. Low but not high salt decreased LLF on PD1 in the underfed compared to the control group. On PD 3 the undernourished pups showed significant increases of LLF with low quinine compared with the control rats, but the reverse was observed with high quinine. These data suggest that perinatal undernutrition affects the development of the sensory and hedonic aspects of taste causing changes in GFR expression.

Sweet and Bitter Tastes Evoked Different neuronal Activation in the Rostral Portion of the nucleus of the Solitary Tract of Developing Rats

The impact of the gustatory stimuli on the rostral portion of the nucleus of the solitary tract (rnST) was investigated in developing rats, by using c-fos immunohistochemical staining. Wistar male rats of 5, 15, and 25 days of age were isolated from the mother for 12 h, then stimulated via the intraoral route with quinine, sucrose, or naCl, and sacrificed 90 min later. The water-stimulated group showed minimal c-fos-like immunoreactivity (FLI) compared with taste-stimulated groups that exhibited different FLI in the rnST at the different ages. At all ages the quinine-stimulated group induced FLI in the medial subfield, while sucrose induced FLI in the lateral subfield of the rnST. The intensity of FLI was highest at P15, and it declined at P25. These findings provide detailed insight into the anatomical basis of rnST activation that is involved in early food intake and the learning capacity of the newborn.

Chronic pain, posttraumatic stress disorder, and opioid intake: A systematic review

BACKGROUnD The literature suggests that there is a high degree of co-occurrence between chronic pain and posttraumatic stress disorder(PTSD). An association has been found between PTSD and substance abuse. PTSD is a severe disorder that should be taken into account when opioids are prescribed. It has been found that the prevalence of opioid use disorder(OUD) in chronic pain patients is higher among those with PTSD than those without this *** To perform a systematic review on the association between PTSD, chronic noncancer pain(CnCP), and opioid intake(i.e., prescription, misuse, and abuse).mETHODS We conducted a systematic review following the Preferred Reporting Items for Systematic Reviews and meta-Analyses guidelines. The Patient, Intervention,Comparator, and Outcomes(PICOS) criteria were formulated a priori in the protocol of the systematic review. A search was conducted of the PROSPERO database. In march 2019, searches were also conducted of 5 other databases:Pub med, mEDLInE, Psyc InFO, Web of Science, and PILOTS. The Scottish Intercollegiate Guidelines network checklist for cohort studies was used to assess the selected studies for their methodological quality and risk of bias. Each study was evaluated according to its internal validity, participant sampling,confounding variables, and the statistical *** A total of 151 potentially eligible studies were identified of which 17 were retained for analysis. Only 10 met the selection criteria. All the studies were published between 2008 and 2018 and were conducted in the United States. The eligible studies included a total of 1622785 unique participants. Evolution of Acquired Humoral Immunity after Full Vaccination against SARS-CoV-2. IgG Levels in Healthcare Workers at 6 and 9 months

Background: The COVID-19 pandemic continues to be a major worldwide health problem. The present study aims to contribute to surveillance of the immune and clinical response of vaccines to SARS-CoV-2. methods: Observational medication study on acquired immunity and effectiveness of vaccines. Population: 620 workers in the health service of Almansa (Spain). Representative sample of 150 individuals. Sociodemographic, clinical, and epidemiological data and samples were recorded to determine anti-SARS-CoV-2 serum IgG levels 6 and 9 months after vaccination with Pfizer. Results: mean age 46.45 years;76% women;85.1% working in a hospital. 19.3% had had COVID-19 in the year prior to vaccination. 96.7% were fully vaccinated with Pfizer/BionTech. At 6 months, 100% seropositivity and mean IgG levels of 3017.2 AU/ml. Significant variations in IgG levels in individuals with prior COVID-19 infection and smokers. At 9 months, 99.3% remained seropositive;2.8% infected after vaccination. The repeated measures analysis showed a difference in means of 669.0 AU/ml (significant decrease in IgG levels of 28.9%). Conclusion: Antibody levels remained positive 6 and 9 months after vaccination, although IgG levels were found to decay.

YKL40 expression in CD14^+ liver cells in acute and chronic injury

AIm:To demonstrate that CD14 + cells are an important source of the growth factor YKL40 in acute and chronic liver ***:Rats were inoculated with one dose of CCl4 to induce acute *** biopsies were obtained at 0,6,12,24,48 and 72 *** chronic damage,CCl4 was administered three days per week for 6 or 8 *** samples were collected,and cellular populations were isolated by liver digestion and purified by cell ***40 mRnA and protein expression were evaluated by realtime polymerase chain reaction and western ***:Acute liver damage induced a rapid increase of YKL40 mRnA beginning at 12 *** peaked at 24 h,with a 26fold increase over basal *** 72 h however,YKL40 expression levels had nearly returned to control *** the other hand,chronic damage induced a sustained increase in YKL40 expression,with 7and 9fold higher levels at 6 and 8 wk,*** pattern of YKL40 expression in different subpopulations showed that CD14+cells,which include Kupffer cells,are a source of YKL40 after acute damage at 72 h[0.09 relative expression units(REU)]as well as after chronic injury at 6 wk(0.11 REU).Hepatocytes,in turn,accounted for 0.06 and 0.01 REU after 72 h(acute)or 6 wk(chronic),*** rest of the CD14cells(including T lymphocytes,B lymphocytes,natural killer and natural killer T cells) yielded 0.07 and 0.15 REU at 72 h and 6 wk,***40 protein expression in liver was detected at 72 h as well as 6 and 8 wk,with the highest expression relative to controls(11fold;P≤0.05)seen at 6 *** were stimulated by *** demonstrate that under these conditions,these cells showed maximum expression of YKL40 at 12 h,with Pmpared with ***:Hepatic CD14 + cells are an YKL40 mRnA and protein source in acute and chronic liver injury,with expression patterns similar to growth factors implicated in inflammationfibrogenesis.

Colorectal cancer(CRC)is the second leading cause of cancer-related death *** five-year survival rate of CRC patients depends on the stage at diagnosis,being higher than 80%when CRC is diagnosed in the early stages but lower than 10%when CRC is diagnosed in advanced *** against specific CRC autoantigens(tumor-associated antigens(TAAs))in the sera of patients have been widely demonstrated to aid in early ***,we herein aim to identify autoantigens target of autoantibodies specific to CRC that possess a significant ability to discriminate between CRC patients and healthy individuals by means of liquid *** that end,we examined the protein content of the exosomes released by five CRC cell lines and tissue samples from CRC patients by means of immunoprecipitation coupled with mass spectrometry analysis.A total of 103 proteins were identified as potential autoantigens specific to *** bioinformatics and meta-analysis,we selected 15 proteins that are more likely to be actual CRC autoantigens in order to evaluate their role in CRC prognosis by Western blot(WB)and immunohistochemistry(IHC).We found dysregulation at the protein level for 11 of these proteins in both tissue and plasma exosome samples from patients,along with an association of nine of these proteins with CRC *** validation,all but one showed a statistically significant high diagnostic ability to distinguish CRC patients and individuals with premalignant lesions from healthy individuals,either by luminescence Halotag-based beads,or by a multiplexed biosensing platform involving the use of magnetic microcarriers as solid support modified with covalently immobilized Halotag fusion proteins constructed for CRC *** together,our results highlight the usefulness of the approach defined here to identify the TAAs specific to chronic diseases;they also demonstrate that the measurement of autoantibody levels in plasma against the TAAs identified here

Quito municipal Schools—Cohort Study: Self-Perception of Body Image and Factors Related with It

Self-perception of body image is one factor to be considered when planning and performing preventive initiatives and interventions in childhood and adolescence. The aim of this study was to analyse self-perception of body image in relation to body-mass index, physical inactivity, not eating breakfast, dieting, smoking and alcohol consumption or toxic habits. materials and methods: using data from the “Quito municipal schools” cohort study 6964 students aged from 9 to 17 years were described using a multivariate log linear model of the multidimensional table generated by the variables. A logistic regression model was then fitted to assess associations via OR. Results: 8.2% perceived themselves as overweight. Of those subjects overweight according to their BmI, 21.8% perceived themselves as such, while among those not overweight according to BmI, 96.8% considered themselves as slim or of normal weight. Among students who were dieting, 15.1% perceived themselves as overweight. Among those pupils who perceived themselves has having excess weight, the most common reasons for dieting were: lose weight (56.8%), be healthier (22.6%), and maintain current weight (8.4%). Self-perception of excess weight interacts with excess weight (ORadjusted 8.42;CI95% 6.92-10.25), no breakfast (ORadjusted 2.83;CI95% 2.13-3.77), diet (ORadjusted 2.38;CI95% 1.95-2.89), and with all the variables except toxic habits (ORadjusted 1.01;CI95% 0.78 a 1.29). Conclusion: Interventions to prevent obesity in childhood and adolescence ought to take account of specific determinants within the personal, behavioural and socio-environmental factors, such as the promotion of a positive body image.
