Defects in phospholipase C zeta cause polyspermy and low fertilization after conventional IVF:not just ICSI failure

Phospholipase C zeta(PLC)is a key sperm-borne oocyte-activating factor that triggers Ca^(2+)oscillations and the subsequent block to polyspermy following gamete *** in PLCZ1,the gene encoding PLCζ,cause male infertility and intracytoplasmic sperm injection(ICSI)fertilization failure;and PLCζ expression and localization patterns are significantly correlated with ICSI fertilization rate(FR).However,in conventional in vitro fertilization(cIVF),whether and how sperm PLCζ affects fertilization remain ***,we identified one previously reported and two novel PLCZ1 mutations associated with polyspermy in vitro that are characterized by excessive sperm-zona binding and a delay in pronuclei(PN)*** staining and oocyte activation testing revealed that virtually all spermatozoa from patients lacked functional PLCζ and were thus unable to evoke Ca^(2+) *** with an artificial oocyte activation treatment successfully rescued the polyspermic phenotype and resulted in a live ***,we analyzed PLCζ in an additional 58 males after cIVF treatment in the Reproductive and Genetic Hospital of CiTiC-Xiangya(Changsha,China)between February 2019 and January *** found that the proportion of spermatozoa that expressed PLCζ was positively correlated with both 2PN rate and total *** optimal cutoff value below which males were likely to experience low FR(total FR≤30%)after clVF was 56.7%for the proportion of spermatozoa expressing *** study expands the mutation and the phenotypic spectrum of PLCZ1 and further suggests that PLCζ constitutes a promising biomarker for identifying low FRs cases in cIVF due to sperm-related oocyte activation deficiency and that sperm PLCζ analysis may benefit the widermale population and not onlymen with IcsI failure.

非小细胞肺癌(non-small cell lung cancer, NSCLC)是临床常见的恶性肿瘤,癌细胞的侵袭和转移是导致NSCLC患者死亡的主要原因,肿瘤相关巨噬细胞(tumor-associated marcrophages, TAMs)与此密切相关。TAMs是肿瘤浸润免疫细胞的重要组成部分,在肿瘤微环境(tumor microenvironment, TME)中有向M2表型分化的倾向,具有促进肿瘤细胞增殖、侵袭及迁移的作用。TAMs的分化受多种细胞因子和信号通路的调节,Notch信号通路是其中之一。该文就Notch信号通路调节TAMs分化影响NSCLC细胞侵袭性的相关研究进行综述,为NSCLC的诊治提供有价值的资料。

富含半胱氨酸的酸性分泌蛋白(secreted protein acidic and rich in cysteine, SPARC)是肿瘤微环境中重要的基质成分,它作为正常细胞恶变和肿瘤发生发展过程中一个重要的分子,能够调节多种肿瘤相关细胞因子及其受体的相互作用,并调节多种信号通路以及蛋白水解酶的表达水平。由于它抑制和促进肿瘤进展的能力取决于细胞类型、肿瘤分期等,因此在多种类型的肿瘤中差异表达,并可能成为肿瘤早期诊断的新型生物标记物以及治疗的新靶点。本文就近年来SPARC在肿瘤研究中的进展作一综述。

Penile sensory thresholds in subtypes of premature ejaculation： implications of comorbid erectile dysfunction

Penile hypersensitivity plays an important role in premature ejaculation （PE）, but differences in penile sensitivity among subtypes of PE are unknown. Therefore, we compared penile sensory thresholds in PE subtypes of lifelong and acquired PE, PE with and without erectile dysfunction （ED）, PE with an intravaginal ejaculation latency time 〈1 min and 〉1 min, and PE with and without orgasmic pleasure perceptual dysfunction. During August 2014 to January 2016, 136 patients with PE were included. Penile warm, cold, and vibratory thresholds were measured. Data of clinical characteristics, sexual life, Premature Ejaculation Diagnostic Tool （PEDT） score, and the 5-item version of the International Index of Erectile Function （IIEF-5） score were collected. Vibratory thresholds of the PE with ED group were higher in the right coronal sulcus （median amplitude： 4.92 vs 3.65 μm, P= 0.02） and the right penile shaft （median amplitude： 3.87 vs 3.30 μm, P = 0.03）, while differences in penile sensory thresholds between other subtypes were not significant. The median PEDT score was lower in the PE without ED group （12 vs 14, P 〈 0.001）. The IIEF-5 and PEDT scores were negatively correlated （r = -0.29, P 〈 0.001）. Patients with orgasmic pleasure perceptual dysfunction had a lower median IIEF-5 score （20 vs 21, P = 0.02）. Patients with PE and ED had lower penile sensitivity, and ED was associated with more severe symptoms and weaker orgasmic pleasure perception. In men with PE, management of comorbid ED is necessary. In case of side effects in erectile function, topical anesthetics should be cautiously used in men with PE and ED.

选矿浮选过程中浮选槽中的泡沫图像,受到工业摄像角度和光照点位置影响,导致泡沫图像颜色特征以及纹理特征的提取达不到预期效果。为解决上述问题,提出一种基于显著性检测的自适应全变差去高光算法。将处于RGB颜色空间的图像转换到处于YUV颜色空间中,根据显著值的大小,判定某个像素点是否为高光像素点,并修复图像高光区域。构建改进的全变差修复模型,并对图像修复模型完成求解。实验结果可知,所提算法对图像高光区域的识别以及细微处的处理具有一定的提高,且均方误差值与峰值信噪比在一定程度上得到了改善,可以有效的提取泡沫图像存在的亮点区域且修复。

K~π=5^- rotational bands in neutron-rich Mo,Ru,and Pd nuclei

The collective rotations of the K^π=5^- configuration in neutron-rich Mo, Ru and Pd isotopes were systematically investigated by the configuration-constrained cranking shell model based on the Skyrme Hartree-Fock method with pairing treated by shell-model diagonalization. The calculations efficiently reproduce the experimental moments of inertia of both the ground-state and side bands. Rotational bands built on two-particle K^π=5^- configurations have been the subject of intense study. Possible configurations were assigned to the observed 5^- bands in ^102-106Mo,^108-112Ru and ^112-114Pd. We predict the existence of the 5^- bands in ^108,110Mo. These results provide deep insights into the structure of neutron-rich nuclei, and provide useful information for future experiments.

Enhanced cycling stability of Mg-F co-modified LiNi_(0.6)Co_(0.2)Mn_(0.2-y)Mg_yO_(2-z)F_z for lithium-ion batteries

The layered LiNi0.6Co0.2Mn0.2-yMgyO2-zFz（0≤y≤0.12, 0≤z≤0.08） cathode materials were synthesized by combining co-precipitation method and high temperature solid-state reaction, with the help of the ball milling, to investigate the effects of F-Mg doping on LiNi0.6Co0.2Mn0.2O）2. Compared with previous studies, this doping treatment provides substantially improved electrochemical performance in terms of initial coulombic efficiency and cycle performance. The LiNi0.6Co0.2Mn0.11Mg0.09O1.96F0.04 electrode delivers an high capacity retention of 98.6% during the first cycle and a discharge capacity of 189.7 m A·h/g（2.8-4.4 V at 0.2 C）, with the capacity retention of 96.3% after 100 cycles. And electrochemical impedance spectroscopy（EIS） results show that Mg-F co-doping decreases the charge-transfer resistance and enhances the reaction kinetics, which is considered to be the major factor for higher rate performance. It is demonstrated that LiNi0.6Co0.2Mn0.11Mg0.09O1.96F0.04 is a promising cathode material for lithium-ion batteries for excellent electrochemical properties.

Nb_(2)P_(5) is a recently discovered Bardeen,Cooper,Schrieffer theory s-wave superconductor hosting nontrivial topological nodal-line structure,which could serve as an excellent platform for studying the interplay between the superconductivity and nontrivial topological *** report herein the superconducting and structural properties of Nb2P5studied by measuring the electrical resistivity and structure under high pressure up to ~28 GPa and performing first principles *** superconducting transition temperature TCdecreases with applied pressure,and reaches a minimum of 1.9 K at ~20.0 GPa,followed by a continuous enhancement up to 50.0 GPa without ***,at pressures above ~8.2 GPa,it is likely that a second superconducting transition appears and coexists with the initial one even up to~28 *** calculations rule out the correlation between the second superconducting-like transition and ***,the Nb2P5showed remarkable axial anisotropic compressibility,in which the b-axis is more compressible than the a-and *** variation of TCis discussed via considering the evolution of density of states around the Fermi level and the phonon spectrum.

In our previous work, reactive extraction using di(2-ethylhexyl) phosphate acid(D2EHPA) has been successfully used for the separation of 4-Hydroxylpyridine(4HP) from the aqueous solution. This study investigates the separation of 4HP by synergistic extraction. Several factors including the extraction agents,p H value,the concentrations of co-extractant and 4HP, which influence the co-extraction of 4HP are also studied. The obtained results suggest that the co-extractant of D2 EHPA and tri-n-butyl-phosphate(TBP) could more effectively be used to extract 4HP. Optimum conditions of synergistic extraction are obtained as follows: the molar ratio of D2 EHPA to TBP(Rs) is 2:1, the aqueous equilibrium p H is found to be one of the key factors that sharply affect the distribution ratio(D)values and the best p H is 4-4.8. Moreover, the extraction mechanism is analyzed by the Fourier infrared spectrum, and it is found that the reactive extraction of 4HP by D2EHPA+TBP is mainly driven by two mechanisms: the ionic association synthetic salt and hydrogen bond association.
