Construction and expression of a Rev-dependent TNF-R1 expressing HIV-infected-cell injurious vectors

Background Rev is necessary for exporting unspliced and incompletely spliced intron containing HIV mRNAs and for HIV replication. The aim of this study is to develop a kind of selective suicide construct that can specifically and directly induce HIV infected cells into apoptosis based on the high affinity of Rev and Rev response element （RRE）. Methods Molecular-cloning technique was used to synthesis Rev dependent TNF-R1 expression construct pDM128-TNF-R1 （pT128） that contains RRE and TNFR1 gene. Restriction digestion, polymerase Chain Reaction （pCR） and DNA sequencing were processed and the exactness and correctness of the inserted TNF-R1 gene in pT128 were confirmed repeatedly. The expression of pT128 co-transfected with different combination of other plasmids by calcium phosphate-DNA co-precipitation in Helas and by gene gun transfection in keratinocytes was further tested by flow-cytometry and cell counted under microscope. Results The new plasmid specifically expressed TNF-R1 in Helas when co-transfected with pRev but did not when without pRev. Indirect expression of TNF-R1 from pT128 was slower than the direct expression of that from Hu p60 TNFR1 in pDC302 （pT60） , but all those pT60 or pT128 transfected cells showed apoptosis at last while TNF-R1 was sufficiently expressed. Other kinds of Rev expression construct such as pAD8 and a chimeric HIV vaccine also can switched on the selective expression of pT128. Not only Rev-dependent expression in Helas, pT128 also normally expressed its TNF-R1 in keratinocytes. Co-transfected with prey or pAD8 that expressed Rev, pT128 expressed TNF-R1 and induced apoptosis of green fluorescent keratinocytes in skin explant. The number of green fluorescent keratinocytes co-transfected by pT128 plus prey or pAD8 was gradually outnumbered by that co-transfected by pT128 only. The difference was more significant after culturing for 72 hours. Conclusions Rev dependent pT128 is able to selectively induce apoptosis of HIV-infect

pb-Sn mixed perovskites are becoming increasingly popular as narrowbandgap(1.2–1.3 eV)light absorbers in single-junction perovskite solar cells(pSCs)and as bottom cells for all-perovskite tandem solar cells,for highefficiency,low-cost,lightweight,roll-to-roll printable photovoltaic(pV)*** the first report of planar pb:Sn mixed pSCs in 2014,the power conversion efficiencies(pCE)have increased from 10%to 21%by the end of 2020 with an exponential growth in research conducted in this *** much effort,the performance and stability of pb-Sn mixed pSCs are still limited,which constrains their long-term use in all-perovskite tandem *** review highlights the avenues explored in improving different aspects of pb-Sn mixed pSCs and provides a comprehensive discussion of the interdependent factors affecting the device *** includes compositional engineering of the perovskite crystal,absorber layer fabrication and crystallization methods,bandgap tuning,Sn4+reduction,and surface passivation of the absorber layer,as well as the selection of interlayers and electrodes of the final pSC.

We describe nNpipe for the automated analysis of morphologically diverse catalyst materials. Automated imaging routines anddirect-electron detectors have enabled the collection of large data stacks over a wide range of sample positions at high temporalresolution. Simultaneously, traditional image analysis approaches are slow and hence unsuitable for large data stacks andconsequently, researchers have progressively turned towards machine learning and deep learning approaches. previous studiesoften detail work on morphologically uniform material systems with clearly discernible features, limited workable image sizes andtraining data that may be biased due to manual labelling. The nNpipe data-processing method consists of two standaloneconvolutional neural networks that were exclusively trained on multislice image simulations and enables fast analysis of2048 × 2048 pixel images. Inference performance compared between idealised and real industrial catalytic samples and insightsderived from subsequent data analysis are placed into the context of an automated imaging scenario.

Objective: To observe the activity of repeated extracts of bone matrix and the production of purified bone morphogenetic proteins (BMps). Methods: BMps were extracted 1- 4 times from fresh bovine cortical bone by the modified Urist’s method, with each collected precipitate separated and lyophilized as partially purified BMps. Another fresh bovine bone was extracted three times and the precipitates were mixed and lyophilized. Meanwhile,the alkaline phosphatase (ALp) activity was measured by an in vitro assay employing cultured C2C12 mouse myoblast cells through the osteoinductivity of bovine BMps extracted four times at days 1, 4, 7, and 14, and the correlation between BMps quantities and costing during extraction processes was ***: The BMps purified and the cost showed a positive correlation (r=(0.969)). To separate and lyophilize each collected precipitate as partially purified BMps raised the cost, and mixed precipitates also cost much. ALp activities of the 1st and mixed extractions of BMps were shown to be highly osteoinductive and keep a significantly high level (ppared with the 2nd, 3rd and 4th extractions, especially the control group. However, the more times the extraction was done, the less activity of BMps was shown and more costing was. The x-ray and histological analysis also showed that the 1st extraction of BMps induced more ossicles and new bone ***: The results indicated that BMps enhanced the abilities of osteoinductivity in C2C12 culture in vitro. The first extraction of BMps from bone is fitfull, the second extraction should be enough, while, the 3rd and 4th extractions are unnecessary for they cost more and waste more time, say nothing of mixed extractions.

Canadian boreal mixedwood forests are extensive,with large potential for carbon sequestration and storage;thus,knowledge of their carbon stocks at different stand ages is needed to adapt forest management practices to help meet climate-change mitigation *** stocks were quantified at three Ontario boreal mixedwood sites.A harvested stand,a juvenile stand replanted with spruce seedlings and a mature stand had total carbon stocks(±SE)of 133±13 at age 2,130±13 at age 25,and 207±15 Mg C ha^-1 at age 81 *** the clear-cut site,stocks were reduced by about 40%or 90 Mg C ha^-1 at *** held 27,34 and 62%of stocks,while detritus held 34,29 and 13%of stocks at age 2,25 and 81,*** soil carbon stocks averaged 51 Mg C ha^-1,and held 38,37 and 25%of *** net primary productivity(±SE)in the harvested and juvenile stand was 2.1±0.2 and 3.7±0.3 Mg C ha^-1 per annum(p.a.),compared to 2.6±2.5 Mg C ha^-1 *** the mature *** mature canopies studied had typical boreal mixedwood composition and mean carbon densities of 208 Mg C ha^-1,which is above average for managed Canadian boreal forest ecosystems.A comparison of published results from Canadian boreal forest ecosystems showed that carbon stocks in mixedwood stands are typically higher than coniferous stands at all ages,which was also true for stocks in vegetation and ***,aboveground net primary productivity was typically found to be higher in mixedwood than in coniferous boreal forest stands over a range of *** from this study,together with those published from the other boreal forest stands demonstrate the potential for enhanced carbon sequestration through modified forest management practices to take advantage of Canadian boreal mixedwood stand characteristics.

OBJECTIVE The objective of this study was to characterize the neurotransmitter systems that cause constriction of murine *** Murine precision cut lung slices(pCLS)and trachea were prepared,placed into perfusion chambers equipped with platinum electrodes and stimulated transmurally(1.0 ms,50 V,0.1-30 Hz).To measure pCLS constriction,changes in airway luminal area in response to electric field stimulation(EFS)were captured as video images quantified using Image J *** trachea,changes in isometric tension were recorded using Grass force *** response curves were generated in the absence and the presence of the inhibitors magnesium,atropine and capsaicin and responses analyzed and compared using a student's t-test(ppendent manner that was best fit by a biphasic ***-specific stimulation was verified by Mg^(2+) *** airway constriction to 30 Hz EFS in pCLS was(51.8±3.0)%while tracheal constriction averaged(551±80)***,in pCLS the muscarinic receptor antagonist atropine(10μmol·L^(-1))blocked(99.5±7.2)%of EFS induced constriction at 1 Hz,but only blocked(23.3±3.8)%of EFS induced constriction at 30 Hz and eliminated the first phase but not the second phase of the biphasic EFS *** with capsaicin to deplete sensory neurotransmitters significantly increased EFS constriction supporting the presence of sensory neurotransmitter systems in *** These data are consistent with parasympathetic constriction of airways by acetylcholine at lower EFS frequencies while higher frequencies release sensory dilator *** data provide evidence for multiple nerve types innervating airways which may provide novel targets for treatment of lung disease.

Bleeding outcomes after non-emergency percutaneous coronary intervention in the very elderly

Background Octogenarians constitute an increasing proportion of patients presenting for non-emergency percutaneous coronary intervention （pCI）. Methods This study evaluated the in-hospital procedural characteristics and outcomes, including the bleeding events of 293 octogenarians presenting between January 2010 and December 2012 for non-emergency pCI to a single large volume tertiary care Aus- tralian center. Comparisons were made with 293 consecutive patients aged less than or equal to 60 years, whose lesions were matched with the octogenarians. Results Non-ST elevation myocardial infarction was the most frequent indication for non-emergency pCI in octoge- narians. Compared to the younger cohort, they had a higher prevalence of co-morbidities and more complex coronary disease, comprising more type C and calcified lesions. peri-procedural use of low molecular weight heparin （LMWH; 1.0% vs. 5.8%; p 〈 0.001） and glycopro- tein IIb/IIIa inhibitors （2.1% vs. 9.6%; p 〈 0.001） was lower, while femoral arterial access was used more commonly than in younger patients （80.9% vs. 67.6%; p 〈 0.001）. Overall, there was a non-significant trend towards higher incidence of all bleeding events in the elderly （9.2% vs. 5.8%; p = 0.12）. There was no significant difference in access site or non-access site bleeding and major or minor bleeding between the two cohorts. Sub-analysis did not reveal any significant influence on bleeding rates by the use of LMWH, glycoprotein IIb/IIIa inhibitors or femoral arterial access. In addition, there were no significant differences in the rates of in-hospital mortality, stroke or acute stent thrombosis between the two groups. Conclusions In this single center study, we did not observe significant increases in adverse in-hospital outcomes including the incidence of bleeding in octogenarians undergoing non-emergency pCI.
