The Rosaceae crop family(including almond,apple,apricot,blackberry,peach,pear,plum,raspberry,rose,strawberry,sweet cherry,and sour cherry)provides vital contributions to human well-being and is economically significant across the *** 2003,industry stakeholder initiatives prioritized the utilization of genomics,genetics,and breeding to develop new cultivars exhibiting both disease resistance and superior horticultural ***,rosaceous crop breeders lacked certain knowledge and tools to fully implement dNA-informed breeding—a“chasm”existed between existing genomics and genetic information and the application of this knowledge in *** RosBREEd project(“Ros”signifying a Rosaceae genomics,genetics,and breeding community initiative,and“BREEd”,indicating the core focus on breeding programs),addressed this challenge through a comprehensive and coordinated 10-year effort funded by the USdA-NIFA Specialty Crop Research *** was designed to enable the routine application of modern genomics and genetics technologies in *** crop breeding programs,thereby enhancing their efficiency and effectiveness in delivering cultivars with producer-required disease resistances and market-essential horticultural *** review presents a synopsis of the approach,deliverables,and impacts of RosBREEd,highlighting synergistic global collaborations and future *** technologies and tools developed are described,including genome-wide scanning platforms and dNA diagnostic *** of dNA-informed breeding use by project participants are presented for all breeding stages,including pre-breeding for disease resistance,parental and seedling selection,and elite selection *** chasm is now bridged,accelerating rosaceous crop genetic improvement.

Prior to the availability of whole-genome sequences,our understanding of the structural and functional aspects of Prunus tree genomes was limited mostly to molecular genetic mapping of important traits and development of EST *** public release of the peach genome and others that followed,significant advances in our knowledge of Prunus genomes and the genetic underpinnings of important traits *** this review,we highlight key achievements in Prunus genetics and breeding driven by the availability of these whole-genome *** the structural and evolutionary contexts,we summarize:(1)the current status of Prunus whole-genome sequences;(2)preliminary and ongoing work on the sequence structure and diversity of the genomes;(3)the analyses of Prunus genome evolution driven by natural and man-made selection;and(4)provide insight into haploblocking genomes as a means to define genome-scale patterns of evolution that can be leveraged for trait selection in pedigree-based Prunus tree breeding programs ***,we summarize recent and ongoing work that leverages whole-genome sequences to identify and characterize genes controlling 22 agronomically important Prunus *** include phenology,fruit quality,allergens,disease resistance,tree architecture,and ***,we explore the application of sequence-based marker-assisted breeding technologies and other sequence-guided biotechnological approaches for Prunus crop ***,we present the current status of publically available Prunus genomics and genetics data housed mainly in the Genome database for Rosaceae(GdR)and its updated functionalities for future bioinformatics-based Prunus genetics and genomics inquiry.

本文旨在以下两个方面建立有效算法:(1)基于时间偏移速度建立地震速度模型;(2)将时间偏移成像结果转换到深度域。本文借助于旁轴射线追踪理论,分别在2d和3d情况下建立了时间偏移速度和地震速度之间的关系。2d情况下的理论分析表明,常规dix速度是地震层速度与成像射线的几何扩散值的比值。文章通过公式推导建立了由dix速度得到地震速度的反问题,并且找到了解决的数值计算方法。方法包括两个步骤:①计算成像射线的几何扩散值,并由dix速度计算时间域坐标系下的真地震速度;②计算时间域坐标系到深度域的转换矩阵,实现真地震速度时间域到深度域的转换。在步骤1中,我们推导出联系dIX速度和成像射线的几何扩散值的偏微分方程(PdE)。这是一个非线性椭圆偏微分方程。结合此方程的物理意义我们提出一个柯西问题。这个问题是不适定的,但是我们可以在要求的时段从两个方面得到其数值解,不过这个时段要足够小。一种是受Lax-Friedrichs法的启发得到的有限差分法;另一种是频谱—契比雪夫法。而在步骤2中,受Sethian的快速前进法启发建立了一种有效的似dijkstra解决方案。最后结合一个合成数据模型和野外实例对上述数值计算方法进行了测试,结果表明该地震速度分析法较常规dix反演,可明显提高计算精确性。该算法可以用于深度偏移中速度模型的建立。

A controlled hydroponic experiment was undertaken to investigate Cd uptake in relation to the activity of Cd species in solution other than the free ion （Cd^2＋） by maintaining a constant Cd^2＋ activity under variable SO4^2- and Cl^- concentrations exposed to maize （Zea mays vat. cameron） plants. The objectives of these experiments were：（1） to distinguish and quantify the different uptake rates of free and inorganic-complexed Cd from nutrient solution, and （2） to model the uptake of Cd by maize with a Biotic Ligand Model （BLM） in a system which facilitates the close examination of root characteristics. Results of the current experiments suggest that, in addition to the free ion, CdSO4^0 complexes are important factors in determining Cd uptake in nutrient solution by maize plants. Higher nominal SO4^2- concentrations in solution generally resulted in a greater Cd accumulation by maize plants than predicted by the Cd^2＋ activity. A better integration of the complete dataset for the 3 harvest times （6, 9 and 11 days after treatment） was achieved by including consideration of both the duration of Cd exposure and especially the root surface area to express Cd uptake. Similarly, the fit of the BLM was also improved when taking into account exposure time and expressing uptake in terms of root morphological parameters.

Using directed mutagenesis and phage display on a soluble fragment of the human immunoglobulin superfamily receptor ILT2(synonyms:LIR1,MIR7,Cd85j),we have selected a range of mutants with binding affinities enhanced by up to 168,000-fold towards the conserved region of major histocompatibility complex(MHC)class I *** in a dimeric form,either by chemical cross-linking with bivalent polyethylene glycol(PEG)derivatives or as a genetic fusion with human IgG Fc-fragment,the mutants exhibited a further increase in ligand-binding strength due to the avidity effect,with resident half-times(t1/2)on the surface of MHC I-positive cells of many *** novel compounds antagonized the interaction of Cd8 co-receptor with MHC I in vitro without affecting the peptide-specific binding of T-cell receptors(TCRs).In both cytokine-release assays and cell-killing experiments the engineered receptors inhibited the activation of Cd8^(+)cytotoxic T lymphocytes(CTLs)in the presence of their target cells,with subnanomolar potency and in a dose-dependent *** a selective inhibitor of Cd8^(+)CTL responses,the engineered high affinity ILT2 receptor presents a new tool for studying the activation mechanism of different subsets of CTLs and could have potential for the development of novel autoimmunity therapies.

Interest in Heat Recovery Coke Oven（HRCO） usage and processes has increased due to their improved environmental performance and ability to use a wide range of metallurgical and non-coking coals. To better understand the HRCO process, the Canadian Carbonization Research Association（CCRA） embarked on a program to construct a pilot-scale HRCO. Hatch completed the design of this unique pilot oven needed to reproduce the industrial scale HRCO heating profile and coal charge, and to minimize coal heating edge effect. different aspects of the pilot coke oven design concept are presented, including oven geometry and wall insulation design, heating strategy using electric heaters, structural buckstay and oven door designs. Several 2d and 3d thermal transient finite element analyses were carried out to calculate the temperature profile of the coal/coke bed and oven walls during coking, capturing the thermal nonlinear behavior of the coal and oven wall bricks and to size the coal bed insulation for minimum edge heating effect. The structural stability of the oven arches was achieved using a spring loaded buckstay for lateral support and expansion allowance. The pilot oven, referred to as the Energy Recovery Coke Oven（ERCO）, is being constructed at the Canmet ENERGY Laboratory in Ottawa, Canada.
