Expression and clinical significance of short-chain fatty acids in patients with intrahepatic cholestasis of pregnancy

BACKGROUND Intrahepatic cholestasis of pregnancy(ICP)is a pregnancy-specific liver condition that typically arises in the middle and late stages of ***-chain fatty acids(SCFAs),prominent metabolites of the gut microbiota,have significant connections with various pregnancy complications,and some SCFAs hold potential for treating such ***,the metabolic profile of SCFAs in patients with ICP remains *** To investigate the metabolic profiles and differences in SCFAs present in the maternal and cord blood of patients with ICP and determine the clinical significance of these *** Maternal serum and cord blood samples were collected from both patients with ICP(ICP group)and normal pregnant women(NP group).Targeted metabolomics was used to assess the SCFA levels in these *** Significant differences in maternal SCFAs were observed between the ICP and NP *** SCFAs exhibited a consistent declining trend in cord blood samples from the ICP group,mirroring the pattern seen in maternal *** analysis revealed a positive correlation between maternal serum SCFAs and cord blood SCFAs[r(Pearson)=0.88,P=7.93e-95].In both maternal serum and cord blood,acetic and caproic acids were identified as key metabolites contributing to the differences in SCFAs between the two groups(variable importance for the projection>1).Receiver operating characteristic analysis demonstrated that multiple SCFAs in maternal blood have excellent diagnostic capabilities for ICP,with caproic acid exhibiting the highest diagnostic efficacy(area under the curve=0.97).CONCLUSION Compared with the NP group,significant alterations were observed in the SCFAs of maternal serum and cord blood in the ICP group,although they displayed distinct patterns of ***,the SCFA levels in maternal serum and cord blood were significantly positively ***,certain maternal serum SCFAs,specifically caproic and acetic acids,demonst

Practical cloud storage auditing using serverless computing

Cloud storage auditing research is dedicated to solving the data integrity problem of outsourced storage on the cloud. In recent years, researchers have proposed various cloud storage auditing schemes using different techniques. While these studies are elegant in theory, they assume an ideal cloud storage model;that is, they assume that the cloud provides the storage and compute interfaces as required by the proposed schemes. However, this does not hold for mainstream cloud storage systems because these systems only provide read and write interfaces but not the compute interface. To bridge this gap, this work proposes a serverless computing-based cloud storage auditing system for existing mainstream cloud object storage. The proposed system leverages existing cloud storage auditing schemes as a basic building block and makes two adaptations. One is that we use the read interface of cloud object storage to support block data requests in a traditional cloud storage auditing scheme. Another is that we employ the serverless computing paradigm to support block data computation as traditionally required. Leveraging the characteristics of serverless computing, the proposed system realizes economical, pay-as-you-go cloud storage auditing. The proposed system also supports mainstream cloud storage upper layer applications(e.g., file preview) by not modifying the data formats when embedding authentication tags for later auditing. We prototyped and open-sourced the proposed system to a mainstream cloud service, i.e., Tencent Cloud. Experimental results show that the proposed system is efficient and promising for practical use. For 40 GB of data, auditing takes approximately 98 s using serverless computation. The economic cost is 120.48 CNY per year, of which serverless computing only accounts for 46%. In contrast, no existing studies reported cloud storage auditing results for real-world cloud services.

Upgrading waste NO_(x) into amino acids via electrocatalysis on Co nanoparticles encapsulated in hollow carbon nanofibers

Electrocatalytic conversion of waste nitrogen oxide(NO_(x))into value-added nitrogenous chemicals,especially amino acid,is an emerging strategy to alleviate NO_(x)***,this process suffers from the coexistence of multiple competitive reactions leading to complex product distribution and low efficiency,and thus the rational design and modulation of efficient catalysts remains a formidable ***,well-designed Co nanoparticles encapsulated in hollow carbon nanofiber(CoHCNF)were constructed to promote the amino acid electrosynthesis via dealing waste NO_(x)coupling with biomass-derived keto *** to the rapid reactant diffusion,electron transport and efficient contact with abundant accessible Co catalytic sites stemmed from the unique hollow nanotubular structure,Co-HCNF manifested impressive glycine synthesis performance in one-pot adopting NO as the nitrogen source with Faradaic efficiency(FE)of 41.7%and a corresponding yield rate of 222.3μmol h^(-1)as well as 120 h robust ***,it also demonstrates an excellent performance with the FE of 42.8%for NO_(3)~-solution as the nitrogen ***,this strategy displays a broad universality for the synthesis of other amino *** work not only highlights the importance of hollow-structuring electrocatalyst towards upgrading wasted NO_(x)species into amino acids,but also promotes the further development of electrosynthetic systems for high-valued N-containing organic chemicals.

Study on cumulative effects of biological craniocerebral trauma under repeated blast

Plasma microRNA-15a/16-1-based machine learning for early detection of hepatitis B virus-related hepatocellular carcinoma

Background and aims:Hepatocellular carcinoma(HCC),which is prevalent worldwide and has a high mortality rate,needs to be effectively *** aimed to evaluate the significance of plasma microRNA-15a/16-1(miR-15a/16)as a biomarker of hepatitis B virus-related HCC(HBV-HCC)using the machine learning *** study was the first large-scale investigation of these two miRNAs in HCC plasma ***:Using quantitative polymerase chain reaction,we measured the plasma miR-15a/16 levels in a total of 766 participants,including 74 healthy controls,335 with chronic hepatitis B(CHB),47 with compensated liver cirrhosis,and 310 with *** diagnostic performance of miR-15a/16 was examined using a machine learning model and compared with that of alpha-fetoprotein(AFP).Lastly,to validate the diagnostic efficiency of miR-15a/16,we performed pseudotemporal sorting of the samples to simulate progression from CHB to ***:Plasma miR-15a/16 was significantly decreased in HCC than in all control groups(P<0.05 for all).In the training cohort,the area under the receiver operating characteristic curve(AUC),sensitivity,and average precision(AP)for the detection of HCC were higher for miR-15a(AUC=0.80,67.3%,AP=0.80)and miR-16(AUC=0.83,79.0%,AP=0.83)than for AFP(AUC=0.74,61.7%,AP=0.72).Combining miR-15a/16 with AFP increased the AUC to 0.86(sensitivity 85.9%)and the AP to 0.85 and was significantly superior to the other markers in this study(P<0.05 for all),as further demonstrated by the detection error tradeoff ***,miR-15a/16 impressively showed potent diagnostic power in early-stage,small-tumor,and AFP-negative HCC.A validation cohort confirmed these ***,the simulated follow-up of patients further validated the diagnostic efficiency of miR-15a/***:We developed and validated a plasma miR-15a/16-based machine learning model,which exhibited better diagnostic performance for the early diagnosis of HCC compared to that of AFP.

Osteosarcoma is a malignant tumor originating from bone tissue that progresses rapidly and has a poor patient *** has shown great potential in the treatment of ***,the immunosuppressive microenvironment severely limits the efficacy of osteosarcoma *** dual pH-sensitive nanocarrier has emerged as an effective antitumor drug delivery system that can selectively release drugs into the acidic tumor ***,we prepared a dual pH-sensitive nanocarrier,loaded with the photosensitizer Chlorin e6(Ce6)and CD47 monoclonal antibodies(aCD47),to deliver synergistic photodynamic and immunotherapy of *** laser irradiation,Ce6 can generate reactive oxygen species(ROS)to kill cancer cells directly and induces immunogenic tumor cell death(ICD),which further facilitates the dendritic cell maturation induced by blockade of CD47 by ***,both calreticulin released during ICD and CD47 blockade can accelerate phagocytosis of tumor cells by macrophages,promote antigen presentation,and eventually induce T lymphocyte-mediated antitumor ***,the dual pH-sensitive nanodrug loaded with Ce6 and aCD47 showed excellent immune-activating and anti-tumor effects in osteosarcoma,which may lay the theoretical foundation for a novel combination model of osteosarcoma treatment.

The interaction mechanism between zein and folic acid in alkaline aqueous solutions:an experimental and molecular simulation study

Objectives:Folic acid is a vitamin that is not highly soluble in water and is sensitive to the ***,it is important to find suitable *** study aimed to exemplify the interaction of folic acid with zein in alkaline aqueous solutions and shed light on how zein can be used as a carrier for folic *** and Methods:Zein and folic acid were separately dissolved in NaOH solutions with a PH of *** solution and folic acid solution were combined in specific *** methods including multi-spectroscopy,dynamic light scattering,and electron microscopy combining with molecular dynamics simulations were used to study the interaction mechanism between zein and folic acid in alkaline aqueous ***:Fluorescence spectroscopy showed that the quenching of zein by folic acid was mainly static,and the main driving force behind this interaction was van der Waals forces and hydrogen *** formation of zein–folic acid complexes was confirmed by ultraviolet–visible spectroscopy,Fourier transform infrared spectroscopy,and circular dichroism *** results also showed that the structure of zein changed when it interacted with folic *** light scattering analysis revealed that the addition of folic acid caused proteins to *** aggregates of the complexes had an irregular shape and were large,as observed by scanning electron microscopy and transmission electron *** simulation was used to further investigate the interaction *** to these findings,the folic acid molecule interacted with zein in a shallow recess near the protein *** dominant forces at play in the zein–folic acid interaction were van der Waals forces and electrostatic forces,including hydrogen ***:The zein alkali-soluble system is very suitable for folic acid delivery.

Nutritional Assessment Tools for Patients with Cancer:A Narrative Review

Cancer patients are at high risk of malnutrition,which can lead to adverse health outcomes such as prolonged hospitalization,increased complications,and increased *** and timely nutritional assessment plays a critical role in effectively managing malnutrition in these ***,while many tools exist to assess malnutrition,there is no universally accepted *** different tools have their own strengths and limitations,there is a lack of narrative reviews on nutritional assessment tools for cancer *** address this knowledge gap,we conducted a non-systematic literature search using PubMed,Embase,Web of Science,and the Cochrane Library from their inception until May 2023.A total of 90 studies met our selection criteria and were included in our narrative *** evaluated the applications,strengths,and limitations of 4 commonly used nutritional assessment tools for cancer patients:the Subjective Global Assessment(SGA),Patient-Generated Subjective Global Assessment(PG-SGA),Mini Nutritional Assessment(MNA),and Global Leadership Initiative on Malnutrition(GLIM).Our findings revealed that malnutrition was associated with adverse health *** of these 4 tools has its applications,strengths,and *** findings provide medical staff with a foundation for choosing the optimal tool to rapidly and accurately assess malnutrition in cancer *** is essential for medical staff to be familiar with these common tools to ensure effective nutritional management of cancer patients.
