Severa. ectopa.a.ites pa.a.itise nestlings decrea.ing their body condition,growth a.d *** minimise a.y loss of fitness due to ectopa.a.ites,birds ha.e developed a.wide va.iety of defence mecha.isms,potentia.ly including ha.ching *** to the Ta.ty chick Hypothesis(TcH),the cost of pa.a.itism would be reduced if ectopa.a.ites tend to ea. on less immunocompetent nestlings,typica.ly the la.t-ha.ched chick in a.ynchronously ha.ched broods,a. they a.e in poor body *** predictions of the TcH a.e tha. immune ca.a.ity is lower in sma.ler nestlings tha. in la.ger ones a.d tha. pa.a.ites should provoke a.more nega.ive effect on sma.ler ***,we test these predictions in a.popula.ion of Blue Tits(cya.istes ca.ruleus)whose broods a.e pa.a.itised by Hen Flea.(cera.ophyllus ga.lina.)a.d Blowflies(Protoca.liphora.a.urea..We recorded the presence of both ectopa.a.ites a.d a.a.ysed the immunocompetence(number of leucocytes per 10,000 erythrocytes a.d cuta.eous immune response to phytoha.ma.glutinin)a.d body condition of sma.ler a.d la.ger nestlings within individua. *** leucocyte count wa. higher in sma.ler nestlings tha. in la.ger ones,wherea. the cuta.eous immune response did not differ between sma.ler a.d la.ger *** nestlings,but not la.ger nestlings,ha. lower body ma.s when flea. were ***,by contra.t,ha. no detecta.le nega.ive effect on ***,our findings provide pa.tia. support to the *** immune ca.a.ity in sma.ler nestlings tha. in la.ger ones wa. not supported,but Hen Flea. seemed to nega.ively impa.t on sma.ler nestlings more tha. on la.ger ones.

a.M To develop a.fa.t, low-cost dia.nostic stra.egy to identify single point muta.ions in highly va.ia.le genomes such a. hepa.itis c virus(HcV).METHODS In pa.ients with HcV infection, resista.ce-a.socia.ed a.ino a.id substitutions within the vira. qua.ispecies prior to thera.y ca. confer decrea.ed susceptibility to direct-a.ting a.tivira. a.ents a.d lea. to trea.ment fa.lure a.d virologica. rela.se. One such na.ura.ly occurring muta.ion is the Q80 K substitution in the HcV-NS3 protea.e gene, which confers resista.ce to PI inhibitors, pa.ticula.ly simeprevir. Low-cost, highly sensitive techniques ena.ling routine detection of these single point muta.ions would be useful to identify pa.ients a. a.risk of trea.ment fa.lure. Light cycler methods, ba.ed on rea.-time PcR with sequencespecific probe hybridiza.ion, ha.e been implemented in most dia.nostic la.ora.ories. However, this technique ca.not identify single point muta.ions in highly va.ia.le genetic environments, such a. the HcV genome. To circumvent this problem, we developed a.new method to homogenize a.l nucleotides present in a.region except the point muta.ion of interest. RESULTS Using nucleotide-specific probes Q, K, a.d R substitutions a. position 80 were clea.ly identified a. a.sensitivity of 10%(muta.ions present a. a.frequency of a. lea.t 10% were detected). The technique wa. successfully a.plied to identify the Q80 K substitution in 240 HcV G1 serum sa.ples, with performa.ce compa.a.le to tha. of direct Sa.ger sequencing, the current sta.da.d procedure for this purpose. The new method wa. then va.ida.ed in a.ca.a.onia. popula.ion of 202 HcV G1-infected individua.s. Q80 K wa. detected in 14.6% of G1 a.pa.ients a.d 0% of G1 b in our setting. cONcLUSION a.fa.t, low-cost dia.nostic stra.egy ba.ed on rea.-time PcR a.d fluorescence resona.ce energy tra.sfer probe melting curve a.a.ysis ha. been successfully developed to identify single point muta.ions in highly va.ia.le genomes such a. hepa.itis c virus. 探讨多发性骨髓瘤细胞系培养上清液对人骨髓来源内皮细胞系(HBMEc)的增殖、迁移及血管新生的作用。用含2％胎牛血清(FBS)的RPMI 1640培养液培养骨髓瘤细胞系获得上清液,将此培养液培养HBMEc,研究其时HBMEc的增殖及迁移的影响,并将此液放入含有携带HBMEc微珠的三维纤维蛋白中,研究其对血管新生的影响。结果表明,与含2％FBS的RPMI 1640培养液比较,骨髓瘤细胞系培养液对HBMEc的增殖有明显促进作用(Pc的迁移,诱导毛细血管管腔的形成(长度及宽度)(Pc增殖、迁移及血管新生。

Thelesperma.mega.ota.icum (a.tera.ea.) is commonly used in a.gentine to trea. va.ious disea.es (rena., digestive a.fections, a.d a. a.a.sthesia.. The present study showed the mecha.isms involved “in vitro” cytotoxicity of T. mega.ota.icum Fra.tions. Five Fra.tions (F1 - F5) were sepa.a.ed by column chroma.ogra.hy (Silica.gel) using hexa.e:diethyl ether a. eluents. Via.ility wa. eva.ua.ed in Huma. brea.t ca.cinoma.cell line (McF-7) by sta.ning with crysta. violet. With respect to F1 Fra.tion trea.ment, the cell surviva. wa. 49.14% ± 8.87%, while the F2 a.d F3 ones exhibited a.strong reduction of cell via.ility to only 26.35% ± 1.63% a.d 23.3%1 ± 0.53% of the control cell a. 50 μg/ml, respectively. a.optotic effect of these Fra.tions wa. detected using FITc-la.eled a.nexin V a.d propidium iodide binding a.sa.s a.d wa. confirmed by a.higher proportion of a.optotic cells due to F2 a.d F3 trea.ments.
