Ketogenic diet therapy in children with epilepsy caused by SLc2A1 mutations:a single-center single-arm retrospective study

Background This retrospective study assessed the efficacy and safety of ketogenic diet therapies in children with epilepsy caused by SLc2A1 genetic mutations and glucose transporter type 1 deficiency *** Pediatric patients with epilepsy symptoms admitted to our medical center between January 2017 and October 2021 were included if they presented with an SLc2A1 genetic mutation on whole-exome *** analyzed the patients’convulsions and treatment with antiepileptic *** patients were followed up at different time periods after ketogenic diet *** Six patients with SLc2A1 mutations were included in this *** patients had seizures of different types and frequencies,and they took antiepileptic drugs to relieve their *** were then treated with a ketogenic diet for at least four *** analyzed epilepsy control rates at 1,2,3,6,and 12 months after ketogenic diet *** patients were seizure-free within a month of receiving the diet *** patients were followed up for six months,three were followed up for 12 months after the treatment,and there was no recurrence of epilepsy during this *** antiepileptic drug withdrawal,none of the patients experienced seizure relapse when receiving ketogenic diet treatment *** severe adverse events occurred during the *** Ketogenic diet therapy is very effective and safe for the treatment of epilepsy caused by SLc2A1 ***,patients with glucose transporter type 1 deficiency syndrome caused by SLc2A1 mutations should begin ketogenic diet treatment as soon as possible.

Identi?cation and Analysis of the Intermediates from Photodegradation of 3,3′-diamino-4,4′-azoxyfurazan (DAAF)by SERS and HPLc–MS/MS

Understanding the structural changes and degradation mechanisms of organic pollutants is critical in environmental *** this work, a xenon lamp was used as an analog light source to simulate the eff ect of sunlight on 3,3′-diamino-4,4′-azoxyfurazan (DAAF) in aqueous solution. combining high-performance liquid chromatography coupled with tandem mass spectrometry (HPLc–MS/MS), surface-enhanced Raman spectroscopy (SERS) with density functional theory (DFT), the degradation intermediates and mechanisms of DAAF were investigated. The sequential breakdown of the–NH 2and–O–N=bonds, followed by the removal of atomic O from–N=N(O)–, was emphasized and investigated using two-dimensional correlation spectroscopy (2D-cOS) and SERS spectra. Three intermediates with mass-to-charge ratios (m/z) 181, 251 and180 were identifi ed and characterized by integrating experimental SERS data with DFT-calculated Raman spectra based on structures hypothesized from MS spectra. The fi ndings from HPLc–MS and SERS not only corroborate each other but also provide a foundation for a detailed examination of the DAAF degradation process.

Workers'task specialization and division of labor are critical features of social insects'ecological *** has been proposed that the division of labor relies on response threshold models:individuals varying their sensitivity(and responsiveness)to biologically relevant stimuli and performing a specific task when a stimulus exceeds an internal *** this work,we study carbohydrate and protein responsiveness and their relation to worker task specialization in Vespula germanica,an invasive social *** sucrose and peptone responsiveness of two different subcastes,preforagers and foragers,was determined by stimulating the antenna of the wasps with increasing concentrations of the solution and quantifying whether each concentration elicited a licking *** studied responsiveness in five different ways:(1)response threshold,(2)concentration 50(concentration to which at least 50%of wasps responded),(3)maximum response,(4)mean scores and(5)median *** results suggest that V germanica foragers are more sensitive to sucrose(lower thresholds)than preforager ***,we found no differences for peptone thresholds(i.e.,a protein resource).Nonetheless,this is the first study to investigate response thresholds for protein *** intercaste variation in sucrose responsiveness shown in our work contributes to the existing knowledge about response threshold theory as a mechanism for task specialization observed in V germanica.

Aniba rosaeodora essential oil(RO) has been traditionally used in natural medicine as a substitute for antibiotics due to its notable antidepressant and antibacterial properties. Salmonella, a prevalent pathogen in foodborne illnesses, presents a major challenge to current antibiotic treatments. However, the antibacterial efficacy and mechanisms of action of RO against Salmonella spp. remain underexplored. This study aims to elucidate the chemical composition of RO, evaluate its antibacterial activity and mechanisms against Salmonella in vitro, and further delineate its anti-inflammatory mechanisms in vivo during Salmonella infection. Gas chromatography-mass spectrometry(Gc-MS) was utilized to characterize the chemical constituents of RO. The antibacterial activity of RO was assessed using minimal inhibitory concentration(MIc) and time-kill assays. Various biochemical assays were employed to uncover the potential bactericidal mechanisms. Additionally, mouse and chick models of Salmonella infection were established to investigate the prophylactic effects of RO treatment. RO exhibited significant antibacterial activity against both Gram-positive and Gram-negative bacteria, with an MIc of 4 mg·mL^(-1) for Salmonella spp. RO treatment resulted in bacterial damage through the disruption of lipid and purine metabolism. Moreover, RO reduced injury and microbial colonization in infected mice and chicks. RO treatment also modulated the host inflammatory response by inhibiting proinflammatory pathways. In conclusion, our findings demonstrate that RO is effective against Salmonella infection, highlighting its potential as an alternative to antibiotics for antibacterial therapy.
