NecAB family of neuronal calcium-binding proteins in health and disease

The N-terminal eF-hand calcium-binding proteins 1–3(NecAB1–3) constitute a family of predominantly neuronal proteins characterized by the presence of at least one eF-hand calcium-binding domain and a functionally less well characterized c-terminal antibiotic biosynthesis monooxygenase domain. All three family members were initially discovered due to their interactions with other proteins. NecAB1 associates with synaptotagmin-1, a critical neuronal protein involved in membrane trafficking and synaptic vesicle exocytosis. NecAB2 interacts with predominantly striatal G-protein-coupled receptors, while NecAB3 partners with amyloid-β A4 precursor protein-binding family A members 2 and 3, key regulators of amyloid-β production. This demonstrates the capacity of the family for interactions with various classes of proteins. NecAB proteins exhibit distinct subcellular localizations: NecAB1 is found in the nucleus and cytosol, NecAB2 resides in endosomes and the plasma membrane, and NecAB3 is present in the endoplasmic reticulum and Golgi apparatus. The antibiotic biosynthesis monooxygenase domain, an evolutionarily ancient component, is akin to atypical heme oxygenases in prokaryotes but is not wellcharacterized in vertebrates. Prokaryotic antibiotic biosynthesis monooxygenase domains typically form dimers, suggesting that calcium-mediated conformational changes in NecAB proteins may induce antibiotic biosynthesis monooxygenase domain dimerization, potentially activating some enzymatic properties. However, the substrate for this enzymatic activity remains uncertain. Alternatively, calcium-mediated conformational changes might influence protein interactions or the subcellular localization of NecAB proteins by controlling the availability of protein–protein interaction domains situated between the eF hands and the antibiotic biosynthesis monooxygenase domain. Human papillomavirus 16 e6 is associated with the nuclear matrix of esophageal carcinoma cells

AIM: To explore the etiologic role of HPV infection in esophageal carcinoma, and the association of HPV-16 e6with the nuclear metrix of carcinoma cells.MeTHODS: Two esophageal carcinoma cell lines, ec/cUHK1 and ec/cUHK2, were tested for HPV-16 e6subgenetic fragment by polymerase chain reaction amplification of virus DNA associated nuclear matrix. RT-PcR and immunocytochemistry were also used to visualizethe expression of e6 subgene in the cells.ReSULTS: The HPV-16 e6 subgenetic fragment wes found to be present in nuclear metrix-associeted DNA, e6oncoprotein localized in the nucleus where it is tightly associated with nuclear matrix after sequential extraction in ec/cUHK2 cells. It was not detected, however, in ec/cUHK1 cells.cONcLUSION: The interaction between HPV-16 e6 and nuclear matrix may contribute to the virus induced carcinogenesis in esophageal carcinoma.

Recent Advancements in the Optimization capacity configuration and coordination Operation Strategy of Wind-Solar Hybrid Storage System

Present of wind power is sporadically and cannot be utilized as the only fundamental load of energy *** paper proposes a wind-solar hybrid energy storage system(HeSS)to ensure a stable supply grid for a longer period.A multi-objective genetic algorithm(MOGA)and state of charge(SOc)region division for the batteries are introduced to solve the objective function and configuration of the system capacity,***/Simulink was used for simulation *** optimization results show that for a 0.5 MW wind power and 0.5 MW photovoltaic system,with a combination of a 300 Ah lithium battery,a 200 Ah lead-acid battery,and a water storage tank,the proposed strategy reduces the system construction cost by approximately 18,000 ***,the cycle count of the electrochemical energy storage systemincreases from4515 to 4660,while the depth of discharge decreases from 55.37%to 53.65%,achieving shallow charging and discharging,thereby extending battery life and reducing grid voltage fluctuations *** proposed strategy is a guide for stabilizing the grid connection of wind and solar power generation,capability allocation,and energy management of energy conservation systems.

Liposarcoma is one of the most common soft tissue sarcomas,however,its occurrence rate is still rare compared to other *** to its rarity,in vitro experiments are an essential approach to elucidate liposarcoma *** cell culture-based research(2D cell culture)is still playing a pivotal role,while several shortcomings have been recently under *** vivo,mouse models are usually adopted for pre-clinical analyses with expectations to overcome the issues of 2D cell ***,they do not fully recapitulate human dedifferentiated liposarcoma(DDLPS)***,three-dimensional(3D)culture systems have been the recent research focus in the cell biology field with the expectation to overcome at the same time the disadvantages of 2D cell culture and in vivo animal models and fill in the gap between *** the liposarcoma rarity,we believe that 3D cell culture techniques,including 3D cell cultures/co-cultures,and Patient-Derived tumor Organoids(PDOs),represent a promising approach to facilitate liposarcoma investigation and elucidate its molecular mechanisms and effective therapy *** this review,we first provide a general overview of 3D cell cultures compared to 2D cell *** then focus on one of the recent 3D cell culture applications,Patient-Derived Organoids(PDOs),summarizing and discussing several PDO ***,we discuss the current and future applications of PDOs to sarcoma,particularly in the field of liposarcoma.

Merging bound states in the continuum in an open acoustic resonator

Bound states in the continuum （BIcs） are perfectly localized resonances despite embedding in the continuum ***,an isolated BIc is very sensitive to the structure ***,we report merging acoustic BIcs in a single open resonator,robust against the structure *** find that both symmetry-protected BIc and Friedrich-Wintgen BIc are sustained in a single coupled waveguide-resonator *** varying the height and length of the resonator,these two BIcs move toward each other and merge into a single one at a critical *** to an individual BIc,the merged BIc is robust against fabrication error because its Q-factor is proportional to ΔL-4,whereΔL embodies the structure *** essence of this extraordinary phenomenon is perfectly explained by the two-and three-level approximations of the effective nonHermitian ***,we present direct experimental demonstrations of the moving and merging of BIcs in a coupled 3D waveguide-resonator,which are evidenced by the vanishing of the linewidth of Fano resonance in the transmission *** results may find exciting applications in designing high-quality acoustic sources,sensors and filters.

Decision and coordination of an e-commerce Supply chain considering Returns and Network externalities

considering the large number of returns in online sales and the network externalities of e-platforms,we develop a decentralized model and a centralized model to explore the impacts of returns and network externalities on e-commerce supply chain(ecSc)*** show that in the decentralized model,the service level,price,market demand,and ecSc members’profits increase with the network externality ***,the service level and price increase,while the market demand and ecSc members’profits decrease with the product return *** centralized model is the optimal operating mode when it is properly *** design the“commission and return cost-sharing”contract to optimize ecSc,in which the proportion of the e-platform’s sharing of the return handling cost is exactly equal to the proportion of the system profit after *** on the decentralized model,we develop two extended models in which we endogenize the impacts of the service level and return rate on the network externality *** comparisons between the extended and decentralized models,we show that high-quality service can improve ecSc’s profitability,while a high return rate hurts its economic performance.

Lowering the synthesis temperature of boron nitride nanotubes(BNNTs)is crucial for their *** primary reason for adopting a high temperature is to enable the effective activation of highmelting-point solid *** this study,we developed a novel approach for efficiently activating boron by introducing alkali metal compounds into the conventional MgO–B *** approach can be adopted to form various low-melting-point AM–Mg–B–O growth *** growth systems have improved catalytic capability and reactivity even under low-temperature conditions,facilitating the synthesis of BNNTs at temperatures as low as 850℃.In addition,molecular dynamics simulations based on density functional theory theoretically demonstrate that the systems maintain a liquid state at low temperatures and interact with N atoms to form BN *** findings offer novel insights into the design of boron activation and are expected to facilitate research on the low-temperature synthesis of BNNTs.

Air transport systems are highly dynamic at temporal scales from minutes to *** dynamic behavior not only characterizes the evolution of the system but also affect the system＇s *** the evolutionary mechanisms is thus fundamental in order to better design optimal air transport networks that benefits companies,passengers and the *** this review,we briefly present and discuss the state-of-the-art on time-evolving air transport *** distinguish the structural analysis of sequences of network snapshots,ideal for long-term network evolution（*** evolution）,and temporal paths,preferred for short-term dynamics（*** evolution）.We emphasize that most previous research focused on the first modeling approach（***-term） whereas only a few studies look at high-resolution temporal *** conclude the review highlighting that much research remains to be done,both to apply already available methods and to develop new measures for temporal paths on air transport *** particular,we identify that the study of delays,network resilience and optimization of resources（aircraft and crew） are critical topics.

considering diseconomies of scale and network externalities in the e-commerce supply chain(ecSc),we construct an e-platform-led benchmark model and derive the optimal ***,the model is extended by endogenizing the impact of service level on network *** service investment that includes fixed and variable investments,the model is further *** the extended models with the benchmark model,we found the following *** the e-platform dominates the ecSc,its profit is lower than the *** corporate profits,service level,and price increase with network *** in diseconomies of scale decrease the corporate profits and service level,but increase the price.A high-quality service combined with network externalities can achieve synergy and improve the e-platform’s economies of scale,further generating a higher *** network externalities promotes the fair profit distributionin ecSc and achieves stable development.
