There has been a growing interest over the past few years in the impact of male nutrition on fertility. Infertility has been linked to male overweight or obesity, and conventional semen parameter values seem to be altered in case of high body mass index （BMI）. A few studies assessing the impact of BMI on sperm DNA integrity have been published, but they did not lead to a strong consensus. Our objective was to explore further the relationship between sperm DNA integrity and BMI, through a 3-year multicentre study. Three hundred and thirty male partners in subfertile couples were included. Using the terminal uridine nick-end labelling （TUNEL） assay, we observed an increased rate of sDerm DNA damage in obese men （odds ratio （95% confidence interval）： 2.5 （1.2-5.1））.

Obesity and associated metabolic disorders represent a major societal challenge in health and quality of life with large psychological consequences in addition to physical disabilities. They are also one of the leading causes of morbidity and mortality. Although, different etiologic factors including excessive food intake and reduced physical activity have been well identified, they cannot explain the kinetics of epidemic evolution of obesity and diabetes with prevalence rates reaching pandemic proportions. Interestingly, convincing data have shown that environmental pollutants, specifically those endowed with endocrine disrupting activities, could contribute to the etiology of these multifactorial metabolic disorders. Within this review, we will recapitulate characteristics of endocrine disruption. We will demonstrate that metabolic disorders could originate from endocrine disruption with a particular focus on convincing data from the literature. Eventually, we will present how handling an original mouse model of chronic exposition to a mixture of pollutants allowed demonstrating that a mixture of pollutants each at doses beyond their active dose could induce substantial deleterious effects on several metabolic end-points. This proof-of-concept study, as well as other studies on mixtures of pollutants, stresses the needs for revisiting the current threshold model used in risk assessment which does not take into account potential effects of mixtures containing pollutants at environmental doses, e.g., the real life exposure. Certainly, more studies are necessary to better determine the nature of the chemicals to which humans are exposed and at which level, and their health impact. As well, research studies on substitute products are essential to identify harmless molecules.

Experiences of Children’s Care Received at Windhoek Residential Child Care Facilities, Namibia

The caring is a very important aspect for every child’s well-being and development. A child should be cared for by biological parents or extended family members when possible. In the absence of these two possibilities, other options are in place such as Residential Child Care Facilities (RCCFs). However, RCCFs are generally regarded as last resorts, as it is noted that they have a negative impact on the well-being of the children. Caregivers in RCCFs should ensure that these children are being adequately cared for as they substitute their biological or extended families. It is also a child’s rights to be provided love, care and support coupled with a sense of belonging, recognition and independence. However, children without or inadequate parental care are likely to be vulnerable from all forms of violence. The aim of this qualitative, exploratory, descriptive, contextual and phenomenological study was to explore and describe the children’s experiences regarding the care received at the Residential Child Care Facilities and to provide recommendations regarding research findings. Data were collected through one on one in-depth interviews and Tech method was used to analyze data. The results of the study show that children in RCCFs have experienced both positive and negative care. The care is experienced through relationships with staff in the facilities, provision of their basic needs, knowledge of their cultural identity and their adult future preparation once they leave the facilities. Thus, it is recommended that a conducive environment that contributes to the quality of care for children in RCCFs be established, taking into consideration human resources capacity, policy guidelines and standards in place while providing care to those children.

Evaluation of Acute Toxicity, Antioxidant and Antibacterial Potential of Leaves Extracts from Two Anacardiaceae’s Species: Lannea microcarpa Engl. &K. Krause and Mangifera indica L.

Background: Leaves of Mangifera indica L or Lannea microcarpa Engl. & K. Krause are used in traditional medicine in Burkina Faso to treat bacterial, parasitic or metabolic diseases. Objective: The aim of this study is to evaluate the acute general toxicity, antioxidant potential and antibacterial activitiy of leave’s aqueous extracts from Lannea microcarpa Engl. and K. Krause and Mangifera indica L. The use of these plants in traditional medicine motivated our choice to lead scientific studies. Methods: The aqueous decoction of the leaves is the form of use recommended by traditional healers. We used the same type of extracts for studies. Acute toxicity was studied in NMRI strain mice, with the dose of 2000 mg/kg body weight, for each plant species. The antioxidant activity is evaluated by the method of reduction of radical DPPH. The phytochemical compounds were detected with specific reagent: Alcaloids with Dragendorff’s reagent, Flavonoids with ammonia (NH4OH), Polyphenols and tannins are revelated by ferric chloride (FeCl3). Saponosides were revelated by their foaming power property. Bacterial inhibiting activity is tested by measuring the diameters’ inhibition of extracts on reference strains. Results: The aqueous extracts were not toxic at the maximum dose of 2000 mg/kg body weight, for each plant species. The extracts showed an antioxidant activity with an IC50, 20 μg/ml for Lannea microcarpa and 18 μg/ml for Mangifera indica. The extracts showed no bacterial activity on three strains of bacteria tested: Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus. The phytochemicals we have identified are for L. microcarpa: tannins and phenolics compounds, triterpenes, saponosides. We identified in M. indica’s extracts: tannins and phenolics compounds, triterpenes, saponosides and flavonoids.

In the present study, the morphological and ultrastructural characteristics of mesenchymal stem cells (MSCs) induced towards osteogenic, adipogenic, and chondrogenic lineages were investigated. The main objective of this paper was to focus on the differentiation capacity of ovine [oMSCs] and equine MSCs [eMSCs]. Bone marrow [BM] MSCs were isolated from ovine and equine patients, expanded in monolayer culture and induced into osteogenic, adipogenic and chondrogenic differentiation. For chondrogenic differentiation, cells were cultured in micromass culture. Also, their ultrastructural phenotypes were studied by transmission electron microscopy [TEM]. This study was aimed to provide an indepth morphological description of BM-derived ovine and equine MSCs induced towards three lineages, it demonstrated that TEM analysis is useful in elucidating detailed structural information.

BACKGROUND Aortic arch stenting is continuously emerging as a safe and effective option to alleviate aortic arch stenosis and arterial *** SUMMARY We present a 15-year-old girl with aortic arch hypoplasia who had undergone implantation of an uncovered 22 mm Cheatham-Platinum stent due to severe(native)aortic arch *** follow-up seven months later,she presented a significant re-stenosis of the aortic arch.A second stent(LD Max 26 mm)was implanted and both stents were dilated up to 16 *** an initially unremarkable post-interventional course,the patient presented with hoarseness five days after the *** and CT scans ruled out an intracranial pathology,as well as thoracic hematoma,arterial dissection,and aneurysm around the intervention *** confirmed left vocal fold paresis attributable to an injury to the left recurrent laryngeal nerve(LRLN)during aortic arch stenting,as the nerve loops around the aortic arch in close proximity to the area of the implanted *** a non-invasive therapeutic approach entailing regular speech therapy,the patient recovered and demonstrated no residual clinical symptoms of LRLN palsy after six *** Left recurrent laryngeal nerve palsy is a rare complication of aortic arch stenting not previously reported.

Using Boosted Regression Trees and Remotely Sensed Data to Drive Decision-Making

Challenges in Big Data analysis arise due to the way the data are recorded, maintained, processed and stored. We demonstrate that a hierarchical, multivariate, statistical machine learning algorithm, namely Boosted Regression Tree (BRT) can address Big Data challenges to drive decision making. The challenge of this study is lack of interoperability since the data, a collection of GIS shapefiles, remotely sensed imagery, and aggregated and interpolated spatio-temporal information, are stored in monolithic hardware components. For the modelling process, it was necessary to create one common input file. By merging the data sources together, a structured but noisy input file, showing inconsistencies and redundancies, was created. Here, it is shown that BRT can process different data granularities, heterogeneous data and missingness. In particular, BRT has the advantage of dealing with missing data by default by allowing a split on whether or not a value is missing as well as what the value is. Most importantly, the BRT offers a wide range of possibilities regarding the interpretation of results and variable selection is automatically performed by considering how frequently a variable is used to define a split in the tree. A comparison with two similar regression models (Random Forests and Least Absolute Shrinkage and Selection Operator, LASSO) shows that BRT outperforms these in this instance. BRT can also be a starting point for sophisticated hierarchical modelling in real world scenarios. For example, a single or ensemble approach of BRT could be tested with existing models in order to improve results for a wide range of data-driven decisions and applications.

Objective: The aim of this study is to screen chemical composition and to evaluate the acute general toxicity of leaves aqueous extracts from Sesbania pachycarpa DC. (Fabaceae), used in traditional medicine in Burkina Faso to treat malaria, helminthiasis, and aqueous extracts of the leaves from Indigofera berhautiana Gillet (Fabaceae) used to treat hepatitis and typhoid fever. Methods: Aqueous decoction of leaves from Sesbania pachycarpa DC. or Indigofera berhautiana Gillett which are the form of use recommended by traditional health practitioners, were used for tests. Aqueous extracts used to evaluate the acute toxicity, were studied on mice of strain NMRI. For the toxicity study, the doses of the aqueous extracts were respectively 5 mg/kg, 50 mg/kg, 300 mg/kg and 2000 mg/kg of body weight. Phytochemical screening was done to identify the phytochemicals contained in extracts. Results: Aqueous extracts of two species: Sesbania pachycarpa DC., Indigofera berhautiana Gillet were not toxic at the maximum dose of 2000 mg/kg of body weight. Phytochemical screening, showed coumarins, flavonoids, tannins, polyphenols compounds, steroids and/or triterpenes and saponosides in the aqueous extracts of the two species. Alkaloids were identified in Sesbania pachycarpa aqueous extract.

Effects of Ginkgo Biloba Special Extract EGb 761^(█) in Very Mild Cognitive Impairment(vMCI)

Objective: To assess effects of EGb 761? on cognition and quality of life in subjects with very mild cognitive impair-ment. Methods: We randomized 300 subjects aged 45 to 65 with cognitive complaints and low functioning (more than one standard deviation below appropriate norm) in at least one cognitive test to double-blind treatment with once daily 240 mg EGb 761? or placebo for 12 weeks. Results: The exploratory intention-to-treat analysis showed significant im-provement (p < 0.025, one-sided) beyond practice effects for EGb 761? in a measure of attention (Vienna Test System - Work Performance Series) and trends in favour of EGb 761? in measures of memory (Wechsler Memory Scale III - Faces I, Appointments Test – delayed recall), and perceived physical health (SF36 - factor score Physical Health). Cognitive effects were more pronounced and more consistent (p < 0.025 in 4 of 5 tests) in subjects with lower memory function at baseline. Specifically, practice effects in the more demanding tests were attenuated or absent in these sub-jects. Conclusion: Ginkgo biloba extract EGb 761? improved cognitive functioning and aspects of quality of life in sub-jects with very mild cognitive impairment.
