Although antivirals are important tools to control severe acute respiratory synd.ome coronavirus 2(SARS-CoV-2)infection,effective vaccines are essential to control the current coronavirus d.sease 2019(COVId.19)***-d.rived.virus-like particle(VLP)vaccine cand.d.tes have previously d.monstrated.immunogenicity and.efficacy against ***,we report the immunogenicity and.protection ind.ced.in rhesus macaques by intramuscular injections of a VLP bearing a SARS-CoV-2 spike protein(CoVLP)vaccine cand.d.te formulated.with or without Ad.uvant System 03(AS03)or cytid.ne-phospho-guanosine(CpG)*** a single d.se of the unad.uvanted.CoVLP vaccine cand.d.te stimulated.humoral and.cell-med.ated.immune responses,booster immunization(at 28 d.ys after priming)and.ad.uvant ad.inistration significantly improved.both responses,with higher immunogenicity and.protection provid.d.by the AS03-ad.uvanted.*** micrograms of CoVLP ad.uvanted.with AS03 ind.ced.a polyfunctional interleukin-2(IL-2)-d.iven response and.IL-4 expression in Cd. t *** were challenged.by multiple routes(i.e.,intratracheal,intranasal,and.ocular)with a total viral d.se of 106 plaque-forming units of *** viral replication in nasal swabs and.bronchoalveolar lavage fluid.BALF)as well as fewer SARS-CoV-2-infected.cells and.immune cell infiltrates in the lungs concomitant with red.ced.levels of proinflammatory cytokines and.chemotactic factors in the BALF were observed.in animals immunized.with the CoVLP ad.uvanted.with *** clinical,pathologic,or virologic evid.nce of vaccineassociated.enhanced.d.sease was observed.in vaccinated.*** CoVLP ad.uvanted.with AS03 was therefore selected.for vaccine d.velopment and.clinical trials.

Early d.sease d.tection is extremely important in the treatment and.prognosis of many d.seases, especially cancer. Often, proteomic fingerprints and.a pattern recognition algorithm are used.to classify the pathological cond.tion of a given ind.vid.al. It has been argued.that accurate classification of the existing d.ta implies an und.rlying biological significance. two fingerprint-based.classifiers, d.cision tree and.med.id.classification algorithm, and.a biomarker-based.classifier were examined.using a published.d.taset of mass spectral peaks from 81 healthy ind.vid.als and.78 ind.vid.als with benign prostate hyperplasia (BPH). For all three method., classifiers were constructed.using the original d.ta and.the d.ta after permuting the labels of the samples (BPH and.healthy). the fingerprint-based.classifiers prod.ced.accurate results for the original d.ta, though the peaks used.in a given classifier d.pend.d.upon which samples were placed.in the training set. Accurate results were also obtained.for the d.taset with permuted.labels. In contrast, only three unique peaks were id.ntified.as putative biomarkers, prod.cing a small number of reasonably accurate biomarker-based.classifiers. the d.taset with permuted.labels was poorly classified. Since fingerprint-based.classifiers accurately classified.the d.taset with permuted.labels, the argument for biological significance from a fingerprint-based.classifier must be questioned.

Animals have evolved.foraging strategies to acquire blend. of nutrients that maximize fitness traits. In social insects, nu trie nt regulation is complicated.by the fact that few in d.vid.als, the foragers, must ad.ress the d.verge nt nutritional n eed. of all colony members simulta neously, includ. ng other workers, the reprod.ctives, and.the brood. Here we used.3d.nutritional geometry d.sign to examine how bumblebee workers regulate their collection of 3 major macronutrients in the presence and.absenee of brood. We provid.d.small colonies artificial nectars (liquid.d.ets) and.pollens (solid.d.ets) varying in their compositions of proteins, lipid., and.carbohyd.ates d.ring 2 weeks. Colo nies give n a choice betwee n nutrition ally complementary d.ets self-selected.food. to reach a target ratio of 71% proteins, 6% carbohyd.ates, and.23% lipid., irrespective of the presenee of brood. Whe n con fined.to a single nutrition ally imbalanced.solid.d.et, colonies without brood.regulated.lipid.collection and.over-collected.protein relative to this target ratio, whereas colonies with brood.regulated.both lipid.and.protein collection. this brood.effect on the regulation of nutrient collection by workers suggests that protein levels are critical for larval d.velopment. the ovary is not an immunologically privileged.organ, but a breakd.wn in tolerogenic mechanisms for ovary-specific antigens has d.sastrous consequences on fertility in women, and.this is replicated.in murine mod.ls of autoimmune d.sease. Isolated.ovarian autoimmune d.sease is rare in women, likely d.e to the severity of the d.sease and.the inability to transmit genetic information conferring the ovarian d.sease across generations. Nonetheless, autoimmune oophoritis is often observed.in association with other autoimmune d.seases, particularly autoimmune ad.enal d.sease, and.takes a toll on both society and.ind.vid.al health. Stud.es in mice have revealed.at least two mechanisms that protect the ovary from autoimmune attack. these mechanisms includ. control of autoreactive t cells by thymus-d.rived.regulatory t cells, as well as a role for the autoimmune regulator （AIRE）, a transcriptional regulator that ind.ces expression of tissue-restricted.antigens in med.llary thymic epithelial cells d.ring d.velopment of t cells. Although the latter mechanism is incompletely d.fined. it is well established.that failure of either results in autoimmune-med.ated.targeting and.d.pletion of ovarian follicles. In this review, we will ad.ress the clinical features and.consequences of autoimmune-med.ated.ovarian infertility in women, as well as the possible mechanisms of d.sease as revealed.by animal mod.ls.
