Marine algae and bacteria produce approximately eight billion tonnes of the organosulfur molecule dimethylsulfoniopropionate(DMSP)in Earth's surface oceans *** is an antistress compound and,once released into the environment,a major nutrient,signaling molecule,and source of climate-active *** methionine transamination pathway for DMSP synthesis is used by most known DMSP-producing algae and *** S-directed S-adenosylmethionine(SAM)-dependent 4-methylthio-2-hydroxybutyrate(MTHB)S-methyltransferase,encoded by the dsyB/DSYB gene,is the key enzyme of this pathway,generating S-adenosylhomocysteine(SAH)and 4-dimethylsulfonio-2-hydroxybutyrate(DMSHB).DsyB/DSYB,present in most haptophyte and dinoflagellate algae with the highest known intracellular DMSP concentrations,is shown to be far more abundant and transcribed in marine environments than any other known S-methyltransferase gene in DMSP synthesis ***,we demonstrate in vitro activity of the bacterial DsyB enzyme from Nisaea denitrificans and provide its crystal structure in complex with SAM and SAH-MTHB,which together provide the first important mechanistic insights into a DMSP synthesis *** and mutational analyses imply that DsyB adopts a proximity and desolvation mechanism for the methyl transfer *** analysis suggests that this mechanism may be common to all bacterial DsyB enzymes and also,importantly,eukaryotic DSYB enzymes from e.g.,algae that are the major DMSP producers in Earth's surface oceans.

Primary and metastatic liver tumors are an increasing global health problem,with hepatocellular carcinoma(HCC)now being the third leading cause of cancer-related mortality *** treatment options for HCC remain limited,with Sorafenib as the only prospectively validated agent shown to increase overall *** resection and/or transplantation,locally ablative therapies and regional or locoregional therapies have filled the gap in liver tumor treatments,providing improved survival outcomes for both primary and metastatic *** invasive local therapies have an increasing role in the treatment of both primary and metastatic liver *** patients with low volume disease,these therapies have now been established into consensus practice *** review highlights technical aspects and outcomes of commonly utilized,minimally invasive local therapies including laparoscopic liver resection(LLR),radiofrequency ablation(RFA),microwave ablation(MWA),high-intensity focused ultrasound(HIFU),irreversible electroporation(IRE),and stereotactic body radiation therapy(SBRT).In addition,the role of combination treatment strategies utilizing these minimally invasive techniques is reviewed.

The development of rapid genome sequencing has greatly enhanced our understanding of the molecular biology underlying many *** exome sequencing has highlighted the individualistic nature of malignancies on a patient-to-patient basis and begun to revolutionize therapeutic *** recent years,whole genome sequencing of urothelial malignancies has identified a host of somatic mutations which contribute to growth,progression,and metastasis of urothelial carcinoma of the bladder and upper tract urothelial *** genetic sequencing continues,additional targets will be identified,allowing development of novel therapeutic agents targeting cancer on a molecular level,with the goal of delivering highly individualized care based on the underlying mutational profile of the patient’s *** this review,we aim to discuss known genetic alterations of urothelial malignancy and the implications these mutations carry in terms of prognostication and development of targeted therapeutic *** will focus on RNA-expression profiling and genomic DNA profiling,with a focus on comprehensive whole exome and whole genome sequencing relative to selected urothelial carcinoma-associated genes and circulating tumor DNA analysis.

The coupled magnetorheological(MR) damping system addressed in this paper contains rubber spring and magnetorheological damper. The device inherits the damping merits of both the rubber spring and the magnetorheological *** a fractional-order constitutive equation is introduced to study the viscoelasticity of the combined damper. An introduction to the definitions of fractional calculus, and the transfer function representation of a fractional-order system are given. The fractional-order system model of a magnetorheological vibration platform is set up using fractional calculus, and the function of displacement is presented. It is indicated that the fractional-order constitutive equation and the transfer function are feasible and effective means for investigating of magnetorheological vibration device.

Sound Modulation in Singing Katydids Using Ionic Polymer-Metal Composites （IPMCs）

Many insect families have evolved to produce and detect complex singing patterns for the purposes of mating, display of dominance, predator escape, and other needs. While the mechanisms of sound production by insects have been thoroughly studied, man-machine exploitation of such mechanisms has remained unreported. We therefore describe a method to modulate the frequency spectrum in the chirp call of a singing insect, Gampsocleis gratiosa （Orthoptera： Tettigoniidae）, a large katydid indigenous to China and commonly known as Guo Guo or Chinese Bush Cricket. The chirp modulation was achieved through the contact of a ribbon of lonic Polymer-Metal Composite （IPMC） against wing of the insect. The IPMC effectively served as an actuator when a small DC voltage was applied to the ribbon＇s faces. By applying a sequential on/off voltage waveform to the IPMC ribbon, the katydid＇s chirp was modulated in a corresponding manner. This configuration can be used as part of a broader application of using singing insects to harness their acoustic power to produce and propagate machine-induced messages into the acoustic environment.

Geographical distribution of GmTfl1 alleles in Chinese soybean varieties

Stem growth habit is an important agronomic trait in soybean and is subject to artificial selection. This study aimed to provide a theory for genotypic selection of stem growth habit for breeding purposes by analyzing the alleles of Gm Tfl1 gene in Chinese soybean varieties and establishing a database of Gm Tfl1 variation. Using knowledge of insertion and deletion(Indel) in the non-coding region and four single-nucleotide polymorphisms(SNPs) in the coding sequences of the Gm Tfl1 gene, four CAPS and one Indel markers were developed and used to test 1120 Chinese soybean varieties. We found that the dominant Gm Tfl1 allele was prevalent in accessions from the Northern ecoregion, whereas the recessive allele, Gmtfl1, was more common in the Southern ecoregion, and the proportions of Gm Tfl1 and recessive alleles were respectively 40.1% and 59.9% in the Huang-Huai ecoregion. The proportion of Gm Tfl1 decreased and that of Gmtfl1 increased, gradually from north to south. Allele Gm Tfl1-a was present in higher proportions in the Huang-Huai spring, Huang-Huai summer, and Northern spring sub-ecoregions than that in the other sub-ecoregions. Gm Tfl1-b was common in the Northeast spring, Northern spring and Southern summer sub-ecoregions. Gmtfl1-ta was found mainly in the Huang-Huai spring,Huang-Huai summer and Southern spring sub-ecoregions. The Gmtfl1-ab allele was distributed in all six soybean sub-ecoregions. The Gmtfl1-bb allele was distributed mainly in the Huang-Huai spring and summer and Southern spring and summer sub-ecoregions,but the Gmtfl1-tb allele was detected only in the Huang-Huai summer sub-ecoregion. The distributions of Gm Tfl1 and Gmtfl1 have shown no large changes in nearly 60 years of breeding, but the frequency of the recessive genotype Gmtfl1 has shown a rising trend in the last 20 years. This study provides a theoretical foundation for breeding new soybean varieties for different ecoregions.

Utility of MtCOI polymerase chain reaction-restriction fragment length polymorphism in differentiating between Q and B whitefly Bemisia tabaci biotypes

The invasive, insecticide-resistant, Q whitefly biotype, has gradually spread to other countries including the US via human-mediated movement of plant materials. We assessed the utility of the VspI-based mtCOI （mitochondrion cytochrome oxidase I） polymerase chain reaction-restriction fragment length polymorphism （PCR-RFLP） technique as a rapid, cost-effective, and reliable alternative for differentiating the Q from the dominant B biotype in Arizona. Using the standard mtCOI gene sequencing and mtCOI PCR-RFLP techniques, we biotyped eight whitefly strains of five individuals each collected from poinsettia and cotton at different locations in Arizona. Complete concordance was observed between the two methods, with three strains being identified as the Q biotype and five samples as the B biotype. We also scanned the mtCOI gene sequences for VspI polymorphisms in the B and Q biotype whiteflies currently available in the GenBank database. This global screening revealed the existence of three and four VspI polymorphic types for the Q and B biotypes, respectively. Nevertheless, all three VspI polymorphic Q biotype whiteflies shared a common and unique VspI site that can be used to differentiate Q biotype from the four VspI polymorphic B biotype whiteflies identified. These results demonstrate that the VspI-based mtCOI gene PCR-RFLP provides a reliable diagnostic tool for differentiating the Q and B biotype whiteflies in the US and elsewhere.

Low-dose morphine elicits ventilatory excitant and depressant responses in conscious rats: Role of peripheral <i>µ</i>-opioid receptors

The systemic administration of morphine affects ventilation via a mixture of central and peripheral actions. The aims of this study were to characterize the ventilatory responses elicited by a low dose of morphine in conscious rats;to determine whether tolerance develops to these responses;and to determine the potential roles of peripheral μ-opioid receptors (μ-ORs) in these responses. Ventilatory parameters were monitored via unrestrained whole-body plethysmography. Conscious male Sprague-Dawley rats received an intravenous injection of vehicle or the peripherally-restricted μ-OR antagonist, naloxone methiodide (NLXmi), and then three successive injections of morphine (1 mg/kg) given 30 min apart. The first injection of morphine in vehicle-treated rats elicited an array of ventilatory excitant (i.e., increases in frequency of breathing, minute volume, respiratory drive, peak inspiratory and expiratory flows, accompanied by decreases in inspiratory time and end inspiratory pause) and inhibitory (i.e., a decrease in tidal volume and an increase in expiratory time) responses. Subsequent injections of morphine elicited progressively and substantially smaller responses. The pattern of ventilatory responses elicited by the first injection of morphine was substantially affected by pretreatment with NLXmi whereas NLXmi minimally affected the development of tolerance to these responses. Low-dose morphine elicits an array of ventilatory excitant and depressant effects in conscious rats that are subject to the development of tolerance. Many of these initial actions of morphine appear to involve activation of peripheral μ-ORs whereas the development of tolerance to these responses does not.
