LHP1 Interacts with ATRX through Plant-Specific Domains at Specific Loci Targeted by PRC2

Heterochromatin Protein 1 （HP1） is a major regulator of chromatin structure and function. In animals, the network of proteins interacting with HP1 is mainly associated with constitutive heterochromatin marked by H3K9me3. HP1 physically interacts with the putative ortholog of the SNF2 chromatin remodeler ATRX, which controls deposition of histone variant H3.3 in mammals. In this study, we show that the Arabidopsis thaliana ortholog of ATRX participates in H3.3 deposition and possesses specific conserved domains in plants. We found that plant Like HP1 （LHP1） protein interacts with ATRX through domains that evolved specifically in land plant ancestors. Loss of ATRX function in Arabidopsis affects the expression of a limited subset of genes controlled by PRC2 （POLYCOMB RePReSSIVe COMPLeX 2）, including the flowering time regulator FLC. The function of ATRX in regulation of flowering time requires novel LHPl-interacting domain and ATPase activity of the ATRX SNF2 helicase domain. Taken together, these results suggest that distinct evolutionary pathways led to the interaction between ATRX and HP1 in mammals and its counterpart LHP1 in plants, resulting in distinct modes of transcriptional regulation.

The joint european Space Agency and Chinese Academy of Sciences Solar wind Magnetosphere Ionosphere Link explorer(SMILe)mission will explore global dynamics of the magnetosphere under varying solar wind and interplanetary magnetic field conditions,and simultaneously monitor the auroral response of the Northern Hemisphere *** these large-scale responses with medium and fine-scale measurements at a variety of cadences by additional ground-based and space-based instruments will enable a much greater scientific impact beyond the original goals of the SMILe ***,we describe current community efforts to prepare for SMILe,and the benefits and context various experiments that have explicitly expressed support for SMILe can offer.A dedicated group of international scientists representing many different experiment types and geographical locations,the ground-based and Additional Science Working group,is facilitating these *** include constructing an online SMILe Data Fusion Facility,the discussion of particular or special modes for experiments such as coherent and incoherent scatter radar,and the consideration of particular observing strategies and spacecraft *** anticipate growing interest and community engagement with the SMILe mission,and we welcome novel ideas and insights from the solar-terrestrial community.

Cyber operations are relatively a new phenomenon of the last two *** that period,they have increased in number,complexity,and agility,while their design and development have been processes well kept under *** a consequence,limited data(sets)regarding these incidents are *** various academic and practitioner public communities addressed some of the key points and dilemmas that surround cyber operations(such as attack,target identification and selection,and collateral damage),still methodologies and models are needed in order to plan,execute,and assess them in a responsibly and legally compliant *** on these facts,it is the aim of this article to propose a model that i))estimates and classifies the effects of cyber operations,and ii)assesses proportionality in order to support targeting decisions in cyber *** order to do that,a multi-layered fuzzy model was designed and implemented by analysing real and virtual realistic cyber operations combined with interviews and focus groups with technical e military *** proposed model was evaluated on two cyber operations use cases in a focus group with four technical e military *** the design and the results of the evaluation are revealed in this article.

BACKgROUND Splanchnic vein thrombosis(SVT)is a major complication of moderate and severe acute *** is no consensus on whether therapeutic anticoagulation should be started in patients with acute pancreatitis and *** To gain insight into current opinions and clinical decision making of pancreatologists regarding SVT in acute *** A total of 139 pancreatologists of the Dutch Pancreatitis Study group and Dutch Pancreatic Cancer group were approached to complete an online survey and case vignette *** threshold to assume group agreement was set at 75%.ReSULTS The response rate was 67%(n=93).Seventy-one pancreatologists(77%)regularly prescribed therapeutic anticoagulation in case of SVT,and 12 pancreatologists(13%)for narrowing of splanchnic vein *** most common reason to treat SVT was to avoid complications(87%).Acute thrombosis was the most important factor to prescribe therapeutic anticoagulation(90%).Portal vein thrombosis was chosen as the most preferred location to initiate therapeutic anticoagulation(76%)and splenic vein thrombosis as the least preferred location(86%).The preferred initial agent was low molecular weight heparin(LMWH;87%).In the case vignettes,therapeutic anticoagulation was prescribed for acute portal vein thrombosis,with or without suspected infected necrosis(82%and 90%),and thrombus progression(88%).Agreement was lacking regarding the selection and duration of long-term anticoagulation,the indication for thrombophilia testing and upper endoscopy,and about whether risk of bleeding is a major barrier for therapeutic *** In this national survey,the pancreatologists seemed to agree on the use of therapeutic anticoagulation,using LMWH in the acute phase,for acute portal thrombosis and in the case of thrombus progression,irrespective of the presence of infected necrosis.

Nature-based coastal protection is increasingly recognised as a potentially sustainable and cost-effective solution to reduce coastal flood *** uses coastal ecosystems such as mangrove forests to create resilient designs for coastal flood ***,to use mangroves effectively as a nature-based measure for flood risk reduction,we must understand the biophysical processes that govern risk reduction capacity through mangrove ecosystem size and *** this perspective,we evaluate the current state of knowledge on local physical drivers and ecological processes that determine mangrove functioning as part of a nature-based flood *** show that the forest properties that comprise coastal flood protection are well-known,but models cannot yet pinpoint how spatial heterogeneity of the forest structure affects the capacity for wave or surge ***,there is relatively good understanding of the ecological processes that drive forest structure and size,but there is a lack of knowledge on how daily bed-level dynamics link to long-term biogeomorphic forest dynamics,and on the role of combined stressors influencing forest *** simulation models of forest structure under changing physical(*** to sea-level change)and ecological drivers with hydrodynamic attenuation models will allow for better projections of long-term natural coastal protection.

Prevalence of-and risk factors for work disability in Dutch patients with inflammatory bowel disease

AIM To determine the prevalence of work disability in inflammatory bowel disease(IBD), and to assess risk factors associated with work *** For this retrospective cohort study, we retrieved clinical data from the Dutch IBD Biobank on july 2014, containing electronic patient records of 3388 IBD patients treated in the eight University Medical Centers in the Netherlands. Prevalence of work disability was assessed in 2794 IBD patients and compared with the general Dutch population. Multivariate analyses were performed for work disability(sick leave, partial and full disability) and long-term full work disability(> 80% work disability for > 2 years).ReSULTS Prevalence of work disability was higher in Crohn's disease(CD)(29%) and ulcerative colitis(UC)(19%) patients compared to the general Dutch population(7%). In all IBD patients, female sex, a lower education level, and extra-intestinal manifestations, were associated with work disability. In CD patients, an age > 40 years at diagnosis, disease duration > 15 years,smoking, surgical interventions, and anti-TNFα use were associated with work disability. In UC patients, an age > 55 years, and immunomodulator use were associated with work disability. In CD patients, a lower education level(OR = 1.62, 95%CI: 1.02-2.58), and in UC patients, disease complications(OR = 3.39, 95%CI: 1.09-10.58) were associated with long-term full work *** The prevalence of work disability in IBD patients is higher than in the general Dutch population. early assessment of risk factors for work disability is necessary, as work disability is substantial among IBD patients.

This is shown via a transformation of our inventory system into a network of erlang loss queues with so-called hierarchical jurnp-over *** product-forrn solution enables fast procedures for the optimization of basestock levels,and it implies that the steady-state distribution and several relevant performance measures only depend on the distributions for the repair and replenishment leadtirnes via their means（i.e.,they are insensitive for the underlying distributions）.

The aim of this study was to characterize tinctures and microcapsules loaded with an ethanol extract of red propolis through chemical, physicochemical and microbiological assays in order to establish quality control tools for nutraceutical preparations of red propolis. The markers(isoflavonoids, chalcones, pterocarpans,flavones, phenolic acids, terpenes and guttiferones) present in the tinctures A and B were identified and confirmed using LC/eSI/FTMS/Orbitrap. Four compositions(A, B, C and D) were prepared to contain B tincture of the red propolis with some pharmaceutical excipients and submitted to two drying processes, i. ***-drying and freeze-drying to obtain microcapsules loaded with the red propolis extract. The tinctures and microcapsules of the red propolis were submitted to the total flavonoid content and antioxidant activity *** antibacterial activity and minimum inhibitory concentration(MIC) were tested using Staphylococcus aureus ATCC 25293 and Pseudomonas aeruginosa ATCC 27853 strains. The tinctures and microcapsules presented high flavonoid quantities from 20.50 to 40.79 mg/100 mg of the microcapsules. The antioxidant activity and IC50 were determined for the tinctures A and B(IC50: 6.95 μg/m L and 7.48 μg/m L), the spray-dried microcapsules(IC50: 8.89–15.63 μg/m L) and the freeze-dried microcapsules(IC50: 11.83–23.36 μg/m L). The tinctures and microcapsules were proved to be bioactive against gram-positive and gram-negative bacteria with inhibition halos superior to 10 mm at concentration of 200 μg/m L and MIC values of 135.87–271.74 μg/m L using gram-positive strain and 271.74–543.48 μg/m L using gram-negative strain. The tinctures and microcapsules of the red propolis have a potential application for nutraceutical products.
