Multitarget stool DNA(mt-sDNA) testing was approved for average risk colorectal cancer(CRC) screening by the United states Food and Drug Administration and thereafter reimbursed for use by the Medicare program(2014).The United states Preventive services Task Force(UsPsTF) October 2015 draft recommendation for CRC screening included mt-s DNA as an "alternative" screening test that "may be useful in select clinical circumstances",despite its very high sensitivity for early stage *** evidence supporting mt-s DNA for routine screening use is *** clinical efficacy of mt-s DNA as measured by sensitivity,specificity,life-years gained(LYG),and CRC deaths averted is similar to or exceeds that of the other more specifically recommended screening options included in the draft document,especially those requiring annual testing *** a population with primarily irregular screening participation,tests with the highest point sensitivity and reasonable specificity are more likely to favorably impact CRC related morbidity and mortality than those depending on annual *** paper reviews the evidence supporting mt-s DNA for routine screening and demonstrates,using UsPsTF's modeling data,that mt-s DNA at three-year intervals provides significant clinical net benefits and fewer complications per LYG than annual fecal immunochemical testing,high sensitivity guaiac based fecal occult blood testing and 10-year colonoscopy screening.

Roost selection of the endangered spotted Greenshank(Tringa guttifer) in critical habitat in the Inner Gulf of Thailand

Background: Roost-site quality can significantly affect the individual fitness of shorebirds, but roost sites remain poorly described for many threatened species on the East Asian–Australasian Flyway. We studied roost-site selection of the globally endangered spotted Greenshank (Tringa guttifer) in the Inner Gulf of Thailand, an area which supports approximately 24% of their global wintering population, during two non-breeding seasons (October 2014–May 2015 and December 2015–February 2016). Methods: We measured nine variables associated with roost site characteristics including water depth, indicators of disturbance/predation risk, and associations with other shorebird species. We predicted that roost ponds with shallow water in proximity to foraging sites would receive higher usage than those further away. Results: A total of 94 sites were measured of which 46 were used for roosts with 23 used repeatedly. All used sites were human-modified ponds, of which 44 were used for salt farming and two used for aquaculture. Roosts were on average 1.10 ± 0.78 (sE) km from foraging sites and 5.8 ± 2.4 cm deep. The most supported model indicated that roost sites were negatively associated with distance to foraging sites and positively associated with the presence of Grey Plover (Pluvialis squatarola) and water depth. Conclusions: Traditional saltpans and other artificial wetlands near (s serve as the primary high-tide roost habitat in the Inner Gulf of Thailand for this spotted Greenshank population and perhaps seven other globally threatened or near-threatened species. Critically, all observed roost sites are on private land with no formal protection and thus will require creative public–private partnerships to manage sustainably.

Geomorphic Habitat Type, Drift Cell, Forage Fish and juvenile salmon： Are They Linked？

The role of geomorphic habitat type, drift cell scale, and geographic scale in defining fish use of nearshore habitats is poorly known, particularly for Pacific salmon and their prey. In this study, key areas of nearshore habitat in central and western strait of juan de Fuca were categorized by geomorphic habitat type and assessed for fish use within a degraded (Elwha) and intact comparative drift cells over a one year period. juvenile Chinook and coho salmon were also sampled for genetic analysis to define regional dispersal patterns. Key findings are: (1) Ecological function of the area"s nearshore is complex, with very strong seasonal variation in fish use both within and across GMHT (geomorphic habitat type); (2) GMHT link to nearshore function for fish use differs depending on the fish species and time of year. surf smelt and sand lance were the most abundant. And they were seasonally used embayed, spit, and bluff shorelines more than lower rivers. juvenile Chinook, coho, and chum salmon occurred in much lower density than forage fish species, and used lower rivers more than other GMHTs; (3) When GMHTs were combined and analyzed at the drift cell scale, the degraded drift cell had different ecological patterns than the intact drift cell; (4) Cross regional juvenile fish use of nearshore is an important component of habitat use: juvenile Chinook and coho from as far away as the Columbia River Oregon and Klamath River California utilize central strait of juan de Fuca shorelines. Forage fish species may do so as well. Drift cell and cross regional scales are therefore most important for accurately defining nearshore ecological function, management, and restoration actions.

Fast-neutron （FN） irradiation has been used to create mutagen- ized collections of many plant species （Bolon et al., 2014）, FN- induced mutagenesis has clear advantages： it is an efficient means of saturating the genome, and it does not involve time- consuming plant transformation or tissue culture. In rice, most mutant collections, although highly valuable, were generated us- ing either T-DNA insertion or transposon tagging approaches that often induce mutations unlinked to the insertion and complicating analysis （Wang et al., 2013）. Another disadvantage of some of these collections is that they were created in rice varieties that are photoperiod-sensitive and often have long generation times.

The crystalline terrane of the Tongbai-Dabie region,centralChina,comprising the Earth’s largest ultrahigh-pressure(UHP) exposure was formed during Triassic collision between the sino-Korean andYangtze *** apatite fission-track(AFT) data presented here from the UHP terrane,extends over a significantly greater area than reported in previous studies,and includes the(eastern) Dabie,the Hong’an(northwestern Dabie) and Tongbai *** new data yield ages ranging from44±3 to 142±36Ma and mean track lengths between□10 and 14.4μ.*** history models based on the AFT data taken together with published 40Ar/39Ar,K -Ar,apatite and zircon(U-Th)/He and U-Pb data,exhibit a three-stage cooling pattern that is similar across the study region,commencing with an Early Cretaceous rapid cooling event, followed by a period of relative thermal stability during which rocks remained at temperatures within the AFT partial annealing zone(□60-110℃) and ending with a possible renewed phase of accelerated cooling during Pliocene to Recent *** first cooling phase followed large-scale transtensional deformation between□140 and 110 Ma and is related to Early Cretaceous eastward tectonic escape and Pacific back arc *** this phase and the subsequent slow cooling phase,a transition period from□120 to 80Ma(to□70 to 45Ma along the Tan-Lu fault) was characterised by a relatively low cooling rate(□3-5℃/Ma).This transition is likely related to a tectonic response associated with the mid-Cretaceous subduction of the IzanagiPacific plate aswell as lithospheric extension and thinning in eastern *** present regional AFTage pattern is therefore basically controlled by the Early Cretaceous rapid cooling event,but finally shaped through active Cenozoic *** the transition phase the subsequent slow cooling phase pattern implies a net reduction in horizontal compressional stress corresponding to increased extensionrates along the continental margin due to the de

Microgeographic Variation and Inter-Riffle Migration of <i>Rhinichthys atratulus</i>(Pisces: Cyprinidae) in a small Connecticut stream, United states

We examined the genetic diversity on a microgeographic scale of Rhinichthys atratulus (Eastern Blacknose Dace) in Allyn Brook, a small tributary in the upper Coginchaug River drainage in Connecticut. By looking at gene flow on a microgeographic scale among populations that had no physical barriers to migration, we tested the null hypothesis that the populations should be homogeneous. We resolved seven polymorphic microsatellite loci and one mitochondrial gene, nd2, in three adjacent populations (se populations to the two closest populations (>5 km) in the Coginchaug River. A dam from the 1920’s in lower Allyn Brook has isolated Allyn-Brook populations from Coginchaug-River populations. Allyn Brook was selected because there are only three riffle habitats in the brook and, therefore, there can be no immigration from upstream populations. Each population has private (i.e., unique) alleles and haplotypes, and there are significant genetic differences between all sites. The Allyn Brook populations are almost as different from one another as they are from the distant populations in the Coginchaug River from which they have been isolated for more than 80 years. These results point to in situ evolution and little migration or gene flow among populations on a microgeographic scale. This raises interesting questions for conservation of genetic diversity of stream fishes.

背景:在根治幽门螺杆菌(Hp)的标准三联治疗中,甲硝唑是其中的一种一线药物。因此,甲硝唑耐药是一个重大健康问题。RdxA失活被认为是甲硝唑耐药的主要机制。然而,表达功能性RdxA蛋白的Hp菌珠更容易对甲硝唑耐药,提示还存在其他一些导致甲硝唑耐药的机制。方法:我们对121份Hp菌珠进行了全基因组测序,其中73份是甲哨唑耐药菌珠。对包含全长RdxA基因的Hp临床菌珠的核心蛋白簇进行序列比对分析。对耐药菌珠与敏感菌珠的差异位点进行分析,以鉴定出差异基因及其导致甲硝唑耐药的氨基酸改变。结果:在73份甲硝唑耐药菌珠中,25份(34%)是由于RdxA无义突变所导致。对来自剩余48份甲硝唑耐药菌珠和48份甲硝唑敏感菌株的核心蛋白簇进行分析发现,有4个差异位点与甲硝唑耐药显著相关。这4个位点包括RdxA的R16H/C位点、内膜蛋白RclC(HP0565)的D85N位点、biotin carboxylase蛋白(HP0370)的V265I位点和putative threonylcarbamoyl-AMP合酶(HP0918)的A51V/T位点。结论:我们发现了Hp对甲硝唑耐药的一些新的潜在机制,值得进一步研究。

Responses of Macroinvertebrate Community Metrics to a Wastewater Discharge in the Upper Blue River of Kansas and Missouri, UsA

The Blue River Main wastewater treatment facility (WWTF) discharges into the upper Blue River (725 km2), and is recently upgraded to implement biological nutrient removal. We measured biotic condition upstream and downstream of the discharge utilizing the macroinvertebrate protocol developed for Kansas streams. We examined responses of 34 metrics to determine the best indicators for discriminating site differences and for predicting biological condition. significant differences between sites upstream and downstream of the discharge were identified for 15 metrics in April and 12 metrics in August. Upstream biotic condition scores were significantly greater than scores at both downstream sites in April (p = 0.02), and in August the most downstream site was classified as non-biologically supporting. Thirteen EPT taxa (Ephemeroptera, Plecoptera, Trichoptera) considered intolerant of degraded stream quality were absent at one or both downstream sites. Increases in tolerance metrics and filtering macroinvertebrates, and a decline in ratio of scrapers to filterers all indicated effects of increased nutrient enrichment. stepwise regressions identified several significant models containing a suite of metrics with low redundancy (R2 = 0.90 - 0.99). Based on the rapid decline in biological condition downstream of the discharge, the level of nutrient removal resulting from the facility upgrade (10% - 20%) was not enough to mitigate negative effects on macroinvertebrate communities.
