Molecular basis for identification of species/isolates of gastrointestinal nematode parasites

Gastrointestinal（GI） parasitism is the most serious constraint throughout the world in small ruminants which causes significant production loss in animals.GI parasites are major contributor to reduce productivity in terms of meat,milk and wool in animals.Control of GI parasite is done primarily by anthelmintic treatment where choice and schedule of treatment is done after identification and quantitation of individual parasite.Identification of GI parasites is done through microscopic method by identifying specific morphological characteristics of egg and larva（Lsub>3sub>）.since most of parasite eggs are having similar morphological characteristics, identification up to species level through microscopy is not possible in most of cases.To address this issue,molecular techniques are the viable alternative for identification of species as well as molecular level differences within a species（isolates） of parasites.Different DNA based molecular techniques viz.PCR,AFLP,RAPD,RFLP,PCR-ssCP,real time PCR,DNA microarray etc.have been used for identification and to assess the genetic diversity among parasite population.For identification of species,the characteristic sequence of genomic DNA of different species should differ to allow the delineation of species,but at the same time,no/minor variation within the species should exist.In contrast,for purpose of identifying population variants（strains/isolates）, a considerable degree of variation in the sequence should exist within a species.Various target regions,including nuclear ribosomal DNA（rDNA）,mitochondrial DNA（mtDNA） or repetitive DNA elements（microsatellite loci）,which show considerable variation in the number of repeats within individuals have been employed to achieve the identification of parasites species or strain.

Detection of canine echinococcosis by coproantigen ELIsA

Objective:To study the canine echinococcosis by coproantigen ELIsA method.Methods:During the present investigation experimental infection was established using evaginated worms of Echinococcus granulosus(E.granulosus).To check cross reactivity two pups were infected with Taenia hydatigena(T.hydatigena).In order to detect the presence of antigen,hyperimmune sera were raised against excretory-secretory products of adult worms E.chinococcus granulosus. Faecal sample collected either from experimentally infected pups or from other sources were heated at 70℃to detect heat stable soluble antigen.Results:Pups harbouring less than 104 worms showed negative results.samples collected from 14 days onwards from experimentally infected animals harbouring more than 104 worms showed positive value.The maximum positive samples were detected in samples collected from in and around slaughter house and the least number of samples were detected positive maintained by dog squad.Conclusions:The affinity purified IgG exhibited promising results for detection of canine echinococcosis by indirect ELIsA.

Prevalence of Deg Nala disease in eastern India and its reproduction in buffaloes by feeding Fusarium oxysporum infested rice straw

Objective:To undertake a study on prevalence of Deg Nala disease in eastern states of India and to reproduce the disease in buffaloes by the Fusarium spp.,isolated from the affected region. Methods:During this investigation,a survey was conducted covering four states of eastern region to identify the Deg Nala cases as well as to isolate and characterize the causative agent(s). An experimental study was carried out to reproduce the disease in healthy male buffaloes(2-3 years age) by randomly dividing them into five groups(four in each group).Each individual group was fed with rice straw artificially infested with either of the two representative isolates of Fusarium oxysporum(F.oxysporum)(F01,F02) or representative reference strains of Fusarium equiseti(F.equiseti)(ITCCF-2470) and Fusarium moniliforme(F.moniliforme)(ITCCF-4821) for 30 days,whereas the control group was fed with normal rice straw only.Results:A total of 658 Deg Nala cases were recorded and 12 Fusarium isolates were identified from the mouldy rice straw collected from these affected areas.The characterization of the isolates revealed three species viz.,F.oxysporum,F.equiseti and F.moniliforme,among which F.oxysporum was predominant.The disease was artificially reproduced in three buffaloes in F01 group and one in F02 group within 20-23 days by feeding F.oxysporum infested rice straw which resembled the clinical symptoms and gross lesions of natural Deg Nala cases.Conclusions:The field investigation and laboratory studies,including experimental production of Deg Nala disease suggest the possible involvement of mycotoxins.However,further investigations needs to be done to understand nature of the toxic factors involved in production of the Deg Nala disease.

Additively manufactured Ti–Ta–Cu alloys for the next-generation load-bearing implants

Bacterial colonization of orthopedic implants is one of the leading causes of failure and clinical complexities for load-bearing metallic implants. Topical or systemic administration of antibiotics may not offer the most efficient defense against colonization, especially in the case of secondary infection, leading to surgical removal of implants and in some cases even limbs. In this study, laser powder bed fusion was implemented to fabricate Ti3Al2V alloy by a 1:1 weight mixture of CpTi and Ti6Al4V powders. Ti-Tantalum(Ta)–Copper(Cu) alloys were further analyzed by the addition of Ta and Cu into the Ti3Al2V custom alloy. The biological,mechanical, and tribo-biocorrosion properties of Ti3Al2V alloy were evaluated. A 10 wt.% Ta(10Ta) and 3 wt.% Cu(3Cu) were added to the Ti3Al2V alloy to enhance biocompatibility and impart inherent bacterial resistance. Additively manufactured implants were investigated for resistance against Pseudomonas aeruginosa and staphylococcus aureus strains of bacteria for up to 48 h. A 3 wt.% Cu addition to Ti3Al2V displayed improved antibacterial efficacy, i.e.78%–86% with respect to CpTi. Mechanical properties for Ti3Al2V–10Ta–3Cu alloy were evaluated, demonstrating excellent fatigue resistance, exceptional shear strength, and improved tribological and tribo-biocorrosion characteristics when compared to Ti6Al4V. In vivo studies using a rat distal femur model revealed improved early-stage osseointegration for alloys with10 wt.% Ta addition compared to CpTi and Ti6Al4V. The 3 wt.% Cu-added compositions displayed biocompatibility and no adverse infammatory response in vivo. Our results establish the Ti3Al2V–10Ta–3Cu alloy’s synergistic effect on improving both in vivo biocompatibility and microbial resistance for the next generation of load-bearing metallic implants.

The present study deals with the development of a low carbon high strength steel by thermomechanical controlled processing on a pilot scale.The continuous cooling transformation has yielded a flat top "C" curve with the phase field occupied by a mixture of bainite and martensite.The microstructure of water quenched steel essentially consists of highly dislocated lath martensite along with fine (Ti,Nb)CN precipitates and twins.High strength steel with 1217-1298 MPa yield strength and 1372-1513 MPa ultimate tensile strength along with 16-12% total elongation has been obtained in the range of 850 to 750℃ finish rolling temperature.The impact toughness value in the range of 45-72J was also achieved in the present steel.

An Autopsy and Histopathology-based study to Ascertain the Cause of Death of Brought Dead Cases

Background and Objectives:The cases of apparent natural deaths are encountered in the hospital casualties who are noted dead at the time of the first medical attention and as such are labeled as“Brought Dead.”These include the cases of sudden death,unexplained death,and unattended death.The cause of such deaths is reported in the literature to emanate principally from the cardiovascular system,though there are some documented geographical,racial,and other variations.In the present study,we endeavored to detect the cause of such natural deaths in terms of the disease and the organ system involved,in the deceased individuals of the contemporary Bengali population,with cases due to obvious unnatural causes being excluded.Materials and Methods:An observational,cross-sectional study with 105(n=105,male:89,female:16)study subjects of age group of 20-80 years,selected by systemic random sampling,from the brought dead subjects coming to the mortuary of the NRs Medical College and Hospital for medicolegal autopsy from April 2011 to March 2012,excluding cases having a history of the obvious unnatural event.Organ system-wise gross examination was done,followed by tissue sampling for histopathological examination.The case-wise findings were tabulated and analyzed for descriptive statistics using sPss software.Results:In the present study,89 cases were male(84.8%)and 16 cases were female(15.2%),which reflects a male:female ratio of 5.56:1.The dead bodies are mostly recovered from the footpaths(47 cases,44%).sixty-four(61%)cases are of known identity.In 54 cases out of 105(51.4%),morbid pathology is detected in the respiratory system(Rs),lobar pneumonia,and cavitary pulmonary tuberculosis being the most frequent disease entity,in both genders.Fifty-one years-sixty-five years is found to be the most commonly affected age group,in both genders.Interpretation and Conclusions:The present study shows that the most common organ system involved in brought dead cases is the Rs,which is supported by a few Indian studies.

Groundwater assists farmers to irrigate crops for fulfilling the crop-water requirement.Indian agriculture system is characterized by three cropping seasons known as Kharif(monsoon),Rabi(post-monsoon)and summer(pre-monsoon).In tropical countries like India,monitoring cropping practices using optical remote sensing during Kharif and Rabi seasons is constraint due to the cloud cover,which can be well addressed by microwave remote sensing.In the proposed research,the strength of C-band polarimetric synthetic Aperture Radar(sAR)time series images were evaluated to classify groundwater irrigated croplands for the Kharif and Rabi cropping seasons of the year 2013.The present study was performed in the Berambadi experimental watershed of Kabini river basin,southern peninsular India.A total of fifteen polarimetric variables were estimated includes four backscattering coefficients(HH,HV,VH,VV)and eleven polarimetric indices for all Radarsat-2 sAR images.The cumulative temporal sum(seasonal and dual-season)of these parameters was supervised classified using support Vector Machine(sVM)classifier with intensive ground observation samples.Classification results using the best equation(highest accuracy and kappa)shows that the Kharif,Rabi and irrigated double croplands are respectively 9.58 km2(20.6%),16.14 km2(34.7%)and 6.22 km2(13.4%)with a kappa coefficient respectively 0.84,0.74 and 0.94.

Additive manufacturing provides achievability for the fabrication of bimetallic and multi-material structures;however,the material compatibility and bondability directly affect the parts’formability and final quality.It is essential to understand the underlying printability of different material combinations based on an adapted process.Here,the printability disparities of two common and attractive material combinations(nickel-and iron-based alloys)are evaluated at the macro and micro levels via laser directed energy deposition(DED).The deposition processes were captured using in situ high-speed imaging,and the dissimilarities in melt pool features and track morphology were quantitatively investigated within specific process windows.Moreover,the microstructure diversity of the tracks and blocks processed with varied material pairs was comparatively elaborated and,complemented with the informative multi-physics modeling,the presented non-uniformity in mechanical properties(microhardness)among the heterogeneous material pairs was rationalized.The differences in melt flow induced by the unlike thermophysical properties of the material pairs and the resulting element intermixing and localized re-alloying during solidification dominate the presented dissimilarity in printability among the material combinations.This work provides an in-depth understanding of the phenomenological differences in the deposition of dissimilar materials and aims to guide more reliable DED forming of bimetallic parts.

Highly specific interactions between biomolecules,such as antigen-antibody,protein-ligand,or nucleic acid base pair complementary are on the basis of the organization of complex organisms.The same principles may be tentatively used in molecular medicine for diagnosis and therapeutics.A molecule can be designed to selectively bind a protease and thereby inhibit the production of a peptide that forms toxic aggregates in the brain or an antibody may be produced to bind specifically to that peptide for detection or clearance purposes.Unfortunately,interference in biological systems is not that simple.For a start there is the inhibition of the physiological role of the protease;moreover,several cleavage fragments may be produced,which may continue to diverge due to putative post-translational modification and self-assembly processes,hiding the toxic target in a“soup”of peptide species varying in size,structure and chemical composition.A perspective of the current status and challenges in targeting peptide species for diagnosis and treatment in the context of Alzheimer’s disease is given.

Thermoelectric performance largely depends on the reduction of lattice thermal conductivity(kL).The study of the thermal conductivity(k)of a sb_(2)Te_(3)/graphite nanocomposite system demonstrates-40%reduction in kL with graphite incorporation.A plausible explanation of intrinsic low kL observed in sb_(2)Te_(3) based system is presented by modeling experimental specific heat(Cp)data.Raman spectroscopy measurement combined to X-Ray diffraction data confirms the presence of graphite as separate phase in the composite sample.It is found that phonon scattering dominates heat transport mechanism in the nanostructured sb_(2)Te_(3)/graphite composite.Large reduction in kL is accomplished by intensifying scattering rate of phonons via various sources.Graphite introduces effective scattering sources,i.e.,defects of different dimensionalities in synthesized nanocomposite sample.Furthermore,graphite mediates phonon-phonon coupling and enhances lattice anharmonicity,which causes an intrinsic scattering of phonons with all frequencies in the sb_(2)Te_(3)/graphite nanocomposite sample.Dislocation density and phonon anharmonicity of the synthesized samples are estimated from in depth analysis of temperature dependent synchrotron powder diffraction and Raman spectroscopic data.kL value as low as 0.8 W m1 K1 at 300 K,achieved with graphite dispersion in sb_(2)Te_(3) based composite system makes the present comprehensive study an interesting concept to be developed in thermoelectric materials.
