MiR-450b-5p enhances the radiosensitivity of HR^(+) and HER2^(−) breast cancer by targeting CDK6

Background:The sensitivity of breast cancer cells to radiation is a key cause of locoregional recurrence after postoperative *** studies have reported that microRNAs(miRNAs)are involved in the radiosensitivity of human breast cancer *** miRNA microarray study showed that miR-450b-5p was overexpressed 13.3-fold in patients with estrogen receptor–positive(ER^(+))and human epidermal growth factor receptor 2–negative(HER2−)breast cancer and no local relapse compared with local relapse ***,its underlying mechanism of action remains ***:The predicted target mRNAs of miR-450b-5p were screened using the TargetScan,miRDB,and miRWalk *** blotting,quantitative polymerase chain reaction,and dual-luciferase reporter assays explored the association between cyclindependent kinase 6(CDK6)and *** cell counting kit-8 assay and flow cytometry detected the proliferation of transfected MCF7 *** formation and xenograft tumors detected the radiosensitivity of the transfected MCF7 ***:Bioinformatics analysis,Western blotting,quantitative polymerase chain reaction,and dual-luciferase reporter assays demonstrated that CDK6 was the target gene of ***,in vitro and in vivo experiments showed that miR-450b-5p inhibited MCF7 cell proliferation and cell cycle progression,increased the sensitizer enhancement ratio,and decreased the volume of xenograft tumors after irradiation by regulating ***:This study demonstrates that miR-450b-5p enhances the radiosensitivity of hormone receptor–positive(HR^(+))and HER2−breast cancer cells and elucidates its ***-450b-5p may be considered a therapeutic target in HR^(+)and HER2−breast cancer treated with radiotherapy.

In situ enhanced bioremediation of dichlorvos by a phyllosphere Flavobacterium strain

A bacterium capable of degrading dichlorvos was isolated from the rape phyllosphere and designated YD4. The strain was identified as Flavobacterium sp., based on its phenotypic features and 16S rRNA gene sequence. Strain YD4 was able to utilize dichlorvos as the sole source of phosphorus. In situ enhanced bioremedia- tion of dichlorvos by YD4 was hereafter studied. Chlorpyrifos and phoxim could also be degraded by this strain as the sole phosphorus source. A higher degradation rate of dichlorvos was observed after spraying YD4 onto the surface of rape leaves when compared to the sterilized- YD4 and water-treated samples. The results indicated that pesticide-degrading epiphytic bacterium could become a new way for in situ phyllosphere bioremediation where the hostile niche is unsuitable for other pesticide-degrading bacteria isolated from soil and water.

Composition of algal pigments in surface freshen layer after ice melt in the central Arctic

Seasonal meltwater input creates a thin freshen layer in surface seawater under ice, which largely shifts the algae assemblages. Our recent observation of photosynthetic pigments in the high Arctic showed that ice bottom and 5 m of seawater under ice contained relatively high concentration of fucoxanthin, while chlorophyll b and lutein were the major diagnostic pigments in ice-water interface and 0 m of seawater under ice. Additionally, a notable change of dominant phytoplankton occurred in the top 5 m of seawater under ice, from chlorophytes-dominated at surface to diatoms-dominated at 5 m depth, which might attribute to the sharp salinity gradient （salinity from 12.5 to 28.1） in the surface seawater under ice. Our results imply that phytoplankton community in surface layer under ice would become more chlorophytes in the future warming Arctic Ocean.

Experiments were conducted to evaluate the microstructure and tensile properties of a medium carbon Cr-Ni-W-Mo steel processedthermo-mechanical controlled processing(TMCP)with cooling at different conditions in water,oil,air or lime followedlow *** to normal heat-treatment processing,TMCP with water-cooling after deformation enhances the yield strength and tensile strength of the steelabout 323 MPa and about 251 MPa,respectively,due to higher dislocation strengthening and grain boundary ***,it increases the elongation by ;about 1.76%attributed to the increase in volume percentage of the retained austenite and the refined laths of tempered *** the cooling rate after deformation during TMCP leads to a decrease in the *** results the coupling effectsthe reduction in dislocation density and volume fraction of tempered martensite together with the coarseness in martensite ***,cooling rate decreasing has less influences on ductility becathe improved elongation the increase in the volume fractions of both retained austenite and lower bainite together with dislocation density decreasing is compensatedthe reduced elongation coarsened grains.

Japanese encephalitis following liver transplantation: A rare case report

BACKGROUND Japanese encephalitis(JE) is a serious public health concern with a high mortality rate in many Asian countries. For many years, JE virus(JEV) was considered the major cause of viral encephalitis in Asia. Although most JE cases are asymptomatic, the case fatality rate approaches 30%, and approximately 30%–50% of survivors have long-term neurological sequelae. To the best of our knowledge, JEV infection has never been reported following liver *** SUMMARY We report a case of a woman who underwent liver transplantation for autoimmune liver disease but presented with fever and neurological symptoms 13 d after transplantation. Magnetic resonance imaging revealed JEV infection,and positive immunoglobulin M antibody to JEV in blood and cerebrospinal fluid confirmed JE. The patient was treated with antiviral agents, immune regulation,and organ function support. No neurological sequelae were present after 1 year of *** Imaging and lumbar puncture examination should be performed as soon as possible in patients with fever and central nervous system symptoms after liver transplantation, and the possibility of atypical infection should be considered,which is helpful for early diagnosis and improved prognosis.

Mitochondria-localized iridium(Ⅲ) complexes with anthraquinone groups as effective photosensitizers for photodynamic therapy under hypoxia

Photodynamic therapy（PDT） is a potential way for the tumor treatment. However, it notably suffers the limitation of hypoxia in solid tumors. Thus, it is significant to develop effective photosensitizers which can exhibit excellent therapeutic performance under both normoxia and hypoxia. Herein, we reported four ionic iridium（III） complexes（Ir1–Ir4） with anthraquinone groups which can regulate their excited state energy levels effectively. Among them, the energy gap of Ir1 was between 1.63 and 2.21 eV, which can match well with that of O2, and the HOMO energy of Ir1 is less than-5.51 eV. Compared with Ir2–Ir4, the luminescent quantum efficiency of Ir1 was the highest. Particularly, Ir1 can specifically target the mitochondria of the tumor cells. Meanwhile, Ir1 showed high singlet oxygen quantum yields（ΦΔ） in both solutions and living cells with low *** results of PDT experiments revealed that Ir1, as a photosensitizer, exhibited excellent therapeutic effect not only in normoxia but also in hypoxia condition. We believe that this work is meaningful for developing excellent PDT agents based on cyclometalated Ir（III） complexes via rational ligand modification.

Overlooked contribution of the biological pump to the Pacific Arctic nitrogen deficit

The nutrient-rich Pacific Ocean seawater that flows through the Bering Strait into the Chukchi Sea is generally considered to be the most important source of nutrients to the Arctic euphotic *** inflow is characterized by nitrogen deficit and low nitrate/phosphate(N/P)ratios;this is ascribed to sedimentary denitrification on the Chukchi shelf by preoccupant ***,the Chukchi Sea also has high primary production,which raises the question of whether the biological pump may also significantly modulate nutrient properties of the ***,we show that nitrate concentrations of the Pacific inflow gradually decrease northward in association with notable biological *** phytoplankton N/P uptake ratio was 8.8±2.27,higher than the N/P ratio of Pacific inflow water(5-6).This uptake ratio,in combination with efficient vertical nitrogen export,serves to preferentially remove nitrogen(relative to phosphorus)from upper waters,thereby further intensifying the Arctic nitrogen ***,as large as about 111.7×10^(9)mol N yr^(−1)of nitrate was extra consumed,according to the real N/P uptake ratio rather than the ratio of the Pacific inflow,which may be as great as half the nitrogen loss ascribed to sedimentary *** findings suggest that besides sedimentary denitrification,biological disproportionate utilization of nutrients in the Chukchi Sea upper water is another important contributor to the nitrogen limitation and excess phosphorus in the upper Arctic *** the rapid Arctic change era,the predicted reinforced biological carbon pump could further impact the nutrient dynamics and biogeochemical process of the Arctic Ocean.

Poly-ion complex(PIC)integrating non-antibiotic theranostics holds great promise in the combat against drug-resistant *** with aggregation-induced emission(AIE)characteristic are particularly intriguing theranostic agents,but incorporating them into antibacterial PIC to enable both fluorescence and reactive oxygen species(ROS)generation turn-on is deemed a great *** we report the development of a PIC that can dually boost the fluorescence and ROS generation in the presence of pathogen *** PIC is constructed based on an anionic polydiacetylene poly(deca-4,6-diynedioic acid)(PDDA),which completely degrades in the presence of ROS.A cationic polymer quaternized poly(2-(dimethylamino)ethyl methacrylate)(PQDMA)that can disrupt bacterial membrane is co-loaded together with a highly efficient AIE photosensitizer TPCI in the *** is nonfluorescent initially in that PDDA can quench the AIE of TPCI in *** pathogenic bacteria are present,they can disturb the assembly of PIC to release TPCI,whose fluorescence turns on sensitively to indicate the existence of *** on-demand irradiation can be subsequently applied to excite TPCI,which generates ROS to degrade PDDA and deform the *** a result,TPCI and PQDMA are completely released to eliminate bacteria through a synergy of turned-on photodynamic therapy(PDT)and membrane *** highly efficient detection and inhibition against both Gramnegative and Gram-positive bacteria have validated this polydiacetylene-based PIC system as an effective non-antibiotic antibacterial theranostic platform as well as a new strategy to enable“turn-on”fluorescence sensing and imaging of AIE fluorophores.
