Fusion of a rice endogenous N-methylpurine DNA glycosylase to a plant adenine base transition editor AbE8e enables A-toK base editing in rice plants

Engineering of a new type of plant base editor for simultaneous adenine transition and transversion within the editing window will greatly expand the scope and potential of base editing in directed evolution and crop ***,we isolated a rice endogenous hypoxanthine excision protein,N-methylpurine DNA glycosylase(OsMPG),and engineered two plant A-to-K(K=G or T)base editors,rAKbE01 and rAKbE02,for simultaneous adenine transition and transversion base editing in rice by fusing OsMPG or its mutant mOsMPG to a plant adenine transition base editor,*** further coupled either OsMPG or mOsMPG with a transactivation factor VP64 to generate rAKbE03 and rAKbE04,*** these four rAKbEs,at five endogenous loci in rice protoplasts,indicated that rAKbE03 and rAKbE04 enabled higher levels of A-to-G base transitions when compared to AbE8e and ***,whereas rAKbE01 only enabled A-to-C/T editing at one endogenous locus,in comparison with rAKbE02 and rAKbE03,rAKbE04 could significantly improve the A-to-C/T base transversion efficiencies by up to 6.57-and 1.75-fold in the rice protoplasts,***,although no stable lines with A-to-C transversion were induced by rAKbE01 and rAKbE04,rAKbE04 could enable simultaneous A-to-G and A-to-T transition and transversion base editing,at all the five target loci,with the efficiencies of A-to-G transition and A-to-T transversion editing ranging from 70.97 to 92.31%and 1.67 to 4.84%in rice stable lines,***,these rAKbEs enable different portfolios of editing products and,thus,now expands the potential of base editing in diverse application scenario for crop improvement.

Sialyltransferase ST3GAL6 silencing reduces α2,3-sialylated glycans to regulate autophagy by decreasing HSPb8-bAG3 in the brain with hepatic encephalopathy

End-stage liver diseases,such as cirrhosis and liver cancer caused by hepatitis b,are often combined with hepatic encephalopathy(HE);ammonia poisoning is posited as one of its main pathogenesis *** is closely related to autophagy,but the molecular mechanism of ammonia’s regulatory effect on autophagy in HE remains *** is an essential form of *** the nervous system,abnormal sialylation affects various physiological processes,such as neural development and synapse ***3 β-galactoside α2,3-sialyltransferase 6(ST3GAL6)is one of the significant glycosyltransferases responsible for addingα2,3-linked sialic acid to substrates and generating glycan *** found that the expression of ST3GAL6 was upregulated in the brains of mice with HE and in astrocytes after ammonia induction,and the expression levels of α2,3-sialylated glycans and autophagy-related proteins microtubule-associated protein light chain 3(LC3)and beclin-1 were upregulated in ammonia-induced *** findings suggest that ST3GAL6 is related to autophagy in ***,we aimed to determine the regulatory relationship between ST3GAL6 and *** found that silencing ST3GAL6 and blocking or degrading α2,3-sialylated glycans by way of Maackia amurensis lectin-II(MAL-II)and neuraminidase can inhibit *** addition,silencing the expression of ST3GAL6 can downregulate the expression of heat shock proteinβ8(HSPb8)and bcl2-associated athanogene 3(bAG3).Notably,the overexpression of HSPb8 partially restored the reduced autophagy levels caused by silencing ST3GAL6 *** results indicate that ST3GAL6 regulates autophagy through the HSPb8-bAG3 complex.

Facet-dependent activation of oxalic acid over hematite nanocrystals under the irradiation of visible light for efficient degradation of pollutants

Naturally occurring hematite has been widely studied in the Fenton-like system for water pollutant remediation due to its abundance and ***,its inadequate catalytic activity results in difficulty in effectively degrading pollutants in the catalytic degradation system that it ***,we constructed a photochemical system composed of hematite with{001}facet of high activity facet and low-cost and non-toxic oxalic acid(OA)for the removal of various types of *** removal rate for the degradation of metronidazole,tetracycline hydrochloride,Rhodamine b,and hexavalent chromium by hematite nanoplate with the exposed{001}facet activating OA under visible light irradiation was 4.75,2.25,2.33,and 2.74 times than that by the exposed{110}facet,*** functional theory(DFT)calculation proved that the OA molecule was more easily adsorbed on the{001}facet of hematite than that on the{110}facet,which would favor the formation of the more Fe(Ⅲ)-OA complex and reactive *** addition,the reactive site of metronidazole for the attraction of radicals was identified on the basis of the DFT calculation on the molecular occupied orbitals,and the possible degradation pathway for metronidazole included carbon chain fracture,hydroxyethyl-cleavage,denitrogenation,and ***,this finding may offer a valuable direction in designing an efficient iron-based catalyst based on facet engineering for the improved activity of Fenton-like systems such as OA activation.

Orbital pacing and secular evolution of lake-level changes reconstructed by sedimentary noise modeling during the Early Jurassic icehouses-(super)greenhouses

Lake-level changes can significantly affect paleoenvironmental evolution,resource occurrence,terrestrial carbon budget,and biodiversity in continental *** is one of the most critical factors controlling lake-level *** of the Early Jurassic has been evidenced by oscillating icehouses to(super)greenhouses with interrupted intermittent extreme climatic events(hyperthermal and cooling),e.g.,the Toarcian oceanic anoxic event(~183 Ma)and the late Pliensbachian cooling event(~185 Ma).Lake-level evolution and hydrologic cycling on Earth’s surface during the Early Jurassic icehouses-to-(super)greenhouses are thus far poorly understood due to a lack of continuous high-resolution nonmarine *** we present a super-long nonmarine lake level record for this pivotal interval from the early Pliensbachian to Toarcian by sedimentary noise modeling,and construct a 16.7-Myr-long astronomical time scale(174.2 Ma to 190.9 Ma)based on cyclostratigraphy analysis of rock color datasets(CIE b*)of the Qaidam *** results document lake-level oscillations on a 5-to 10-million-year(Myr)scale which shows a pronounced correlation with long-term climate variation and extreme climatic events,and 1-to 2.5-Myr-scale lake-level changes that are prominently paced by the 2.4-Myr long-eccentricity forcing and the 1.2-Myr obliquity *** the Pliensbachian Stage,the 1.2-Myr-scale lake-level changes are in phase with the coeval sealevel *** forced growth and decay of the ephemeral or permanent ice sheets in polar regions are interpreted to control the synchronous ups-and-downs of continental lake level and global sea ***,during the Toarcian ice-free greenhouses to(super)greenhouses,the 1.2-Myr-scale lake-level variations show an anti-phase relationship with global sea level,indicating a‘seesaw’interaction between continental reservoirs(lakes and groundwater)and global *** 2.4-Myr longeccentricity cycles mainly regulate variation

Si anode is of paramount importance for advanced energy-dense lithium-ion batteries(libs).However,the large volume change as well as stress generates during its lithiation-delithiation process poses a great challenge to the long-term cycling and hindering its *** this work,a composite binder is prepared with a soft component,guar gum(GG),and a rigid linear polymer,anionic polyacrylamide(APAM).Rich hydroxy,carboxyl,and amide groups on the polymer chains not only enable intermolecular crosslinking to form a web.like binder,A2G1,but also realize strong chemical binding as well as physical encapsulating to Si *** resultant electrode shows limited thickness change of merely 9%on lithiation and almost recovers its original thickness on *** demonstrates high reversible capacity of 2104.3 mAh g^(-1)after 100 cycles at a current density of 1800 mA g^(-1),and in constant capacity(1000 mAh g^(-1))test,it also shows a long life of 392 ***,this soft-hard combining web.like binder illustrates its great potential in the future applications.

TbAb hydrate formation and growth in a microdevice under static and dynamic conditions

The natural gas hydrate has become one of the most promising future green energy sources on the *** natural gas hydrates mostly exist in the sediments with porous structure, so a solid understanding of the hydrate formation and growth processes in the porous medium is of significance for the exploitation of natural gas hydrate. The micro-packed bed device is one of the efficient microfluidic devices in the engineering field, but it has been rarely used for the hydrate-based research. In this study, a transparent micro-packed bed device filled with glass beads was developed to mimic the porous condition of sediments, and used to in-situ visualize the hydrate formation and growth habits in the pore spaces under both static and dynamic conditions. For the static experiment, two types of hydrate growth patterns in porous medium were observed and identified in the micro-packed bed device, which were the graincoating growth and pore-filling growth. For the dynamic condition, the hydrate formation, growth,distribution habits and hydrate blockage phenomena in the pore spaces were in-situ visually *** impacts of flowrate and subcooling on the pressure variation of the micro-packed bed and the duration of the hydrate growth under dynamic flow condition in pores were in-situ monitored and analyzed. The higher flowrate could result in the faster hydrate growth and more severe blockage in pores, but the effect of subcooling condition might be less significant at the high flowrate.

A connection between matrix orthogonal polynomials and non-ab.lian integrable lattices is investigated in this *** normalization factors of matrix orthogonal polynomials expressed using quasideterminants are shown to be the solutions to the non-ab.lian Toda lattice in semi-discrete and full-discrete ***,with a moment modification method,we demonstrate that the b¨acklund transformation of the non-ab.lian Toda lattice given by Popowicz(1983)is equivalent to the non-ab.lian Volterra lattice,whose solutions can be expressed using quasi-determinants as well.

Synthesis of Highly Ordered Large Size Mesoporous Silica and Effect of Stab.lization as Enzyme Supports in Organic Solvent

The FSM-16, MCM-41 and SbA-15 type hexagonal mesoporous silica materials having a highly arrange in order of the 2-dimensional structure were synthesized by using different silicon sources and surfactants. In the 2-dimensional silicate framework, pore size can be uniformly controlled by the combined use of the surfactants having different alkyl chains length and swelling agents (triisopropyl benzene). Pore-diameter of FSM-16 and MCM-41 can be expanded to 100 A, SbA-15 is 150 A. Crystal regularity decreased with increasing the pore-diameter in the FSM-16 derived from Kanemite (silicon source) and MCM-41 from water glass, its anionic char-acteristics on the pore wall may be higher than SbA-15 derived from oligomeric tetramethoxysilane (TMOS) is also reported. We have successfully used FSM-16 and MCM-41 as immob.lizing agents of enzyme having cationic residues under isoelectric point. The level of adsorption of enzymes in the FSM-16 and MCM-41 was relatively high, but was low in the SbA-15 support. The mechanism of enzyme adsorption in mesopore was suggested to be the ionic interactions. In aqueous solutions, horseradish peroxidase (HRP) immob.lized in FSM-16 containing 89A mesopore showed the highest loaded amounts(183mg/mg FSM),then a FSM-16 of pore-diameter 30 A only loaded a litter amounts (28mg/mg FSM) on the outside surface. The catalytic activity in organic solvent is high when HRP was immob.lized in FSM-16 and MCM-41, but is low in case of SbA- 15.
