AIM To analyze colorectal carcinogenesis and age-related DNA methylation alterations of gene sequences associated with epigenetic clock CpG sites. METHODS In silico DNA methylation analysis of 353 epigenetic clock Cp G sites published by Steve Horvath was performed using methylation array data for a set of 123 colonic tissue samples [64 colorectal cancer(CRC), 42 adenoma, 17 normal; GEO accession number: GSE48684]. Among the differentially methylated agerelated genes, secreted frizzled related protein 1(SFRP1) promoter methylation was further investigated in colonic tissue from 8 healthy adults, 19 normal children, 20 adenoma and 8 CRC patients using bisulfite-specific PCR followed by methylation-specific high resolution melting(MS-HRM) analysis. m RNA expression of age-related "epigenetic clock" genes was studied using Affymetrix HGU133 Plus2.0 whole transcriptome data of 153 colonic biopsy samples(49 healthy adult, 49 adenoma, 49 CRC, 6 healthy children)(GEO accession numbers: GSE37364, GSE10714, GSE4183, GSE37267). bACKGROUND Despite the improvement in the endoscopic hemostasis of non-variceal upper gastrointestinal bleeding(NVUGIb),rebleeding remains a major *** To assess the role of prophylactic transcatheter arterial embolization(PTAE)added to successful hemostatic treatment among NVUGIb *** We searched three databases from inception through October 19th,*** controlled trials(RCTs)and observational cohort studies were *** compared patients with NVUGIb receiving PTAE to those who did not get *** outcomes were rebleeding,mortality,reintervention,need for surgery and transfusion,length of hospital(LOH),and intensive care unit(ICU)*** the quantitative synthesis,odds ratios(ORs)and weighted mean differences(WMDs)were calculated with the random-effects model and interpreted with 95%confidence intervals(CIs).RESULTS We included a total of 3 RCTs and 9 observational studies with a total of 1329 patients,with 486 in the intervention *** was associated with lower odds of rebleeding(OR=0.48,95%CI:0.29–0.78).There was no difference in the 30-d mortality rates(OR=0.82,95%CI:0.39–1.72)between the PTAE and control *** who underwent PTAE treatment had a lower chance for reintervention(OR=0.48,95%CI:0.31–0.76)or rescue surgery(OR=0.35,95%CI:0.14–0.92).The LOH and ICU stay was shorter in the PTAE group,but the difference was non-significant[WMD=-3.77,95%CI:(-8.00)–0.45;WMD=-1.33,95%CI:(-2.84)–0.18,respectively].CONCLUSION PTAE is associated with lower odds of rebleeding and any reintervention in ***,further RCTs are needed to have a higher level of evidence.

AIM： To investigate the prevalence and clinical significance of ＂anti-Hbc alone＂ in an unselected population of patients and employees of a university hospital in southern Germany. METHODS： All individuals with the pattern ＂anti-Hbc alone＂ were registered over a time span of 82 mo. HbVDNA was measured in serum and liver samples, and clinical charts were reviewed. RESULTS： Five hundred and fifty two individuals were ＂anti-Hbc alone＂（of 3004 anti-Hbc positive individuals; 18.4%）, and this pattern affected males （20.5%） more often than females （15.3%; P〈0.001）. HbV-DNA was detected in serum of 44 of 545 ＂anti-Hbc alone＂ individuals （8.1%）, and in paraffin embedded liver tissue in 16 of 39 patients tested （41.0%）. There was no association between the detection of HbV genomes and the presence of biochemical, ultrasonic or histological signs of liver damage. Thirty-eight ＂anti-Hbc alone＂ patients with cirrhosis or primary liver carcinoma had at least one additional risk factor. HCV-coinfection was present in 20.4% of all individuals with ＂anti-Hbc alone＂ and was the only factor associated with a worse clinical outcome. CONCLUSION： In an HbV low prevalence area, no evidence is found that HbV alone causes severe liver damage in individuals with ＂anti-Hbc alone＂. Recommendations for the management of these individuals are given.

Although road agencies need to provide road infrastructure that is beneficial for road users, little is known about how the activities of the agencies influence the value creation of road infrastructure. From a service-dominant logic perspective, the importance of road main- tenance and traffic management activities for the contribution of road infrastructure to the value-creation process of road users is investigated. Road agencies facilitate the value creation of road users by maintaining, upgrading or renewing road infrastructure, the provision of information about the current traffic situation, possible redirection routes in case of traffic jams, and suggestions for appropriate driving behavior. based on a structured questionnaire, data were collected among motorists in Singapore and analyzed by means of a partial least square modeling approach. The analysis revealed that road cleanliness and road evenness have a significant effect on the experience of road maintenance. Important and significant indicators for the experience of traffic management are the clarity of road signs and the efficiency of traffic redirection. A main conclusion of the research is that for traffic-intensive networks, both road maintenance and traffic man- agement activities are important contributors to the value creation of road infrastructure with a slightly stronger contribution of traffic management activities. Road agencies need to find appropriate maintenance strategies which reduce and coordinate simultaneous maintenance interventions on the network to such an extent that traffic management activities are able to minimize any considerable loss of traffic flow.

Primary Research of Immunological Mechanism of Combined Hepatitis A-Measles-Varicella Vaccine

To explore the primary humoral and cellular immunological mechanism of the combined hepatitis A-measles-varicella vaccine, the mice were inoculated with hepatitis A-measles-varicella vaccine by intraperitoneally and two weeks later, blood was collected to observe the mice＇s immunological status. Antibody level was measured to appraise the humoral immunity. At the same time, T lymphocyte surface marker, NK cell activity, LAK cell activity, delayed type hypersensitivity of skin, Mφ phagocytic function, mRNA level of cytokine IL-2 and IFN-γ plus lymphocyte transformation test were used to analyze the cellular immunity. The humoral immunity results show that the combined hepatitis A-measles-varicella vaccine produce the same antibody level as their corresponding univalent vaccine, and maintained fine immunogenicity and security. The result of cellular immunity shows that the combined vaccine could activate physical immunocyte, increase the regulative ability of cytokine, enhance the physical immune function and immune defense ability. The present research proved the security and better humoral and cellular immunity of combined hepatitis A-measles-varicella vaccine from the immunological point of view, which laid good foundation for further study and development.
