Objective:To assess the stability of 10 candidate internal control genes(ICGs),namely GAPDH,ACTB,RPL23,RPS15A,ATPSF1,GLUT5,HMBS,ATP2B4,PPIA,and BRP to normalize the transcriptional data from testes samples of Zebu and crossbred ***:Total RNA was isolated from testicular tissue of Zebu and crossbred bulls(n=6 each)between 2-8 years of *** was synthesized,and the quantitative real-time polymerase chain reaction(PCR)was *** cycle threshold values were used for the analysis of the stability of *** different statistical algorithms:geNorm,Normfinder,BestKeeper,and RefFinder,were used to assess the stability of these ***:ATPSF1,HMBS,PPIA,and RPS15A were the most reliable and stable ICGs for Zebu testes,and ATPSF1,RPL23,and PPIA for crossbred ***:A panel of stable ICGs(ATPSF1,HMBS,PPIA,RPS15A for Zebu and ATPSF1,RPL23,and PPIA for crossbred)for normalization of gene expression data in testes samples can be helpful for researchers to conduct functional genomics studies at the testicular level in cattle bulls.

Early versus delayed laparoscopic cholecystectomy in uncomplicated biliary colic: An observational study

Objective:Biliary colic is a condition treated with laparoscopic ***,the outcomes of surgery depend on early or delayed time *** research findings reported no benefits of early over delayed,on contra,other reported *** study aims to compare the benefits associated with early and delayed laparoscopic cholecystectomy among uncomplicated biliary colic ***:This observational study included patients with right upper abdominal pain and abdominal ultrasound showing *** who were admitted at the first and second visits(within 6 weeks of the first visit)were assigned to the early and delayed groups,*** participants were followed up for one-week *** diagnosis of the patient,postoperative hospital stay,duration of surgery and complications were noted and compared ***:A total of 80 patients were included,40 each in the early group and delayed *** patients in the two groups had comparable mean ages(40.55±13.12 y vs.40.45±12.06 y,p=0.972).The early group had more female patients(72.5%vs.45.0%,p=0.012).The duration of hospital stay(2.18±0.38 d vs.2.68±1.04 d,p=0.009)and duration of surgery(61.63±3.64 min vs.71.13±16.19 min,p=0.001)were found to be significantly different between the early and delayed *** 1(2.5%)patient in both groups was converted to open *** biliary colic requiring hospital admission was seen in 1(2.5%)patient and 6(15.0%)patients,acute cholecystitis in 2(5.0%)and 6(15.0%),biliary pancreatitis in 1(2.5%)and 2(5.0%),and obstructive jaundice in 1(2.5%)and 1(2.5%)in the early and delayed groups,respectively,with insignificant differences(p>0.05).Conclusion:Early laparoscopic cholecystectomy decreases the operating time and duration of hospital *** terms of postoperative complications,our study did not find any significant difference between the groups.

There is intense interest in uncovering design rules that govern the formation of various structural phases as a function of chemical composition in multi-principal element alloys (MPEAs).In this paper,we develop a machine learning (ML) approach built on the foundations of ensemble learning,post hoc model interpretability of black-box models,and clustering analysis to establish a quantitative relationship between the chemical composition and experimentally observed phases of *** originality of our work stems from performing instance-level (or local) variable attribution analysis of ML predictions based on the breakdown method,and then identifying similar instances based on k-means clustering analysis of the breakdown *** also complement the breakdown analysis with Ceteris Paribus profiles that showcase how the model response changes as a function of a single variable,when the values of all other variables are *** from local model interpretability analysis uncover key insights into variables that govern the formation of each *** developed approach is generic,model-agnostic,and valuable to explain the insights learned by the black-box *** interactive web application is developed to facilitate model sharing and accelerate the design of MPEAs with targeted properties.

The study of cordierite ceramic is the need of the hour in present technological world for various advanced engineering ***,which has relatively poor mechanical properties,has induced the use of various dopants to study the improvement in mechanical *** of seeking new materials with good mechanical properties has revived the research on ceria doped *** research conducted on pure cordierite and ceria doped cordierite ceramics has investigated the results of characterization and studied their *** cordierite and cordierite-ceria(CeO_(2))composite ceramics synthesized for various stoichiometric composition(5-20 wt%)were compacted at 240 MPa and sintered between 600℃and 1350℃for 3 *** characterization techniques used in this study were X-ray diffraction(XRD),thermogravimetric(TG)analysis and Fourier transform infrared(FTIR).The density was calculated using the Archimedes *** of the ceria addition on cordierite’s mechanical properties such as hardness,flexural strength and fracture toughness and on thermal properties as thermal expansion was *** results confirmed the presence of cordierite and ceria in the samples heat treated at 1350℃.Results of FTIR and TG analyses revealed the formation of cordierite and the effect of ceria *** mechanical properties studied were found to be encouraging and confirmed the suitability of cordierite-ceria as an alternate material for cordierite.

The Pleurotus florida is recognized as a medicinal and edible mushroom and the present study intends to reveal the active isomeric molecules from this *** *** was cultivated using different nutrient supplements:groundnut husk,maize powder,horse gram powder and coconut oil-cake *** gram supplement showed the higher mushroom yield and henceit was used for the cultivation of *** extract of *** was found to be efficient in antioxidant activity among ethanol,aqueous,ethyl acetate and hexane *** bioactive fraction 3-methoxy-4-hydroxy cinnamic acid(PF5)was isolated and purified from the methanolic extract of *** by column chromatography,thin layer chromatography(TLC)and gas chromatography-mass spectrum(GC-MS)and further it was characterized by Nuclear magnetic resonance(NMR).The PF5 was tested for its DPPH and reducing power assays,and the IC_(50) values were found to be 21.7μg/mL and 105μg/mL,*** found that the cytotoxic effect of 3-methoxy-4-hydroxy cinnamic acid was tested against the lung cancer cell line using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT),sulphorodamine B(SRB)and trypan blue assays which exhibited a higher cytotoxic effect(CTC_(50),645μg/mL).These results suggested that 3-methoxy-4-hydroxy cinnamic acid from *** could be explored as a novel and potent natural antioxidant and cancer preventive agent,alternative to existing synthetic molecules.

Insilico structural analysis of parasporin 2 protein sequences of non-toxic bacillus thuringiensis

The unusual and remarkable property of parasporin 2 of non-insecticidal Bacillus thuringiensis is specifically recognizing and selectively targeting human leukemic cell lines. The 37-kDa inactive nascent protein is proteolytically cleaved to the 30-kDa active form that loses both the N-terminal and the C-terminal segments. Accumulated cytological and biochemical observations on parasporin-2 imply that the protein is a pore-forming toxin. To confirm the hypothesis, insilico analysis was performed using homology modeling. The resulting model of parasporin 2 protein is unusually elongated and mainly comprises long β-strands aligned with its long axis. It is similar to aerolysin-type β-pore-forming toxins, which strongly reinforce the pore-forming hypothesis. The molecule can be divided into three domains. Domain 1, comprising a small β-sheet sandwiched by short α-helices, is probably the target-binding module. Two other domains are both β-sandwiches and thought to be involved in oligomerization and pore formation. Domain 2 has a putative channel-forming β-hairpin characteristic of aerolysin-type toxins. The surface of the protein has an extensive track of exposed side chains of serine and threonine residues. The track might orient the molecule on the cell membrane when domain 1 binds to the target until oligomerization and pore formation are initiated. The β-hairpin has such a tight structure that it seems unlikely to reform as postulated in a recent model of pore formation developed for aerolysin-type toxins. Parasporin 2 (Accession no: BAD35170) protein sequence analysis indicated two different domains namely, aerolysin toxin and clostridium toxin domain based on different database searches (CDD and Pfam). It showed a close similarity with the available PDB template (PDB id: 2ZTB) of parasporin which has cytocidal activity against MOLT-4, HL60 and Jurkat cell lines. Bacillus thuringiensis strains isolated from Madurai, TamilNadu, India were evaluated for their mosquitocidal activity, as well as cry and cyt genes diversity. It revealed that 99% of the parasporal crystal morphology of these isolates was spherical in nature and a variable percentage (0% - 100%) of toxicity was observed against Culex quinquefasciatus and Aedes aegypti. PCR analysis revealed that 53% of the isolates were positive for the various cry and cyt genes tested, whereas 47% did not produce any PCR product for the cry gene analyzed. Diverse pattern of cry and cyt genes distribution was observed even in the isolates from the same sample. B. thuringienis subsp. LDC-9 showed three-fold higher toxicity against Culex quinquefasciatus than that of B. thuringiensis var israelensis which might be used as a potential strain to control mosquitoes in near future after field evaluation.
