A strategy for uncovering germline variants altering anti-tumor CD8 T cell response

Among many factors known to alter the outcomes of T cell receptor(TCR)-induced proximal signaling,the role of human germline variants in dictating the individuality of the anti-tumor CD8 T cell response has remained challenging to ***,we describe a convenient strategy for molecular and functional characterization of phosphotyrosine-altering non-synonymous single nucleotide variations(pTyr-sNVs)that directly impact TCR-induced proximal phosphotyrosine motif-based signaling *** devise an experimental co-cultivation set-up comprising a C57BL/6 mouse-derived metastatic melanoma cell line engineered to constitutively present ovalbumin(OVA)antigens and retrovirally engineered syngeneic major histocompatibility complex(MHC)Class I restricted OVA TCR-transgenic CD8 T cells(OT-I).Using the synthetic version of pTyr-sNV rs1178800678-G/T-encoding integrin alpha 4(ITGA4)p.s1027I variant as a prototype,we show that under identical TCR stimulation conditions,genetically determined membrane-proximal immunoreceptor tyrosin activation motif(ITAM)results in increased tyrosine phosphorylation of 70 kDa zeta-chain-associated protein(ZAP70)and the levels of cytotoxic effector molecule granzyme B(GZMB),which in turn result in enhanced cytotoxic activity against metastatic melanoma cell *** strategy paves the way for rapid molecular and functional characterization of anti-tumor immune response-linked germline pTyr-sNVs so as to improve our understanding of the genetic basis of individual-to-individual differences in anti-tumor CD8 T cell response.

Modelling of electric vehicle charging station and controlled by fuzzy logic controller with different modes of operation

The main objective of implementing charging stations is to ensure the good charging to theElectric Vehicles by using a solar PV array which is interconnected to the battery energy storagesystems. The charging station regulates the supply voltage and frequency without the use of amechanical speed governor. It also assures that energy gained from grid or by the DG set willhave the unity power factor (UPF) when the load is nonlinear. Besides this, the Point of CommonCoupling (PCC) voltage is synchronised with the grid/generator voltage in order to providecontinuous charging. In order to increase the optimal efficiency of the charging stations, thecharging stations will perform the active/reactive power transfer from the vehicle to grid, vehicleto house and vehicle to vehicle (V2V) power transfer. The operational experiment of the chargingstation is simulated and verified by using MATLAB/sIMULINK.

Digital image steganography technique based on hiding the secret data behind of cover image in such a way that it is not detected by the human visual *** paper presents an image scrambling method that is very useful for grayscale secret *** this method,the secret image decomposes in three parts based on the pixel’s threshold *** division of the color image into three parts is very easy based on the color channel but in the grayscale image,it is difficult to *** proposed image scrambling method is implemented in image steganography using discrete wavelet transform(DWT),singular value decomposition(sVD),and sorting *** is no visual difference between the stego image and the cover *** extracted secret image is also similar to the original secret *** proposed algorithm outcome is compared with the existed image steganography *** comparative results show the strength of the proposed technique.

Analysing the combined effect of crystallographic orientation and grain refinement on mechanical properties and corrosion behaviour of ECAPed ZE41 Mg alloy

Two step equal channel angular pressing carried out on as cast ZE41 Mg alloy resulted in a remarkable grain *** compared to grain size of 46p.m in as cast sample,refinement upto 2.5^01 was achieved after 8th pass equal channel angular pressing(ECAP).The combined effect of crystallographic orientation and grain refinement was investigated by analysing the mechanical properties and corrosion behaviour of ZE41 Mg alloy using electron back scattered diffraction(EBsD).The first stage comprises of 1st,2nd,3rd and 4th passes at a processing temperature of 300℃while the 5th,6th,7th and 8th passes were ECAPed at 275℃in second *** mechanical properties of ZE41 Mg 158 yield tensile strength(YTs),230 ultimate tensile strength(UTs)and 7%elongation in as cast condition is enhanced to 236 YTs,295 UTs and 19.76%,respectively,after first stage *** yield tensile strength deteriorated due to the effect of texture predominating grain refinement during the second stage *** corrosion resistance of ZE41 Mg was significantly enhanced by ECAP and is inferred from electrochemical impedance spectroscopy(EIs)and potentiodynamic polarisation *** role of microstructure was minimal on corrosion behaviour of ZE41 Mg due to extra resistance when tested in 0M ***,the influence of grain refinement greatly influenced the improvement in corrosion resistance of ZE4I Mg rather than crystallographic orientation observed from *** contrast,the crystallographic orientation predominated the effect of grain refinement during ZE41 Mg corrosion in chloride containing(0.1 M and 1 M NaCl)*** the observation of results it is found that equal channel angular pressing has the dual advantage of improving mechanical properties and corrosion resista nee of ZE41 Mg alloy.

Nowadays,due to the increase in information resources,the number of parameters and complexity of feature vectors *** offermore practical solutions instead of exact solutions for the solution of this *** Emperor PenguinOptimizer(EPO)is one of the highest performing meta-heuristic algorithms of recent times that imposed the gathering behavior of emperor *** shows the superiority of its performance over a wide range of optimization problems thanks to its equal chance to each penguin and its fast convergence *** traditional EPO overcomes the optimization problems in continuous search space,many problems today shift to the binary search ***,in this study,using the power of traditional EPO,binary EPO(BEPO)is presented for the effective solution of binary-nature *** algorithm uses binary search space instead of searching solutions like conventional EPO algorithm in continuous search *** this purpose,the sigmoidal functions are preferred in determining the emperor *** addition,the boundaries of the search space remain constant by choosing binary ***’s performance is evaluated over twenty-nine benchmarking *** evaluations are made to reveal the superiority of the BEPO *** addition,the performance of the BEPO algorithm was evaluated for the binary feature selection *** experimental results reveal that the BEPO algorithm outperforms the existing binary meta-heuristic algorithms in both tasks.

Demon Genes May Deform Common syndromes: Collagen VI Gene Change in Down syndrome Unifies the Medical and Molecular Approach to Hypermobility Disorders

Purpose: To alert the medical community that whole exome sequencing can find accessory gene changes in well-known syndromes that alter preventive health care and management. Meaning: A collagen type VI gene change adds muscle weakness, hypermobility, and dysautonomia concerns to usual management considerations for Down syndrome. Methods: Commercial whole exome sequencing combined with clinical interpretation of DNA sequence change added new considerations to patient management and parental counsel. Results: An 11-year-old child with the trisomy 21 form of Down syndrome who was evaluated for extraordinary joint laxity had a heterozygous collagen type VI aspartic to glutamic acid (COL6A3 c.6360 C>G ***2120Glu) gene change found by whole exome sequencing. The DNA variant was qualified as having strong relevance to the enhanced hypermobility due to prior association of collagen 6 gene changes with myopathy. Conclusions: Dual diagnosis of Ehlers-Danlos syndrome was not assigned because the patient lacked criteria like bruising, unusual scars, or selected dysautonomia symptoms. The concept of a hypermobility spectrum offers advantages for management of its constituent conditions if clinically guided ascertainment and DNA diagnostics are employed.
