Background Triple-negative breast cancer(TNBC) is the most aggressive breast cancer subtype and leads to the poorest patient outcomes despite surgery and chemotherapy treatment. Exploring new molecular mechanisms of TNBC that could lead to the development of novel molecular targets are critically important for improving therapeutic options for treating *** We sought to identify novel therapeutic targets in TNBC by combining genomic and functional studies with lipidomic analysis, which included mechanistic studies to elucidate the pathways that tie lipid profile to critical cancer cell properties. Our studies were performed in a large panel of human breast cancer cell lines and patient *** Comprehensive lipid profiling revealed that phospholipid metabolism is reprogrammed in TNBC cells. We discovered that patatin-like phospholipase domain-containing lipase 8(PNPLA8)is overexpressed in TNBC cell lines and tissues from breast cancer patients. Silencing of PNPLA8disrupted phospholipid metabolic reprogramming in TNBC, particularly affecting the levels of phosphatidylglycerol(PG), phosphatidylcholine(PC), lysophosphatidylcholine(LPC) and glycerophosphocholine(GPC). We showed that PNPLA8 is essential in regulating cell viability,migration and antioxidation in TNBC cells and promoted arachidonic acid and eicosanoid production, which in turn activated PI3K/Akt/Gsk3β and mAPK *** Our study highlights PNPLA8 as key regulator of phospholipid metabolic reprogramming and malignant phenotypes in TNBC, which could be further developed as a novel molecular treatment target.

Effect of a Wildlife-Livestock Interface on the Prevalence of Intra-Erythrocytic Hemoparasites in Cattle

We conducted a cross-sectional study to establish the effect of proximity of livestock to a wildlife-livestock interface on the relative abundance of intra-erythrocytic hemoparasites in cattle. Blood samples were obtained from 131 randomlyselected cattle raised around Queen Elizabeth National Park. Cattle-farm location was determined by using Global Positioning System device from an arbitrarily reference point. Giemsa-stained blood smears were examined microscopically for intra-erythrocytic hemoparasites. Correlational analysis was used to examine the relationship between farm location and prevalence, whereas risk ratios were used to determine the strength of mixed hemoparasitic infections among cattle, using a significant level of α = 0.05. The location of a cattle farm significantly predicted the prevalence of Anaplasma (rs = 0.33, p 0.05) and Theileria (rs = 0.57, p 0.01) but, farm’s proximity to QENP did not explain the variation in the prevalence of Babesia (rs = 0.14, p 0.2). Although mixed infections occurred in 15% of sampled cattle, concurrent infection of cattle with A. marginale and B. bigemina [RR = 36;95% CI (7.191);p which was recorded. This study demonstrated that unlike the prevalence of B. bigemina, the prevalence of T. parva and A. marginale in livestock significantly increased with close proximity to a wildlife-livestock interface.

背景与目的对于严重面容损毁的成年患者而言，其术后长期心理学影响的评估仍有限。本研究旨在确定此类患者的长期心理学机能，并评估获得性面容损毁患者与无面容损毁者间的不同。同样的，先天性面容损毁者的心理机能程度与其对面容的满意度相关，此类患者会因其他人对其面容的负面评价、自尊心和面部畸形的严重程度而感到不同程度的忧虑。方法本研究共对59例严重先天性面容损毁者、59例创伤获得性面容损毁者和120例无面容损毁者进行分析。让所有被调查者进行标准的心理学、身体检查和人口统计学问卷调查，包括面容负面评价忧虑量表（Fear of Negative Appear—ance Evaluation Scale）、自信心量表（Rosenberg Self-Esteem Scale）、住院焦虑和抑郁量表（Hospital Anxietyand Depression Scale）、阿成贝切自评表（Achenbach Adult Self-Repor）、SF-36健康调查表（SF-36Health Survey）和视觉类比量表（Visual Analog Scale）。结果先天性面容损毁的成年患者有相对正常的心理机能，但相对于无面容损毁的成年患者，其表现出存在藏在心底的问题。与获得性面容损毁者相比，先天性者仅在身体检查量表部分的生活质量方面表现出较少的问题。除了身体检查部分的量表的有关生活质量的方面以外，患者对面容的满意度、对面容负面评价的忧虑和自尊心都是很好的可预测其心理机能不同方面的因素。结论（通过手术）改善患者外观评价满意度、（通过心理支持）增强其自尊心或降低其对面容负面评价的忧虑，将有助于其提高长期心理机能。今后的研究将会把重点放在对个体患者的分析和对不良适应相关危险因素的分析。

Brain antigens in functionally distinct antigen-presenting cell populations in cervical lymph nodes in mS and EAE

Drainage of central nervous system(CNS) antigens to the brain-draining cervical lymph nodes(CLN) is likely crucial in the initiation and control of autoimmune responses during multiple sclerosis(mS). We demonstrate neuronal antigens within CLN of mS patients. In monkeys and mice with experimental autoimmune encephalomyelitis(EAE) and in mouse models with non-inflammatory CNS damage, the type and extent of CNS damage was associated with the frequencies of CNS antigens within the cervical lymph nodes. In addition, CNS antigens drained to the spinal-cord-draining lumbar lymph nodes. In human mS CLN, neuronal antigens were present in pro-inflammatory antigen-presenting cells(APC), whereas the majority of myelincontaining cells were anti-inflammatory. This may reflect a different origin of the cells or different drainage mechanisms. Indeed, neuronal antigen-containing cells in human CLN did not express the lymph node homing receptor CCR7, whereas myelin antigen-containing cells in situ and in vitro did. Nevertheless, CLN from EAE-affected CCR7-deficient mice contained equal amounts of myelin and neuronal antigens as wild-type mice. We conclude that the type and frequencies of CNS antigens within the CLN are determined by the type and extent of CNS damage. Furthermore, the presence of myelin and neuronal antigens in functionally distinct APC populations within mS CLN suggests that differential immune responses can be evoked.
