Combinatorial Complexity in a Transcriptionally Centered Signaling Hub in Arabidopsis

A subfamily of four Phytochrome （phy）-Interacting bHLH transcription Factors （PIFs） collectively promote skotomorphogenic development in dark-grown seedlings. This activity is reversed upon exposure to light, by photoacti- vated phy molecules that induce degradation of the PIFs, thereby triggering the transcriptional changes that drive a tran- sition to photomorphogenesis. The PIFs function both redundantly and partially differentially at the morphogenic level in this process, To identify the direct targets of PIF transcriptional regulation genome-wide, we analyzed the DNA-binding sites for all four PIFs by ChlP-seq analysis, and defined the genes transcriptionally regulated by each PIF, using RNA-seq analysis of pif mutants. Despite the absence of detectable differences in DNA-binding-motif recognition between the PIFs, the data show a spectrum of regulatory patterns, ranging from single PIF dominance to equal contributions by all four. Similarly, a broad array of promoter architectures was found, ranging from single PIF-binding sites, containing single sequence motifs, through multiple PIF-binding sites, each containing one or more motifs, with each site occupied prefer- entially by one to multiple PIFs. Quantitative analysis of the promoter occupancy and expression level induced by each PIF revealed an intriguing pattern. Although there is no robust correlation broadly across the target-gene population, examination of individual genes that are shared targets of multiple PIFs shows a gradation in correlation from strongly positive, through uncorrelated, to negative. This finding suggests a dual-layered mechanism of transcriptional regulation, comprising both a continuum of binding-site occupancy by each PIF and a superimposed layer of local regulation that acts differentially on each PIF, to modulate its intrinsic transcriptional activation capacity at each site, in a quantitative pattern that varies between the individual PIFs from gene to gene. Dysregulated expression and distribution of Kif5αin neurites of wobbler motor neurons

Impaired axonal transport has been observed in patients with amyotrophic lateral sclerosis(ALS)and in animal models,suggesting that transport proteins likely play a critical role in the pathological mechanism of *** of Kinesin-family-member 5α(Kif5α),a neuron-specific isoform of heavy chain kinesin family,has been described in several neurological disorders,in humans and animal models,including *** this study,we determined Kif5αexpression by gene sequencing,quantitative reverse transcription-polymerase chain reaction,and western blot assay in the cervical spinal cord of wobbler mice and immunofluorescence staining in dissociated cultures of the ventral ***,we observed the distribution of Kif5αand mitochondria along motor neuronal branches by confocal *** results showed that Kif5αexpression was greatly dysregulated in wobbler mice,which resulted in altered distribution of Kif5αalong motor neuronal branches with an abnormal mitochondrial ***,our results indicate that dysregulation of Kif5 and therefore abnormal transport in motor neuronal branches in this ALS model could be causative for several pathological findings at the cellular level,like misallocation of cytoskeletal proteins or organelles like mitochondria.

Atm deficient zebrafish model reveals conservation of the tumour suppressor function and a role in fertility

Biallelic loss-of-function variants in ATM (Ataxia Telangiectasia Mutated) cause Ataxia Telangiectasia (AT), a rare disorder associated with cerebellar degeneration and ataxia, cancer predisposition, infertility, growth retardation, etc. ATM is a phosphoinositide 3-kinase-related kinase (PIKK) with a role in DNA repair and maintenance of genome stability. Studying a multisystem genetic disease like AT requires animal models to ascertain its pathogenesis at the level of tissues, organs and the organism. Due to its small size, cheap maintenance, large progeny, rapid development and initial transparency, zebrafish (Danio rerio) is an increasingly popular vertebrate model organism, suitable for genetic modifications and large-scale in vivo therapeutic screens as embryos are chemically permeable to small compounds. Currently, no zebrafish model for AT exists.1 We generated atm knock-outs through CRIPSR-Cas9 mutagenesis. We show that atm conserved its function as a tumour suppressor gene and is involved in gametogenesis and fertility. Therefore, this mutant is of great value for further studies investigating the role of atm in reproduction and tumorigenesis.

24-Hour movement behaviours and COVID-19 among children in the Kingdom of Saudi Arabia:A repeat cross-sectional study

This study investigated how children's 24-hour(24-h)movement behaviours were affected by SARS-CoV-2(COVID-19)in the Kingdom of Saudi *** research examined 24-h movement behaviours in Saudi Arabia seven months after the World Health Organization(WHO)declared COVID-19 a global *** repeat cross-sectional study examined changes in 24-h movement behaviours 12 months after the WHO *** Time 2 survey repeated five months(1 March–15 May 2021)after Time 1 survey(1 October–11 November 2020).The survey was distributed to parents of children aged 6–12 years across Saudi Arabia via an online *** were classified as meeting 24-h movement guidelines if they reported uninterrupted sleep for 9–11 h per night,2 h of recreational sedentary screen time(RST)per day and-60 min of moderate-to vigorous-intensity physical activity(MVPA)per day.A total of 1045 parents from all regions of Saudi Arabia responded(42.4%).Only 1.8%of children met all components of the guidelines,compared to 3.4%in Time *** the present study,girls spent more days per week in MVPA-60 min duration than boys(3.0 vs 2.6;p=0.025),while boys had spent more days per week engaged in activities that strengthened muscle and bone than girls(3.0 vs 2.8;p=0.019).Healthy levels of physical activity(PA),sedentary behaviour(SB)and sleep further declined in Saudi children five months after the Time 1 *** challenges require urgent intervention to ensure children's movement behaviours improve as Saudi Arabia moves out of the COVID-19 pandemic.

Macrocycle-based glycoclusters, on account of their promising anti-adhesive properties against bacteria,are potential therapeutic alternatives to classic antibiotics through the much less explored anti-adhesive strategy. In this study, a series of constitutionally-pure pentavalent glycoclusters was prepared by conjugating assorted azido-carbohydrates onto a penta-propargyl rim-differentiated pillar[5]arene(RD-P[5])scaffold through Cu(I)-catalyzed azide–alkyne cycloaddition “click” reactions. Their binding towards therapeutically relevant bacterial lectins, such as Lec A and Lec B from Pseudomonas aeruginosa and concanavalin A(Con A), were evaluated subsequently by isothermal titration calorimetric studies. Most of these isomerfree RD-P[5] pentavalent glycoclusters, except the fucosylated ones, display good affinities to ***, the dissociation constants observed are similar to those displayed by an analogous pentavalent glycocluster consisting of four P[5] constitutional isomers, in which the RD-P[5] component merely accounts for 7% in the mixture. Our results revealed that high constitutional purity is not essential for achieving effective multivalent interactions between P[5]-based glycoclusters and lectins, presumably as a result of the conformationally labile nature of the P[5] scaffold. This information provides valuable design principles for low-cost and facile syntheses of glycosylated P[5]s for biomedical applications.

Percutaneous abscess drainage is a procedure commonly performed by interventional radiologists to provide source control on infections using CT or ultrasound *** interventionalist has many different sizes and shapes of catheters to treat abscesses of varying sizes and locations,but the general approach to each abscess is similar:provide a percutaneous route for purulence,bacteria,necrotic tissue,and other debris to escape the *** generally considered a low-risk procedure,adverse events can occur due to operator error or other *** present a unique case of an abscess drain placed into a right upper quadrant abscess that formed following laparoscopic cholecystectomy that perforated and entered the *** physicians,both in the emergency department and the radiology reading room,were able to rapidly rule out more common post-operative complications and make the correct diagnosis,likely preventing dangerous sequelae from developing in this patient.

Effects of personalized exercise prescriptions and social media delivered through mobile health on cancer survivors’physical activity and quality of life

Purpose This study aimed to examine the effects of a multi-component mobile health intervention(wearable,apps,and social media)on cancer survivors’(CS')physical activity(PA),quality of life,and PA determinants compared to exercise prescription only,social media only,and attention control *** A total of 126 CS(age=60.37±7.41 years,mean±SD)were recruited from the United *** study duration was 6 months and participants were randomly placed into 4 *** participants received a Fitbit tracker and were instructed to install its companion app to monitor their daily ***(1)received previously established weekly personalized exercise prescriptions via email,(2)received weekly Facebook health education and interacted with one another,(3)received both Conditions 1 and 2,or(4)were part of the control condition,meaning they adopted usual *** PA daily steps,quality of life(i.e.,physical health and mental health),and PA determinants(e.g.,self-efficacy,social support)were measured at baseline,3 months,and 6 *** The final sample size included 123 *** results revealed only the multi-component condition had greater improvements in PA daily steps than the control condition post-intervention(95%confidence interval(95%CI):368–2951;p<0.05).Similarly,those in the multi-component condition had significantly greater increased physical health than the control condition(95%CI:–0.41 to–0.01;p<0.05)over *** addition,the social media condition had significantly greater increased perceived social support than the control condition(95%CI:0.01–0.93;p<0.05).No other significant differences on outcomes were *** The study findings suggest that the implementation of a multi-component mobile health intervention had positive effects on CS PA steps and physical ***,offering social media intervention has the potential to improve CS perceived social support.
