Nicotinamide treatment of plants and plant cell cultures has been shown to promote defense and decrease levels of DNA *** the present study,we used RNA-seq technology to study overall changes in gene expression induced in roots of 3-month-old spruce(Picea abies)seedlings grown from nicotinamide-treated seeds to examine the molecular mechanisms underlying the defense *** 350 genes were identified as differentially expressed in roots after the seed *** response genes,including transcription factors MYb77 and LHY and two chitinase enzymes,were generally upregulated,whereas genes thought to be involved in epigenetic regulation such as DDM1,known to promote DNA methylation,were present at high frequency among the downregulated *** all samples,the expression of downregulated epigenetic-related genes was highly correlated with the nicotinamide treatment,indicating a common *** results support an earlier hypothesis regarding a potential role of nicotinamide as a defense-signal mediator.

The increasing use of cloud-based devices has reached the critical point of cybersecurity and unwanted network *** environments pose significant challenges in maintaining privacy and *** approaches,such as IDS,have been developed to tackle these ***,most conventional Intrusion Detection System(IDS)models struggle with unseen cyberattacks and complex high-dimensional *** fact,this paper introduces the idea of a novel distributed explainable and heterogeneous transformer-based intrusion detection system,named INTRUMER,which offers balanced accuracy,reliability,and security in cloud settings bymultiplemodulesworking together within *** traffic captured from cloud devices is first passed to the TC&TM module in which the Falcon Optimization Algorithm optimizes the feature selection process,and Naie bayes algorithm performs the classification of *** selected features are classified further and are forwarded to the Heterogeneous Attention Transformer(HAT)*** this module,the contextual interactions of the network traffic are taken into account to classify them as normal or malicious *** classified results are further analyzed by the Explainable Prevention Module(XPM)to ensure trustworthiness by providing interpretable *** the explanations fromthe classifier,emergency alarms are transmitted to nearby IDSmodules,servers,and underlying cloud devices for the enhancement of preventive *** experiments on benchmark IDS datasets CICIDS 2017,Honeypots,and NSL-KDD were conducted to demonstrate the efficiency of the INTRUMER model in detecting network trafficwith high accuracy for different *** outperforms state-of-the-art approaches,obtaining better performance metrics:98.7%accuracy,97.5%precision,96.3%recall,and 97.8%*** results validate the robustness and effectiveness of INTRUMER in securing diverse cloud environments against sophisticated cyber threats.

In the rapidly evolving landscape of television advertising,optimizing ad schedules to maximize viewer engagement and revenue has become *** methods often operate in silos,limiting the potential insights gained from broader data analysis due to concerns over privacy and data *** article introduces a novel approach that leverages Federated Learning(FL)to enhance TV ad schedule optimization,combining the strengths of local optimization techniques with the power of global Machine Learning(ML)models to uncover actionable insights without compromising data *** combines linear programming for initial ads scheduling optimization with ML—specifically,a K-Nearest Neighbors(KNN)model—for predicting ad spot *** into account the diversity and the difficulty of the ad-scheduling problem,we propose a prescriptivepredictive approach in which first the position of the ads is optimized(using Google’s OR-Tools CP-SAT)and then the scheduled position of all ads will be the result of the optimization ***,this output becomes the target of a predictive task that predicts the position of new entries based on their characteristics ensuring the implementation of the scheduling at large scale(using KNN,Light Gradient boosting Machine and Random Forest).Furthermore,we explore the integration of FL to enhance predictive accuracy and strategic insight across different broadcasting networks while preserving data *** FL approach resulted in 8750 ads being precisely matched to their optimal category placements,showcasing an alignment with the intended diversity ***,there was a minimal deviation observed,with 1133 ads positioned within a one-category variance from their ideal placement in the original dataset.

The goal of this paper was to present knowledge on changes in the morphodynamic structure of the Ochotnica River(Polish Carpathians)and transformations within its active river zone since the end of the 19th *** study used a set of archival and contemporary cartographic materials and the results of three-fold field mapping of the morphodynamic structure of the *** and indirect human interference with the natural environment of the catchment and the fluvial system was taken into account in the *** of changes in the morphodynamic structure of the Ochotnica and its active river zone since the end of the 19th century indicated significant changes in the development trend of the *** contemporary structure of the Ochotnica is more complex than in the *** has been a fragmentation of the riverbed into sections with different morphodynamic *** proportion of erosive sections has increased,which explains the reduced in the width of the active river zone of the *** average width of the active river zone of the river between 1861 and 2022 has changed from 80 m to 18 *** impact has been identified as the main reason for the changes in the morphodynamic structure of the riverbed and its active river ***,only the unregulated sect ion is characterised by free lateral migration of the riverbed and the widest active river zone(~28 m).

Sweden has witnessed an increase in the rates of sexual crimes including *** of who the offenders of these crimes are is therefore of importance for *** aimed to study characteristics of individuals convicted of rape,aggravated rape,attempted rape or attempted aggravated rape(abbreviated rape+),against a woman≥18years of age,in *** using information from the Swedish Crime Register,offenders between 15 and 60years old convicted of rapeþbetween 2000 and 2015 were *** on substance use disorders,previous criminality and psychiatric disorders were retrieved from Swedish population-based registers,and Latent Class Analysis(LCA)was used to identify classes of rapeþoffenders.A total of 3039 offenders were included in the analysis.A major-ity of them were immigrants(n=1800;59.2%)of which a majority(n=1451;47.7%)were born outside of *** LCA identified two classes:Class A-low offending class(LOC),and Class b—high offending class(HOC).While offenders in the LOC had low rates of previous criminality,psychiatric disorders and substance use disorders,those included in the HOC had high rates of previous criminality,psychiatric disorders and substance use *** HOC may be composed by more“traditional”criminals probably known by the police,the LOC may represent individuals not previously known by the *** two separated classes,as well as our finding in regard to a majority of the offenders being immi-grants,warrants further studies that take into account the contextual characteristics among these offenders.

Increasing our understanding of primate gestural communication can provide new insights into language evolution.A key question in primate communication is the association between the social relationships of primates and their repertoire of *** analyses can reveal how primates use their repertoire of gestural communication to maintain their networks of family and friends,much as humans use language to maintain their social *** this study we examined the association between the repertoire of gestures(overall,manual and bodily gestures,and gestures of different modalities)and social bonds(presence of reciprocated grooming),coordinated behaviors(travel,resting,co-feeding),and the complexity of ecology(***,illumination)and sociality(party size,audience),in wild East African chimpanzees(Pan troglodytes schweinfurthii).A larger repertoire size of manual,visual gestures was associated with the presence of a relationship based on reciprocated grooming and increases in social complexity.A smaller repertoire of manual tactile gestures occurred when the relationship was based on reciprocated grooming.A smaller repertoire of bodily gestures occurred between partners who jointly traveled for *** gesture repertoire size was associated with social complexity,complex ecology also influenced repertoire *** evolution of a large repertoire of manual,visual gestures may have been a key factor that enabled larger social groups to emerge during ***,the evolution of the larger brains in hominins may have co-occurred with an increase in the cognitive complexity underpinning gestural communication and this,in turn,may have enabled hominins to live in more complex social groups.

background:Triple-negative breast cancer (TNbC), characterized by its lack of traditional hormone receptors and HER2, presents a significant challenge in oncology due to its poor response to conventional therapies. Autophagy is an important process for maintaining cellular homeostasis, and there are currently autophagy biomarkers that play an effective role in the clinical treatment of tumors. In contrast to targeting protein activity, intervention with protein-protein interaction (PPI) can avoid unrelated crosstalk and regulate the autophagy process with minimal interference pathways.Methods:Here, we employed Naive bayes, Decision Tree, and k-Nearest Neighbors to elucidate the complex PPI network associated with autophagy in TNbC, aiming to uncover novel therapeutic targets. Meanwhile, the candidate proteins interacting with beclin 2 were initially screened in MDA-Mb-231 cells using beclin 2 as bait protein by immunoprecipitation-mass spectrometry assay, and the interaction relationship was verified by molecular docking and CO-IP experiments after intersection. Colony formation, cellular immunofluorescence, cell scratch and 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) tests were used to predict the clinical therapeutic effects of manipulating candidate PPI.Results:by developing three PPI classification models and analyzing over 13,000 datasets, we identified 3733 previously unknown autophagy-related PPIs. Our network analysis revealed the central role of beclin 2 in autophagy regulation, uncovering its interactions with 39 newly identified proteins. Notably, the CO-IP studies identified the substantial interaction between beclin 2 and Ubiquilin 1, which was anticipated by our model and discovered in immunoprecipitation-mass spectrometry assay results. Subsequently, investigations showed that overexpressing beclin 2 increased Ubiquilin 1, promoted autophagy-dependent cell death, and inhibited proliferation and metastasis in MDA-Mb-231 cells.
