The pres.nt paper mainly deals.with a cas. s.udy of failed dump s.ope in wes.ern coalfield limited, Nagpur, India. A huge mas. of debris.flow had happened during the routine the activity of mining. The failed dump had a height of 75 m with 43? s.ope angle which had s.ipped forward by 18m. Repres.ntative loos. dump material s.mples.were collected from the s.te and tes.ed to determine the phys.co-mechanical properties.of dump material. The dump material cons.s.ed of loos. fragments.and lumps.of friable s.nds.one, s.ale, clay and carbonaceous.s.ale. To evaluate the condition of failure, a well known, s.ear s.rength reduction tech-nique has.been applied to achieve the des.red factor of s.fety us.ng two dimens.onal finite element code. Fi-nally, a economical, s.s.ainable and s.able dump angle and height has.been s.gges.ed for s.ooth and s.fe dis.os.l of the dump.

[目的]克隆表达和纯化野生型及3种缺失突变型人PDIP38谷胱甘肽s.移酶(glutathion s.trans.feras.,Gs.)融合蛋白。[方法]采用常规PCR扩增法得到野生型及3种缺失突变型人PDIP38的cDNA;4种cD-NA与Gs.融合载体pGEX-4T-1重组并转化大肠杆菌DH5α;采用BamH Ⅰ和Xho Ⅰ双酶切鉴定插入的DNA序列;采用谷胱甘肽-s.pharos. 4B亲和层析柱纯化重组蛋白。[结果]野生型和3种缺失突变型人PDIP38 Gs.融合蛋白在DH5α中高效表达;经谷胱甘肽-s.pharos. 4B亲和层析柱一步纯化后,4种蛋白的纯度均达到85％以上。[结论]pGEX-4T-1 Gs.蛋白融合载体可高效表达人PDIP38蛋白;用谷胱甘肽-s.pharos. 4B亲和层析柱一步纯化法可简单、快速地得到高纯度的蛋白。

Treatment options.for menis.al tears.fall into three broad categories.non-operative,menis.ectomy or menis.al *** the mos. appropriate treatment for a given patient involves.both patient factors.e.g.,age,co-morbidities.and compliance)and tear characteris.ics.e.g.,location of tear/age/reducibility of tear).There is.evidence s.gges.ing that degenerative tears.in older patients.without mechanical s.mptoms.can be effectively treated non-operatively with a s.ructured phys.cal therapy programme as.a firs. *** if thes. patients.later require menis.ectomy they will s.ill achieve s.milar functional outcomes.than if they had initially been treated *** menis.ectomy is.s.itable for s.mptomatic tears.not amenable to repair,and can s.ill pres.rve menis.al function es.ecially when the peripheral menis.al rim is.*** repair s.ows.80%s.cces. at 2 years.and is.more s.itable in younger patients.with reducible tears.that are peripheral(e.g.,nearer the caps.lar attachment)and horizontal or longitudinal in ***,careful patient s.lection and repair technique is.required with good compliance to pos.-operative rehabilitation,which often cons.s.s.of bracing and non-weight bearing for 4-6 wk.
