Insights into platinum-induced peripheral neuropathy–current perspective

Cancer is a global health problem that is often successfully addressed by therapy, with cancer survivors increasing in numbers and living longer world around. Although new cancer treatment options are continuously explored, platinum based chemotherapy agents remain in use due to their efficiency and availability. Unfortunately, all cancer therapies affect normal tissues as well as cancer, and more than 40 specific side effects of platinum based drugs documented so far decrease the quality of life of cancer survivors. Chemotherapy-induced peripheral neuropathy is a frequent side effects of platinum-based chemotherapy agents. This cluster of complications is often so debilitating that patients occasionally have to discontinue the therapy. Sensory neurons of dorsal root ganglia are at the core of chemotherapy-induced peripheral neuropathy symptoms. In these postmitotic cells, DNA damage caused by platinum chemotherapy interferes with normal functioning. Accumulation of DNA-platinum adducts correlates with neurotoxic severity and development of sensation of pain. While biochemistry of DNA-platinum adducts is the same in all cell types, molecular mechanisms affected by DNA-platinum adducts are different in cancer cells and non-dividing cells. This review aims to raise awareness about platinum associated chemotherapy-induced peripheral neuropathy as a medical problem that has remained unexplained for decades. We emphasize the complexity of this condition both from clinical and mechanistical point of view and focus on recent findings about chemotherapy-induced peripheral neuropathy in in vitro and in vivo model systems. Finally, we summarize current perspectives about clinical approaches for chemotherapy-induced peripheral neuropathy treatment.

Numerical investigation on the aerodynamic characteristics of high-speed train under turbulent crosswind

Increasing velocity combined with decreasing mass of modern highspeed trains poses a question about the influence of strong crosswinds on its aerodynamics. Strong crosswinds may affect the running stability of high speed trains via the amplified aerodynamic forces and moments. In this study, a simulation of turbulent crosswind flows over the leading and end cars of ICE2 highspeed train was performed at different yaw angles in static and moving ground case scenarios. Since the train aerodynamic problems are closely associated with the flows occurring around train, the flow around the train was considered as incompressible and was obtained by solving the incom pressible form of the unsteady Reynoldsaveraged Navier Stokes （RANS） equations combined with the realizable kepsilon turbulence model. Important aerodynamic coef ficients such as the side force and rolling moment coeffi cients were calculated for yaw angles ranging from 30° to 60° and compared with the results obtained from wind tunnel test. The dependence of the flow structure on yaw angle was also presented. The nature of the flow field and its structure depicted by contours of velocity magnitude and streamline patterns along the train＇s crosssection were presented for different yaw angles. In addition, the pressure coefficient around the circumference of the train at dif ferent locations along its length was computed for yaw angles of 30° and 60°, The computed aerodynamic coef ficient outcomes using the realizable kepsilon turbulencemodel were in good agreement with the wind tunnel data. Both the side force coefficient and rolling moment coeffi cients increase steadily with yaw angle till about 50° before starting to exhibit an asymptotic behavior. Contours of velocity magnitude were also computed at different cross sections of the train along its length for different yaw angles. Morphology versus DNA barcoding： two sides of the same coin. A case study of Ceutorhynchus erysimi and C. contractus identification

Genotyping of 2 well-known weevil species from the genus Ceutorhynchus （Coleoptera： Curculionidae） distributed in west Palearctic, C. erysimi and C. contractus, revealed phenotype versus genotype inconsistencies in a set of 56 specimens （25 C. erysimi and 31 C. contractus） collected from 25 locations in Serbia and Montenegro. An analysis of the mitochondrial cytochrome oxidase subunit I gene （CO1）, widely used as a barcoding region, and a nuclear gene, elongation factor-v （EF-1α）, revealed stable genetic divergence among these species. The average uncorrected pairwise distances for the CO1 and EF-1α genes were 3.8%, and 1.3%, respectively, indicating 2 genetically well-segregated species. However, the genetic data were not congruent with the phenotypic characteristics of the studied specimens. In the first place, C. erysimi genotypes were attached to specimens with phenotypic characteristics of C. contractus. Species-specific PCR-RFLP assays for the barcoding gene COIwere applied for the molecular identification of 101 additional specimens of both morphospecies （33 C. erysimi and 68 C. contractus） and were found to confirm this incongruity. The discrepancy between the genetic and morphological data raises the question of the accuracy of using a barcoding approach, as it may result in misleading conclusions about the taxonomic position of the studied organism. Additionally, the typological species concept shows considerable weakness when genetic data are not supported with phenotypic characteristics as in case of asymmetric introgression, which may cause certain problems, especially in applied studies such as biological control programs in which the biological properties of the studied organisms are the main focus.

Enhanced Osteogenesis of Nanosized Cobalt-substituted Hydroxyapatite

Hydroxyapatite （HAp） is an extensively studied material with known biocompatible and osteoconductive properties in bone tissue reconstruction. The improvement of the osteogenetic potential of HAp has been tested through modification of its structure, by replacing Ca2＋ ions with Co2＋ ions. In our study, we comparatively analyze the osteogenetic potential of the syn- thesized HAp and Co2＋-substituted HAp （HAp/Co） designed on the nano-scale with the aim of specifically stimulating osteo- genesis in vivo. We present a quantitative study of the microscopic organization and structure of the newly formed tissue in a bone defect after 12 weeks and 24 weeks. A quantitative analysis of the calcium, magnesium and phosphorus content in the defect and its close environment was used to determine the deposition of minerals after bone reconstruction. The defect recon- structed with HAp/Co nanoparticles （Co2＋ content 12 wt%） was filled with a new tissue matrix composed of dense collagen fibers containing centers of mineralization after 24 weeks. The mineral deposition rate was also higher when the defect was reconstructed with HAp/Co than when it was filled with pttre HAp. A histological analysis confirmed that the alveolar bone, in which osteoporosis-induced defects were repaired using HAp/Co nanoparticles, was recuperated.

Bioremediation perspective of historically contaminated sediment with polycyclic aromatic hydrocarbons

The purpose of the current study is to compare the influence of different aerobic conditions(biostimulation(BS), bioaugmentation(BA), and a combination of biostimulation and bioaugmentation(BB))on polycyclic aromatic hydrocarbons(PAH) degradation and compare the degraded amount with single step XAD-4 extraction as a new tool for bioavailability assessment for chronically contaminated sediment samples obtained from territory of Autonomous Province Vojvodina of Serbia(S1, S2, and S3). A great number of papers dealing with biodegradation of PAHs in spiked sediment or soil have been published,but to the authors' knowledge, a limited number of papers studied aged, historically polluted sediment and a sum of chosen U.S. Environmental Protection Agency(USEPA) PAHs. A significant reduction(up to67%) in PAH concentration was observed, while the percentage of reduction varied depending on the sediment sample and treatment used. BS treatment successfully stimulated growth of indigenous bacteria. Further, PAH-degrading strain Sphingomonas paucimobilis F8 inoculated in BA and BB treatment survived for up to 7 weeks after it was suppressed by unfavorable conditions or native *** amounts generally showed good correlation with results obtained from XAD-4 *** obtained in the current study represent a good start for standardizing a XAD-4 extraction technique as a simplified, easier, and lower cost method for bioavailability assessment.

Design and Optimization of a GaN GIT Based PFC Boost Converter

The advent of GaN power devices brings the possibility of pushing up frequencies of power electronics system beyond the capacity of conventional Si *** harvest the high frequency operation benefits of GaN technology,not only GaN devices should be used optimally by providing proper switching and conduction conditions to best accommodate parasitic elements inherent with the devices,other components should also be capable of facilitating high frequency *** this paper,PFC boost converter is used as a platform to harvest the benefits of GaN technology and,at the same time,to uncover the limitations in pushing power electronics systems up.A 600W PFC boost converter with a maximum switching frequency of 3MHz is designed and implemented,which reached an average efficiency of 98.5%.The results indicate that,when properly used,a GaN PFC boost converter could operate at a frequency that is beyond the reach of conventional Si *** well,with availability of GaN devices,high frequency operation of power electronics systems is no longer limited by the lack of switching devices but by practical issues *** frequency magnetic materials,high frequency controllers,etc.

This work is focused on immobilization of laccase from Myceliophthora thermophila expressed in Aspergillus oryzae(Novozym 51003?laccase)on amino modified fumed nano-silica(AFNS)and the possible use in ***,for the first time,factors affecting the immobilization of Novozym 51003?laccase on AFNS were investigated for defining the immobilization mechanism and optimizing the utilization of AFNS as support for laccase *** highest specific activity(13.1 IU·mg-1 proteins)was achieved at offered 160 mg per g of AFNS and for the same offered protein concentration the highest activity immobilization yield,reaching68.3%after the equilibrium time,at optimum pH 5.0,was *** immobilization occurs by adsorption as monolayer enzyme binding in 40 min,following pseudo-first-order *** possible use of obtained immobilized preparation was investigated in degradation of pesticide *** 24 h,lindane concentration was reduced to 56.8%of initial concentration and after seven repeated reuses it retained 70%of the original activity.
