Head geometric morphometrics of two Chagas disease vectors from Venezuela

Triatominae species are considered the main vectors of Chagas disease or American Trypanosomiasis. In Venezuela, the principal vectors are Rhodnius prolixus （Stal, 1959） and Triatoma maculata （Erichson, 1848）, which are belonged to the tribe Rhodniini and Triatomini, respectively. The head conformation and size development of these species can reflect ontogenetic changes which contribute with the vectors biology studies, as well to support of instars determination. The goal of the paper is to the application of geometric morphometric techniques for describing head conformation and size of instars of these species. We photographed 140 heads in R. prolixus： First instar （I： 16）, second instar （II： 17）, third instar （III： 18）, fourth instar （IV： 21）, fifth instar （V： 21）, adult female （F： 26） and adult males （M： 21）; in T. maculata heads of 136 specimens were photographed,I： 20, II： 17, III： 26, IV： 15, V： 19, F： 20 and M： 19. Landmark coordinate （x, y） configurations were registered and aligned by Generalized Procrustes analysis. Covariance analyses were implemented with proportions of re-classified groups and MANOVA. Statistical analyses of variance found not significant differences in head isometric size （Kruskal-Wallis） among IV and V instars in both species. The a posteriori re-classification was almost perfect in R. prolixus （82%） and T. maculata （86%）; the main head differences occurs in antenniferous tubercles, postocular and preocular. Our study using quantitative tools for describing the shape differences contributes to explain the morphology variability and development of Chagas disease vectors.

Germanium transport across supported liquid membrane with Cyanex 923: Mathematical modeling

A mathematical model was developed to monitor the facilitated transport of germanium(IV) from oxalic acid solutions through a flat sheet supported liquid membrane(FSSLM) containing four trialkylphosphine oxides(Cyanex 923). The FSSLM modeling was based on the extraction constant(Kext) calculated from the liquid-liquid extraction(LLX) modeling. The LLX model presented a reliable calculation of the extraction constant(Kex= 2.057×103 L/mol4). The FSSLM model was solved using Matlab■ software according to extraction constant, Fick’s law, and diffusional principles. The model predicts the overall mass transfer coefficient(Korg) to be 3.84 cm/s. Using this value, diffusion coefficients(Dm) for various Cyanex 923 concentrations of 0.126, 0.252, 0.378, 0.505, 0.631 and 0.757 mol/L are found to be 8.50×10^-4, 4.30×10^-4, 1.87×10^-4, 5.87×10^-5, 2.57×10^-5, 2.09×10^-5 cm2/s, respectively. The results show that the diffusion rate of the current study is approximately more than that of similar FSSLM systems containing Cyanex 923 used to transport various metals. The modeling values are in good agreement with the experimental data, showing the good reliability of the mathematical model.

AMP-activated kinase （AMPK）, a protein that regulates energy balance and metabolism, has recently been identified in boar spermatozoa where regulates key functional sperm processes essential for fertilization. This work＇s aims are AMPK identification, intracellular localization, and their role in human spermatozoa function. Semen was obtained from healthy human donors. Sperm AMPK and phospho-Thr172-AMPK were analyzed by Western blotting and indirect immunofluorescence. High- and low-quality sperm populations were separated by a 40%-80% density gradient. Human spermatozoa motility was evaluated by an Integrated Semen analysis System （ISAS） in the presence or absence of the AMPK inhibitor compound C （CC）. AMPK is localized along the human spermatozoa, at the entire acrosome, midpiece and tail with variable intensity, whereas its active form, phospho-Thr172-AMPK, shows a prominent staining at the acrosome and sperm tail with a weaker staining in the midpiece and the postacrosomal region. Interestingly, spermatozoa bearing an excess residual cytoplasm show strong AMPK staining in this subcellular compartment. Both AMPK and phospho-Thr172-AMPK human spermatozoa contents exhibit important individual variations. Moreover, active AMPK is predominant in the high motility sperm population, where shows a stronger intensity compared with the low motility sperm population. Inhibition of AMPK activity in human spermatozoa by CC treatment leads to a significant reduction in any sperm motility parameter analyzed： percent of motile sperm, sperm velocities, progressivity, and other motility coefficients. This work identifies and points out AMPK as a new molecular mechanism involved in human spermatozoa motility. Further AMPK implications in the clinical efficiency of assisted reproduction and in other reproductive areas need to be studied.

Increasing evidence of a link between inflammatory bowel disease(IBD) and adverse cardiovascular events has emerged during the last *** 2014,an important number of meta-analyses and cohort studies clarified the subtle dangerous liaisons between gut inflammation and cardiovascular *** evidence suggests that patients with IBD have a significantly increased risk of myocardial infarction,stroke,and cardiovascular mortality,especially during periods of IBD *** populations(e.g.,women,young patients) may have an even greater *** effective treatment of IBD is aimed at disease remission and seems to reduce cardiovascular risk in these patients.A beneficial effect was demonstrated for salicylates,but not for steroids or *** necrosis factor-α antagonists,which are highly effective in the reduction of inflammation and in the restoration of the digestive mucosa,lead to conflicting cardiovascular effects,as they seem to reduce the risk for ischemic heart disease but increase the rate of cerebrovascular *** supplemental treatment strategies that may reduce the atherothrombotic risk during periods of IBD activity should be explored.

Injection of bone marrow mesenchymal stem cells by intravenous or intraperitoneal routes is a viable alternative to spinal cord injury treatment in mice

In spite of advances in surgical care and rehabilitation, the consequences of spinal cord injury （SCI） are still challenging. Several experimental therapeutic strategies have been studied in the SCI field, and recent advances have led to the development of therapies that may act on the inhibitory microenvironment. Assorted lineages of stem cells are considered a good treatment for SCI. This study investigated the effect of systemic transplantation of mesenchymal stem cells （MSCs） in a compressive SCI model. Here we present results of the intraperitoneal route, which has not been used previously for MSC administration after compressive SCI. We used adult female C57BL/6 mice that underwent laminectomy at the T9 level, followed by spinal cord compression for 1 minute with a 30-g vascular clip. The animals were divided into five groups： sham （anesthesia and laminectomy but without compression injury induction）, MSC i.p. （intraperitoneal injection of 8×10^5 MSCs in 500 μL of DMEM at 7 days after SCI）, MSC i.v. （intravenous injection of 8 × 10^5 MSCs in 500μL of DMEM at 7 days after SCI）, DMEM i.p. （intraperitoneal injection of 500μL of DMEM at 7 days after SCI）, DMEM i.v. （intravenous injection of 500 μL of DMEM at 7 days after SCI）. The effects of MSCs transplantation in white matter sparing were analyzed by luxol fast blue staining. The number of preserved fibers was counted in semithin sections stained with toluidine blue and the presence of trophic factors was analyzed by immunohistochemistry. In addition, we analyzed the locomotor performance with Basso Mouse Scale and Global Mobility Test. Our results showed white matter preservation and a larger number of preserved fibers in the MSC groups than in the DMEM groups. Furthermore, the MSC groups had higher levels of trophic factors （brain-derived neurotrophic factor, nerve growth factor, neurotrophin-3 and neurotrophin-4） in the spinal cord and improved locomotor performance. Electrocardiographic measures of repolarization dispersion and their relationships with echocardiographic indices of ventricular remodeling and premature ventricular beats in hypertension

Objective To examine the relationship between Tpeak- Tend interval （Tpe） and Tpe/QT ratio with occurrence of ventricular premature beats （VPBs） and left ventricular remodeling in hypertension. Methods A total of 52 patients with mild to moderate essential hypertension were included, undergoing echocardiography and 24-hours Holter monitoring. Ventricular remodeling was assessed by left ventricular mass index （LVMI） using the Devereux formula and diastolic fimction by transmitral E and A wave velocities and E/A ratio. Tpe was measured in the precordial leads. The end of the T wave was set by the method of the tangent to the steepest descending slope of the T wave. Results Tpe and Tpe/QT in leads V2 （r = 0.33, P = 0.01; r = 0.27, P = 0.04 respectively） and V3 （r = 0.40, P = 0.002; r = 0.40, P = 0.003, respectively） correlated significantly with LVMI. A significant inverse relationship was observed between E/A ratio and QT （r = -0.33, P = 0.01）, Tpe in V3 （r = -0.39, P = 0.003） and Tpe/QT in V3 （r = -0.31, P = 0.02）. Tpe in V3, V5, mean Tpe and maximum Tpe with cut-offvalues of 60 ms, 59 ms, 62 ms and 71 ms, respectively, associated with the occurrence of ventricular premature beats. Conclusions The repolarization parameters Tpe interval and Tpe/QT ratio correlate with LVMI and indices of left ventricular diastolic function and show better predictive values than traditional parameters such as QT interval and QT dispersion. Lead V3 is the best lead for measuring Tpe and Tpe/QT. These ECG indices can therefore be used in clinical practice to monitor LV remodeling and predict occurrence of VPBs.
