Structural and nucleic acid binding properties of hepatitis delta virus small antigen

AIm To further characterize the structure and nucleic acid binding properties of the 195 amino acid small delta antigen, S-HDAg, a study was made of a truncated form of S-HDAg, comprising amino acids 61-195(?60HDAg), thus lacking the domain considered necessary for dimerization and higher order *** circular dichroism, and nuclear magnetic resonance experiments were used to assess the structure of ?60HDAg. Nucleic acid binding properties were investigated by gel retardation assays. RESULTS Results showed that the truncated ?60HDAg protein is intrinsically disordered but compact, whereas the RNA binding domain, comprising residues 94-146, adopts a dynamic helical conformation. We also found that ?60HDAg fails to multimerize but still contains nucleic acid binding activity, indicating that multimerization is not essential for nucleic acid binding. moreover, in agreement with what has been previously reported for full-length protein, no apparent specificity was found for the truncated protein regarding nucleic acid *** Taken together these results allowed concluding that ?60HDAg is intrinsically disordered but compact; ?60HDAg is not a multimer but is still capable of nucleic acid binding albeit without apparent specificity.

Tolerogenic dendritic cells （Dcs） are widely studied for their possible use in the treatment of inflammatory disorders, such as autoimmune diseases. One of the obstacles for the use of this cell-based therapy is the characterization of drugs that are able to modulate Dcs. We have previously shown that chloroquine （cQ）, an antimalarial agent, has the ability to modulate Dcs towards a tolerogenic phenotype.1 These tolerogenic Dcs are able to suppress the development of experimental autoimmune encephalomyelitis （EAE）, a T cell-driven mouse model of human multiple sclerosis. In addition, several studies have proposed that nitric oxide （NO） plays a major role in the dif- ferentiation of regulatory T cells （Tregs） and the suppression of Thl/Th17 cells.2＂3 However, little is known about the role of Dc-derived NO in the modulation of inflammatory auto- immune responses. Thus, we aimed to evaluate whether NO plays a role in the tolerogenic activity of cQ-treated Dcs （cQ- Dcs）. We found that cQ induces Dc production of NO and expression of indoleamine 2,3-dioxygenase （IDO）, as well as inducible nitric oxide syrtthase （iNOS）.

Venous thromboembolism(VTE) is an illness that has a potentially life-threatening condition that affects a large percentage of the global population. VTE with pulmonary embolism(PE) is the third leading cause of death after myocardial infarction and stroke. In the first three months after an acute PE, there is an estimated 15% mortality among submassive PE, and 68% mortality in massive PE. current guidelines suggest fibrinolytic therapy regarding the clinical severity, however some studies suggest a more aggressive treatment approach. This review will summarize the available endovascular treatments and the different techniques with its indications and outcomes.

major resections of bone tumors of the head and neck, owing to their small dimensions and their proximity to the organs in this region, cause important functional and aesthetic problems that significantly compromise patients’ quality of life. Therefore, therapeutic planning should always extend beyond the resection to include functional and aesthetic reconstruction, preferably immediately. microsurgical reconstruction represents a significant advance, but has not yet been perfected. Rapid prototyping (RP) comes as a new technology for the purpose of assisting the surgeon in the visual and tactile aspects of surgery, providing diagnostic accuracy and increasing the success of surgical planning. The authors demonstrate the technological advances in the manufacture of customized mandibular prostheses with the assistance of RP and practical applications of these methods.

The potential applications of neural stem cells (NSc) in the therapy of degenerative and traumatic diseases of the central nervous system (cNS) have aroused great scientific interest. NScs can be recovered from specific areas of the cNS from fetuses, embryos and from adult brain as well, and under appropriate culture conditions, may be induced to differentiate into the three major neural cell types, neurons, astrocytes and oligodendrocytes. The main neurogenic areas of mammals are the subventricular zone (SVZ) and the dentate gyrus (DG) of the hippocampus, yet other areas of neurogenic potential have been identified, including frontal and parietal cortices, hippocampus and lateral wall of the lateral ventricle. Neural stem cells and differentiated neural cells are usually identified by expression of specific markers, proteins that are expressed by different cell types and/or at different stages of differentiation. The main cell markers are nestin and Sox-2 for undifferentiated NScs, beta-III tubulin (B-III tubulin) for neurons, Galactocerebroside (Galc) for oligodendrocytes and Glial fibrillary acid protein (GFAP) for astrocytes. In general, the main neurogenic areas, the neurogenic potential of NSc and the morphological and biological characteristics of differentiated neural cells are strikingly similar among species, yet some differences have been reported. This article presents a brief review of NScs, neurogenic areas and techniques used for their identification and characterization in humans and experimental animals.

This review was based on a literature search of Pubm ed and Scielo databases using the keywords "quercetin,rutin,isoquercitrin,isoquercitin(IQ),quercetin-3-glucoside,bioavailability,flavonols and favonoids,and cancer" and combinations of all the *** collected relevant scientific publications from 1990 to 2015 about the absorption,bioavailability,chemoprevention activity,and treatment effects as well as the underlying anticancer mechanisms of *** are a group of polyphenolic compounds widely distributed throughout the plant *** subclass of flavonols receives special attention owing to their health *** main components of this class are quercetin,rutin,and IQ,which is a flavonoid and although mostly found as a glycoside,is an aglycone(lacks a glycoside side chain).This compound presents similar therapeutic profiles to quercetin but with superior bioavailability,resulting in increased efficacy compared to the aglycone *** has therapeutic applications owing to its wide range of pharmacological effects including antioxidant,antiproliferative,anti-inflammatory,anti-hypertensive,and *** protective effects of IQ in cancer may be due to actions on lipid *** addition,the antitumor effect of IQ and its underlying mechanism are related to interactions with Wnt signaling pathway,mixed-lineage protein kinase 3,mitogen-activated protein kinase,apoptotic pathways,as well proinflammatory protein *** review contributed to clarifying the mechanisms of absorption,metabolism,and actions of IQ and isoquercitrin in cancer.

Pharmaceutical services contribute to the rational use of medicines by ensuring the provision of appropriate, safe and effective pharmacotherapies. Thus, it is important to investigate whether the geographical distribution of pharmacies causes inequalities in access in countries like Brazil, where there is no federal law regulating the opening of new establishments (in contrast to many European countries). This study analyzed the accessibility of community pharmacies in the city of Sao mateus, Espírito Santo, Brazil. The authors obtained the geographical coordinates of pharmacies and healthfacilities, which scanned and georeferenced the study area. The 40 pharmacies included in the spatial analysis presented a heterogeneous distribution, with the existence of neighborhoods with an accumulation of pharmacies and others with no pharmacies. The authors found that only 43.7% of the study area received pharmaceutical service coverage, when buffer zones of 350 meters are incorporated, and that most of the districts have a minimum distance of 100 meters between health facilities and pharmacies. It was concluded that the use of GIS proved to be very suitable for analysis of pharmaceutical services, enabling knowledge of the local situation and serving as a stimulus for future studies.

Assessing cleaner Energy Alternatives for Bus Transit in Rio de Janeiro:A Life cycle Inventory analysis

From 2003 to 2009 in Brazilian municipalities of over 60,000 inhabitants, buses accounted for more than 25% of urban trips. This trend is not expected to change in the medium term. Worldwide, buses rely on petroleum diesel as fuel. In Brazil, alternative fuels such as biodiesel, natural gas and ethanol are available and the choice among them should depend on the assessment of the entire life cycle of such fuels. This paper uses a Life cycle Inventory, which is essential to the implementation of a Life cycle Assessment, to assess six energy alternatives: petroleum diesel, biodiesel, petroleum diesel with 5% of biodiesel, compressed natural gas, additivated hydrous ethanol and dual-fuel system composed by petroleum diesel with 5% of biodiesel and compressed natural gas. In saving total energy consumption, pure petroleum diesel or mixed with 5% biodiesel and dual-fuel systems stand out, in that order. If renewable energy use and net carbon dioxide emissions reduction are the goals, ethanol and biodiesel should be given preference. The addition of 5% of biodiesel in petroleum diesel increases the share of renewable energy in the supply chain of petroleum diesel by 47.5% with an increase of 0.58% in total energy consumption and a reduction of 3.8% in net cO2 emissions during the life cycle. In the case of biodiesel, the addition of 5% of biodiesel in petroleum diesel increases the share of renewable energy in the supply chain by 51.15% with an increase of 0.03% in the total energy consumption and a decrease of 7% in net cO2 emissions in the life cycle. The use of 5% of biodiesel in petroleum diesel does not significantly affect the use of renewable energy (+0.69%) or total energy consumption (+0.04%) in ethanol supply chain, which already shows a great use of renewable energy input. However, a decrease of 9.29% in the net cO2 emissions in the supply chain occurs, which reaches 5.28% in the life cycle.
