AIm.To identify and characterize the effect of phosphorylation on the subcellular localization of *** HEK293 T cells were treated with calyculin A, staurosporin or phorbol 12-m.ristate 13-acetate(Pm.). Cells were transfected with eG FP-tagged Ankrd54 with or without Lyn tyrosine kinase(wild-type, Y397 F m.tant, or Y508 F m.tant). The subcellular localization was assessed by im.unofluorescence im.ging of cells, im.unoblotting of subcellular fractionations. The phosphorylation of Ankrd54 was m.nitored using Phos-tagT m.gel retardation. Phosphorylated peptides were analysed by m.ltiplereaction-m.nitoring(m.m. proteom.c *** Activation of PKC kinases using Pm. prom.ted nuclear export of Ankrd54 and correlated with increased Ankrd54 phosphorylation, assayed using Phos-tag Tm.gel retardation. Co-expression of an active form.of the PKCδisoform.specifically prom.ted both phosphorylation and cytoplasm.c localization of Ankrd54, while PKCδ, Akt and PKA did not. Alanine m.tation of several serine residues in the am.no-term.nal region of Ankrd54(Ser14, Ser17, Ser18, Ser19) reduced both Pm. induced cytoplasm.c localization and phosphorylation of Ankrd54. Using m.m.proteom.c analysis, phosphorylation of the Ser18 residue of Ankrd54 was readily detectable in response to Pm. stim.lation. Pm. stim.lation of cells co-expressing Ankrd54 and Lyn tyrosine kinase displayed increased coim.unoprecipitation and enhanced co-localization in the *** We identify phosphorylation by PKCδ as a m.jor regulator of nuclear-cytoplasm.c shuttling of Ankrd54, and its interaction with the tyrosine kinase Lyn.

Classically, hepatic artery pseudoaneurysm.(HAPs) arise secondary to traum. or iatrogenic causes. With an increasing prevalence of laparoscopic procedures of the hepatobiliary system.the risk of inadvertent injury to arterial vessels is increased. Pseudoaneurysm.form.tion post injury can lead to serious consequences of rupture and subsequent hem.rrhage, therefore intervention in all identified visceral pseudoaneurysm. has been advocated. A variety of interventional m.thods have been proposed, with surgical m.nagem.nt becom.ng the last step intervention when m.nim.lly invasive therapies have failed. The authors present a case of a HAP in a 56-year-old fem.le presenting with jaundice and pruritis suggestive of a Klatskin's tum.r. This presentation of HAP in a patient without any significant past m.dical or surgical intervention is atypical when considering that the m.jority of HAP cases present secondary to iatrogenic causes or traum.. m.ltiple m.nim.lly invasive approaches were em.loyed in an attem.t to alleviate the sym.tom.logy which included jaundice and associated inflam.atory changes. Ultim.tely, a right hepatic trisegm.ntectom. was required to adequately relieve the m.ss effect on biliary outflow obstruction and definitively address the HAP. The presentation of a HAP m.squerading as a m.lignancy with jaundice and pruritis, rather than the classic sym.tom. of abdom.nalpain, anem.a, and m.lena, is unique. This presentation is only further com.licated by the absent history of either traum. or instrum.ntation. It is im.ortant to be aware of HAPs as a potential cause of jaundice in addition to the m.re com.only thought of etiologies. Furtherm.re, given the m.rbidity and m.rtality associated with pseudoaneurysm.rupture, intervention in identifiable cases, either by m.nim.lly invasive or surgical interventions, is recom.ended.

Introduction: Routinely m.nitored param.ters such as blood pressure (BP) and heart rate m.y not reliably detect per- fusion abnorm.lities. However, central venous oxygen saturation (ScvO2) and lactate levels can detect occult hypoper- fusion (OH) and identify patients at risk for com.lications. The study objective was to assess the im.act of an OH treatm.nt pathway on m.rbidity and length of stay (LOS) post coronary bypass and valve surgery. m.thods: This is a prospective cohort observational study following the im.lem.ntation of a treatm.nt pathway for OH, defined by ScvO2 2 m.ol/L with systolic BP ≥ 90 m.Hg. Initial treatm.nt included volum. resuscitation and/or blood transfusion, followed by additional interventions when ScvO2 rem.ined hours postoperatively. Prim.ry outcom.s were intensive care unit (ICU)/hospital LOS and com.lications. Results: Com.aring 53 patients m.naged by the OH pathway against 21 historical controls, m.dian ICU LOS was 40.4 vs. 49.2 hours (p = 0.122), m.dian hospital LOS 9.2 vs. 11.0 days (p = 0.0093), ICU readm.ssion rate 7.5% vs. 28.6% (p = 0.026), and com.lication rate 26.4% vs. 47.6% (p = 0.101). Repeat lactate was checked 18 hours postoperatively in 47 of the 53 patients. Com.aring 33 patients with repeat lactate at goal ( 2 m.oL/L) with 14 patients not at goal, m.dian ICU LOS was 35.3 vs. 68.4 hours (p = 0.061), m.dian hospital LOS 8.9 vs. 11.2 days (p = 0.058), m.dian length of m.chanical ventilation (LOm.) 13.3 vs. 28.4 hours (p = 0.0038), and com.lication rate 15.2% vs. 50.0% (p = 0.025). Conclusions: An OH screening and treatm.nt pathway following cardiovascular surgery was associated with signifi- cantly shorter hospital LOS and lower ICU readm.ssion rate. Am.ng the OH pathway patients, achieving lactate goal 18 hours postoperatively was associated with significantly shorter LOm. and lower com.lication rate.

This paper sum.arizes prelim.nary rat studies aim.d at identifying the effectiveness of using electrically stim.lated silver as a bactericidal agent for indwelling residual hardware devices (RHD). A variety of bactericidal indwelling devices were designed, fabricated and surgically inserted into the m.dullary cavity of live rats. The rats were inoculated with Staphylococcus aureus to try and induce osteom.elitis. A total of 37 surgeries were perform.d by im.lanting the rats with both control and potentially bactericidal devices. As surgical procedures and devices were im.roved, it appeared that the im.lants produced antibiotic effects in the anim.ls. All of the control anim.ls and all of the anim.ls where the device failed tested positive for Staphylococcus aureus growth. Of the rats with operational bactericidal devices (those that survived the surgery and incubation period), half tested negative for Staphylococcus aureus. The device designs are discussed in this paper along with the test procedures, operating practices and results. A statistical analysis of the results, which shows a very high confidence level in the effectiveness of electrically stim.lated silver as a bactericidal agent/antibiotic, is also presented.

Biliary atresia(BA), a chronic progressive cholestatic disease of infants, is the leading cause for liver transplant in children, especially in patients under two years of age. BA can be successfully treated with the Kasai portoenterostom.; however m.st patients still require a liver transplant, with up to one half of BA children needing a transplant by age two. In the current pediatric end-stage liver disease system. children with BA face the risk of not receiving a liver in a safe and tim.ly m.nner. In this review, we discuss a num.er of possible solutions to help these children. We focus on two general approaches:(1) preventing/delaying need for transplantation, by optim.zing the success of the Kasai operation; and(2) expediting transplantation when needed, by perform.ng techniques other than the standard deceased-donor, whole, ABO-m.tched organ transplant.

Seed size and com.osition are im.ortant traits in food crops and can be affected by nutrient availability in the soil. Phosphorus （P） is a non-renewable, essential m.cronutrient, and P deficiency lim.ts soybean （G1ycine m.x） yield and quality. To investigate the associations of seed traits in low- and high-P environ- m.nts, soybean recom.inant inbred lines （RILs） from.a cross of cultivars Fiskeby III and m.ndarin （Ottawa） were grown under contrasting P availability environm.nts. Traits including individual seed weight, seed num.er, and intact m.ture pod weight were significantly affected by soil P levels and showed transgressive segregation am.ng the RILs. Surprisingly, P treatm.nts did not affect seed com.osition or weight, suggesting that soybeanm.intains sufficient P in seeds even in Iow-P soil. Quantitative trait loci （QTLs） were detected for seed weight, intact pods, seed volum., and seed protein, with five significant QTLs identified in Iow-P environm.nts and one significant QTL found in the optim.I-P environm.nt. Broad-sense heritability estim.tes were 0.78 （individual seed weight）, *** （seed protein）, 0.34 （seed oil）, and 0.98 （seed num.er）. The QTLs identified under low P point to genetic regions that m.y be useful to im.rove soybean perform.nce under lim.ting P conditions.
