Cystic fibrosis (CF) is a genetically inherited disease which is characterized by excessive inflam.ation and inability to resolve infection with pathogens such as Pseudom.nas aeruginosa. Treatm.nt options have im.roved with correctors and potentiators, but a cure rem.ins elusive. Hum.n m.senchym.l stem.cells (hm.Cs) have the potential to be both anti-inflam.atory and anti-m.crobial, which m.kes them.ideal candidates for exploration as an innovative new therapeutic for CF. Using a sublethal CF m.use m.del of chronic Pseudom.nas aeruginosa infection, we show that hm.Cs and wild type bone m.rrow derived m.crophages (Bm.) have the capacity to attenuate inflam.ation while at the sam. tim. im.roving the ability to resolve infection. Anim.ls infected with bacteria and treated with hm.Cs and Bm. had less weight lost, decreased pro-inflam.atory cytokines, decreased severity of gross lung pathology as well as clinical score. Im.ortantly, even though the inflam.ation was decreased in vivo, both Bm. and hm.C treatm.nt resulted in significant decrease in lung bacterial load. The im.rovem.nt in the CF m.del was consistent with hm.C induced anti-inflam.atory and anti-m.crobial activity which m.y involve the cathelicidin LL-37. These studies suggest that both healthy m.Cs and Bm. m.y provide im.ortant new direction toward cell based therapies in CF.

目的利用生物信息学方法筛选湿性年龄相关性黄斑变性(wet age-related m.cular degeneration,w Am.)的氧化应激相关差异表达基因及靶向中药,以期为wAm.的预防和治疗提供新思路。方法从GEO数据库中筛选数据集,利用R软件lim.a包筛选差异基因,从Genecard数据库筛选氧化应激差异基因,并利用韦恩图得到共有基因。应用m.tascape对氧化应激差异基因进行基因本体(gene ontology,GO)和京都基因与基因组百科全书(Kyoto encyclopedia of genes and genom.s,KEGG)分析,并通过String数据库构建蛋白质相互作用(protein-protein interaction,PPI)网络,再通过Cytoscape软件筛选出wAm.中氧化应激的关键(Hub)基因。采用GSE103060数据集验证Hub基因的差异表达。利用Corem.ne m.dical预测靶向中药。结果经筛选共得到差异表达基因1874个,其中上调基因747个,下调基因1127个。KEGG富集分析表明,差异表达基因主要富集于细胞外基质受体相互作用、黏附斑激酶通路、河马(Hippo)信号通路、晚期糖基化终末产物-晚期糖基化终末产物受体(advanced glycation end products-receptor for advanced glycation end products,AGE-RAGE)信号通路、钙信号通路和磷脂酰肌醇-3-羟激酶(phosphatidylinositol-3-hydroxykinase,PI3K)/蛋白激酶B(protein kinase B,Akt)信号通路。通过PPI网络筛选得到与w Am.氧化应激密切相关的8个Hub基因,分别为IL6、EGFR、HIF1A、APOE、FN1、PPARG、EDN1和NGF。最后在GSE103060数据集中验证了以上Hub基因的显著差异表达。利用Corem.ne m.dical预测人参、黄芪、枸杞子、丹参、银杏叶等可作为预防或治疗wAm.的靶向中药。结论通过对w Am.芯片的挖掘和生物信息分析,差异基因主要富集于氧化应激的相关通路与wAm.的氧化应激相关基因引起的。防治wAm.的首选中药为人参、黄芪、枸杞子、丹参和银杏叶等26种。这将对理解和探索wAm.的中医药防治策略有重要意义。

Patients were random.zed 2:1 to receive either atezolizum.b(1,200 m.) or docetaxel(75 m./m.(2)). The prim.ry study endpoint was overall survival(OS) in the intention-to-treat(ITT) population with wild-type epiderm.l growth factor receptor expression(ITT EGFR-WT) and in the overall ITT ***: m.dian OS in the ITT EGFR-WT population(n=467) was 12.3 [95% confidence interval(95% CI),10.3-13.8] m.nths in the atezolizum.b arm.n=312) and 9.9(95% CI, 7.8-13.9) m.nths in the docetaxel arm.n=155;stratified hazard ratio(HR), 0.82;95% CI, 0.66-1.03]. m.dian OS in the overall ITT population was 12.5(95% CI, 10.8-13.8) m.nths with atezolizum.b treatm.nt and 11.1(95% CI, 8.4-14.2) m.nths(n=377) with docetaxel treatm.nt(n=188;stratified HR, 0.87;95% CI, 0.71-1.08). Understanding naturally occurring pine regeneration dynam.cs in response to thinning and burning treatm.nts is necessary not only to m.asure the longevity of the restoration or fuels treatm.nt,but also to assess how well regeneration m.ets forest sustainability guidelines and whether natural regeneration is sufficient for m.intaining a sustainable forest structure and com.osition.A synthesis review was carried out on the effects of m.chanical thinning and prescribed burn treatm.nts on natural pine regeneration response in frequent-fire ponderosa pine forests across the western United *** focus was on site-specific variability in pine regeneration dynam.cs,tem.oral trends in regeneration presence and abundance,and response to treatm.nt as described in the current literature using 29 studies that m.t our evidence-based review *** showed that the effects of thinning and burning treatm.nts on regeneration depended on tim. since *** thinning,prescribed burning,and thinning plus burn treatm.nts all increased seedling density,but there was high variability am.ng sites and *** were m.xed results in the short-term.m.studies(>20 years)concluded that stands can return to pre-treatm.nt densities in term. of total trees per hectare and forest floor duff levels when there are no m.intenance treatm.nts *** studies showed the average ponderosa pine seedling presence,survival and growth found in today’s forests to be at a high density;this com.ined with m.ssed fire cycles could contribute to future fire risk and reduce the efficacy of m.intaining fuel reduction goals.

Water disinfection is an essential process that provides safe water by inactivating pathogens that cause waterborne diseases. However, disinfectants react with organic m.tter naturally present in water, leading to the form.tion of disinfection by-products(DBPs). m.lti-analyte m.thods based on m.ss spectrom.try(m.) are preferred to quantify m.ltiple DBP classes at once however, m.st require extensive sam.le pre-treatm.nt and significant resources. In this study, two analytical m.thods were developed for the quantification of 32 regulated and unregulated DBPs. A purge and trap(P&T) coupled with gas chrom.tography m.ss spectrom.try(GC-m.) m.thod was optim.zed that autom.ted sam.le pre-treatm.nt and analyzed volatile and sem.-volatile com.ounds, including trihalom.thanes(THm.), iodinated trihalom.thanes(I-THm.), haloacetonitriles(HANs), haloketones(HKTs) and halonitrom.thanes(HNm.). LOQs were between 0.02-0.4 μg/L for m.st DBPs except for 8 analytes that were in the low μg/L range. A second m.thod with liquid chrom.tography(LC) tandem.m.ss spectrom.try(m./m.) was developed for the quantification of 10 haloacetic acids(HAAs) with a sim.le clean-up and direct injection. The LC-m./m. direct injection m.thod has the lowest detection lim.ts reported(0.2-0.5 μg/L). Both m.thods have a sim.le sam.le pre-treatm.nt, which m.ke it possible for routine analysis. Hyperchlorination and uniform.form.tion conditions(UFC) form.tion potential tests with chlorine were evaluated with water sam.les containing high and low TOC. Hyperchlorination form.tion potential test m.xim.zed THm. and HAAs while UFC m.xim.zed HANs. Ascorbic acid was found to be an appropriate quencher for both analytical m.thods. Disinfected drinking water from.four water utilities in Alberta, Canada were also evaluated.

The nest environm.nt m.y lim.t the ability of nest-bound birds to hear sounds from.the outside *** vocal learning species,such as hum.ns and songbirds,it is vital for young anim.ls to hear the voices of conspecific anim.ls early in *** songbirds,nest structure varies considerably across species,and the resulting im.act on sound transm.ssion m.y have consequences for vocal learning in *** this study,we tested the hypothesis that the nest environm.nt attenuates song of Savannah Sparrows(Passerculus sandwichensis),grassland songbirds that build nests on the ground under cover of dense *** recorded live Savannah Sparrows singing at variable distances from.21 *** recorded songs using one m.crophone inside the nest(the typical position of a nestling)and another placed 1 m.directly above the nest(a typical position of an adult).We found a substantial reduction in signal-to-noise ratio,where songs recorded inside the nest were an average of 11 dB lower than songs recorded directly above the *** estim.te that the attenuation im.osed by the nest reduced the m.xim.m.acoustic environm.nt from.117.7 m.for recordings above the nest),to 78.6 m.for recordings within the nest),which is analogous to listening from.a position 39.1 m.farther away from.the *** research estim.ted that song transm.ts up to 100 m.in this species,so any adult m.le within 100 m.of a young bird was previously considered a potential vocal *** reducing the nestling acoustic environm.nt from.100 m.to 78.6 m.the num.er of m.le tutors available to nestlings is reduced by an average of 27%.Given the growing evidence that song learning begins very early in life,future research on vocal developm.nt should account for how the structural properties of the natal environm.nt affect the songs that nestlings hear.

To protect from.COVID-19 pandem.c, several vaccines were developed infection with expected im.unity against a SARS-CoV-2 infection. Short tim. side effects are reported. New onset diabetes was reported after SARS-CoV-2 infection. Here we report a case of new onset diabetes presenting with hyperosm.lar hyperglycem.c state, whose sym.tom. followed right after the second dose of Pfizer-BioNTech COVID-19 Vaccine. He is a 56-year old, obese Afro-Am.rican Veteran with no fam.ly history of diabetes and with HbA1C of 5.6 forty-five days prior to the hospitalization. He noted polyurea and excessive thirst following the second dose Pfizer-BioNTech COVID-19 vaccine. Hospitalized with hyperosm.lar state and HbA1C of m.re than 14, he was treated initially with insulin drip and changed to basal, bolus regim.n. In addition, he had new onset of oral thrush, requiring antifungal therapy. He needed higher doses of insulin during hospitalization and at discharge. He rapidly recovered and could be tapered off insulin in 4 m.nths and recovered to norm.l glycem.c state. We conclude that this is the second state to present with hyperosm.lar state, and first case with rapid recovery of glycem.c state.

The im.ortance of predation risk as a key driver of evolutionary change is exem.lified by the Northern Range in Trinidad, where research on guppies living in m.ltiple parallel stream. has provided invaluable insights into the process of evolution by natural selection. Although Trinidadian guppies are now a textbook exam.le of evolution in action, studies have generally categorized predation as a dichotom.us variable, representing high or low risk. Yet, ecologists appreciate that com.unity structure and the attendant predation risk vary substantially over space and tim.. Here, we use data from.a longitudinal study of fish assem.lages at 16 different sites in the Northern Range to quantify tem.oral and spatial variation in predation risk. Specifically we ask: 1) Is there evidence for a gradient in predation risk? 2) Does the ranking of sites (by risk) change with the definition of the predator com.unity (in term. of species com.osition and abundance currency), and 3) Are site rankings consistent over tim.? We find com.elling evidence that sites lie along a continuum.of risk. However, site rankings along this gradient depend on how predation is quantified in term. of the species considered to be predators and the abundance currency is used. Nonetheless, for a given categorization and currency, rankings are relatively consistent over tim.. Our study suggests that consideration of predation gradients will lead to a m.re nuanced understanding of the role of predation risk in behavioral and evolutionary ecology. It also em.hasizes the need to justify and report the definition of predation risk being used.

The focus of this review is to highlight the im.ortance of glial cell line-derived neurotrophic factor （GDNF） for the m.tor nervous system. GDNF is the m.st potent survival factor for m.tor neurons, where it enhances m.intenance and survival of both developing and m.ture m.tor neurons in vivo and in vitro. GDNF aids in neurom.scular junction form.tion, m.intenance, and plasticity, where skeletal m.scle-derived GDNF m.y be responsible for this phenom.non. Increased levels of physical activity can increase GDNF protein levels in skeletal m.scle, where alterations in acetylcholine and acetylcholine receptor activation m.y be involved in regulation of these changes observed. With inactivity and disuse, GDNF expression shows different patterns of regulation in the central and peripheral nervous system.. Due to its potent effects for m.tor neurons, GDNF is being extensively studied in neurom.scular diseases.
