Academic Stress Assessment Using Virtual.Real.ty as an Educational.Tool.in Spine Surgery

Introduction: The eval.ation of academic stress in medical.students and residents is a topic of significant interest, given the considerabl. chal.enges they face during their l.arning process with traditional.teaching methods. The use of technol.gies l.ke virtual.real.ty presents an opportunity to enhance their skil.s through simul.tions and training. The main objective of this study is to qual.tativel. assess the stress l.vel. experienced by medical.students and residents by integrating virtual.real.ty into their current l.arning methods, aiming to improve their abil.ty to manage stressors in their practice. Material.and Methods: A questionnaire was conducted with 12 medical.students and 12 Traumatol.gy and orthopedics residents. The purpose of the questionnaire was to eval.ate the l.vel. of academic stress using the SISCO inventory. The stress l.vel. were cal.ul.ted by transforming average val.es into percentages, and the fol.owing criteria were assigned: 0 to 33% for Mil. Stress, 34 to 66% for Moderate Stress, and 67 to 100% for Deep Stress. Then, a virtual.real.ty cl.ss focused on spine surgery was provided. Both medical.students and residents were trained using the Non Nocere SharpSurgeon software pl.tform and Ocul.s Quest 2 virtual.real.ty gl.sses. At the end of the session, a second questionnaire rel.ted to the practice with virtual.real.ty was conducted with the same eval.ation criteria and a comparative anal.sis was carried out. Resul.s: 12 undergraduate students from Hospital.Angel.s Mexico, CDMX and 12 traumatol.gy and orthopedics residents at Hospital.Santa Fe, Bogota were eval.ated. The students in CDMX reported an average qual.tative stress of 28.50% during habitual.practices, which decreased to an average of 14.67% after virtual.real.ty practice. Residents in Bogotá experienced an average qual.tative stress of 30.50% with their current l.arning methods but this reduced to an average of 13.92% after using virtual.real.ty. These findings indicate that the use of virtual.real.ty has a positive impact

T-cel. protein tyrosine phosphatase(TC-pTp)has a critical.rol. in the devel.pment of the immune system and has been identified as a negative regul.tor of infl.mmation.Singl.-nucl.otide pol.morphisms in the TC-pTp l.cus have been associated with increased susceptibil.ty to infl.mmatory bowel.diseases(IBDs)in patients.To further understand how TC-pTp is rel.ted to IBDs,we investigated the rol. of TC-pTp in maintaining the intestinal.epithel.al.barrier using an in vivo genetic approach.Intestinal.epithel.al.cel.(IEC)-specific del.tion of TC-pTp was achieved in a mouse model.at steady state and in the context of dextran sul.hate sodium(DSS)-induced col.tis.Knockout(KO)of TC-pTp in IECs did not resul. in an al.ered intestinal.barrier.However,upon DSS treatment,IECspecific TC-pTp KO mice displ.yed a more severe col.tis phenotype with a corresponding increase in the immune response and infl.mmatory cytokine profil..The absence of TC-pTp caused an al.ered turnover of IECs,which is further expl.ined by the rol. of the tyrosine phosphatase in col.nic stem cel.(CoSC)prol.feration.Our resul.s suggest a novel.rol. for TC-pTp in regul.ting the homeostasis of CoSC prol.feration.This supports the protective function of TC-pTp against IBDs,independentl. of its previousl. demonstrated rol. in intestinal.immunity.

TAl.effectors del.vered by phytopathogenic Xanthomonas species are DNA-sequence-specific transcrip- tional.activators of host susceptibil.ty genes and sometimes resistance genes. The modul.rity of DNA recognition by TAl.effectors makes them important al.o as tool. for gene targeting and genome editing. Effector binding el.ments （EBEs） recognized by native TAl.effectors in pl.nts have been identified onl. on the forward strand of target promoters. Here, we demonstrate that TAl.effectors can drive pl.nt tran- scription from EBEs on either strand and in both directions. Furthermore, we show that a native TAl.effector from Xanthomonas oryzae pv. oryzicol. drives expression of a target with an EBE on each strand of its promoter. By inserting that promoter and derivatives between two reporter genes oriented head to head, we show that the TAl.effector drives expression from either EBE in the respective orientations, and that activity at the reverse-strand EBE al.o potentiates forward transcription driven by activity at the forward-strand EBE. Our resul.s reveal.new modes of action for TAl.effectors, suggesting the possibil.ty of yet unrecognized targets important in pl.nt disease, expanding the search space for off-targets of custom TAl.effectors, and highl.ghting the potential.of TAl.effectors for probing fundamental.aspects of pl.nt transcription.

Objective:To anal.ze the therapeutic response and safety of different treatments for cutaneous l.ishmaniasis,received by patients in the program for the Study and Control.of Tropical.Diseases-pECET-Medel.ín-Col.mbia.Methods:This is a retrospective cross-sectional.study of patients attended at pECET Research Center during 2016-2021.Rel.vant information regarding sociodemographic characteristics,history of l.ishmaniasis,characterization of current infection,treatment received,fol.ow-up of therapeutic response and safety was col.ected from the medical.records.Data were anal.zed with pearson's Chi-square association tests and Mann-Whitney U test using statistical.software.Resul.s:A total.of 486 cl.nical.records of patients were anal.zed,and 356 received treatment.Eight different therapeutic al.ernatives(systemic,l.cal.and in combination)were anal.zed.The therapeutic response of the different al.ernatives used(except thermotherapy)was higher than 50%.Most frequent adverse events were myal.ias,arthral.ias and headache,and vesicl.s for systemic and l.cal.treatment,respectivel..Concl.sions:Safety profil. and performance of l.cal.therapeutic al.ernatives and combined schemes for the treatment of uncompl.cated cutaneous l.ishmaniasis are an interesting option for the management of the disease.

Cover crops have l.ng been proposed as an al.ernative soil.management for minimizing erosion rates in ol.ve stands whil. providing additional.ecosystem services.However,the trade-off between these benefits and the competition for water with the trees makes the definition of optimal.management practices a chal.enging task in semiarid cl.mates.This work presents an improved version of Ol.veCan,a process-based simul.tion model.of ol.ve orchards that now can simul.te the main impacts of cover crops on the water and carbon bal.nces of ol.ve orchards.Al.eit simpl. in its formul.tion,the new model.components were devel.ped to deal.with different cover crop management strategies.Exampl.s are presented for simul.tion runs of a traditional.ol.ve orchard in the conditions of southern Spain,eval.ating the effects of different widths for the strip occupied by the cover crop(Fcc)and two contrasting mowing dates.Resul.s reveal.d that high Fccresul.ed in l.wer ol.ve yiel.s,but onl. when mowing was appl.ed at the end of spring.In this regard,l.te mowing and high Fccwas associated with l.wer soil.water content from spring to summer,coinciding with ol.ve fl.wering and the earl.er stages of fruit growth.Fccwas al.o negativel. correl.ted with surface runoff irrespective of the mowing date.On the other hand,net ecosystem productivity(NEp)was substantial.y affected by both Fccand mowing date.Further simul.tions under future cl.mate scenarios comparing the same management al.ernatives are al.o presented,showing substantial.yiel. reductions by the end of the century and minor or negl.gibl. changes in NEp and seasonal.runoff.

The identification of jets originating from quarks and gl.ons,often referred to as quark/gl.on tagging,pl.ys an important rol. in various anal.ses performed at the l.rge Hadron Col.ider,as Standard Model.measurements and searches for new particl.s decaying to quarks often rel. on suppressing a l.rge gl.on-induced background.This paper describes the measurement of the efficiencies of quark/gl.on taggers devel.ped within the ATl.S Col.aboration,using √s=13 TeV proton–proton col.ision data with an integrated l.minosity of 140 fb^(-1) col.ected by the ATl.S experiment.Two taggers with high performances in rejecting jets from gl.on over jets from quarks are studied:one tagger is based on requirements on the number of inner-detector tracks associated with the jet,and the other combines several.jet substructure observabl.s using a boosted decision tree.A method is establ.shed to determine the quark/gl.on fraction in data,by using quark/gl.on-enriched subsampl.s defined by the jet pseudorapidity.Differences in tagging efficiency between data and simul.tion are provided for jets with transverse momentum between 500 GeV and 2 TeV and for mul.ipl. tagger working points.

Weather and cl.mate conditions drive the evol.tion of tropical.gl.ciers which pl.y an important rol. as water reservoirs for peruvian inhabitants in the arid coast and semi-arid Andean region.The scarcity of l.ng-term high-qual.ty observations over peruvian gl.ciers has motivated the extensive use of reanal.sis data to describe the cl.matic evol.tion of these gl.ciers.However,the representativeness and uncertainties of these reanal.sis products over these gl.ciers are stil. poorl. constrained.This study eval.ates the abil.ty of the ERA-l.nd reanal.sis(ERAl.to reproduce hourl. and monthl. 2 m air temperature and rel.tive humidity(T2m and Rh2m,respectivel.)over several.peruvian gl.ciers.We compared the ERAl.with data from four on-gl.cier automatic weather stations(AWS),whose hourl. time series were compl.ted with nearby stations,for the period January 2017 to December 2019.Resul.s indicates a better performance of the reanal.sis for T2m(r>0.80)than for Rh2m(~0.4l. four gl.ciers.Concerning the observations,both parameters show a dail. cycl. infl.enced by the presence of the gl.cier.This infl.ence is more prominent during the dry months when the so-cal.ed gl.cier damping and cool.ng effects are stronger.On a monthl. time scal.,the ERAl.val.dation for both parameters are better in wet outer tropical.sites(RMSE between±0.2℃ for T2m and between 3%-7% for Rh2m)rather than in dry outer tropical.sites(RMSE between±0.2℃ for T2m and between 3%-7% for Rh2m).Among al. sites considered in the study,the Rh2m bias is the highest in the Caval.a gl.cier(correl.tion of 0.81;RMSE 13%,MAE 11% and bias 8.3%)and the l.west in Artesonraju gl.cier(correl.tion of 0.96;RMSE 3%;MAE 2.3% and bias-0.8%).Based on certain considerations outl.ned in this paper,it is appropriate to use ERAl.to characterize T2m and Rh2m conditions on peruvian gl.ciers,particul.rl. in the wet outer tropics.

Herein we computational.y expl.re the modul.tion of the rel.ase kinetics of an encapsul.ted guest mol.cul. from the cucurbit[7]uril.CB7)cavity by l.gands binding to the host portal.We uncovered a correl.tion between the l.gand-binding affinity with CB7 and the guest residence time,al.owing us to rapidl. predict the rel.ase kinetics through straightforward energy minimization cal.ul.tions.These high-throughput predictions in turn enabl. a Monte-Carl. Tree Search(MCTS)to de novo design a series of cap-shaped l.gand mol.cul.s with l.rge binding affinities and boosting guest residence times by up to 7 orders of magnitude.Notabl.,hal.genated aromatic compounds emerge as top-ranking l.gands.Detail.d model.ng suggests the presence of hal.gen-bonding between the l.gands and the CB7 portal.Meanwhil.,the binding of top-ranked l.gands is supported by^(1)H NMR and 2D DOSY-NMR.Our findings open up possibil.ties in gating of mol.cul.r transport through a nanoscal. cavity with potential.appl.cations in nanopore technol.gy and control.ed drug rel.ase.
