Advancing Early Detection of Colorectal Adenomatous Polyps via Genetic Data Analysis: A Hybrid Machine Learning Approach

In this study, a hybrid machine learning (HML)-based approach, incorporating Genetic data analysis (GDA), is proposed to accurately identify the presence of adenomatous colorectal polyps (ACRP) which is a crucial early detector of colorectal cancer (CRC). The present study develops a classification ensemble model based on tuned hyperparameters. Surpassing accuracy percentages of early detection approaches used in previous studies, the current method exhibits exceptional performance in identifying ACRP and diagnosing CRC, overcoming limitations of CRC traditional methods that are based on error-prone manual examination. Particularly, the method demonstrates the following CRP identification accuracy data: 97.7 ± 1.1, precision: 94.3 ± 5, recall: 96.0 ± 3, F1-score: 95.7 ± 4, specificity: 97.3 ± 1.2, average AUC: 0.97.3 ± 0.02, and average p-value: 0.0425 ± 0.07. The findings underscore the potential of this method for early detection of ACRP as well as clinical use in the development of CRC treatment planning strategies. The advantages of this approach are highly expected to contribute to the prevention and reduction of CRC mortality.

Dapoxetine is a newer anti-depressant that is used for severe forms of depression via selective serotonin reuptake inhibition *** aimed to evaluate the effect of different doses of dapoxetine lonely or with acetaminophen(AMP).Seven groups of albino rats were used where dapoxetine was administered orally at the doses of 3,5,and 8 mg/kg body weight for a period of 14 days in groups 2,4,and 6,***(67.5 mg/kg body weight)was co-administered with dapoxetine(3,5,and 8 mg/kg body weight)for 14 days in groups 3,5 and 7,*** in mid and high dose plus AMP increased body weight and hematological *** and rearing were declined with mid and high doses of dapoxetine+AMP,contrary to *** was decreased with high doses of dapoxetine+*** dose of dapoxetine+AMP induced lowering of serotonin and dopamine as well as increasing of *** derived neurotrophic factor was decreased in all *** stress biomarkers were augmented with high dose of dapoxetine+*** doses of dapoxetine and/or AMP induced loss of the Purkinje cell layer at *** high dose of dapoxetine+AMP denotes behavioral,histopathological and neurochemical alterations,even though no serotonin syndrome occurs.

AIM: To assess the diagnostic accuracy,of aminotransferase-to-platelet ratio index(APRI) alone and with antischistosomal antibody(Ab) in patients with hepatitis C virus(HCV) and schistosomiasis coinfection. METHODS: This retrospective study included medical records of three hundred and eighty three Egyptianmen patients who had undergone percutaneous liver biopsy between January 2006 to April 2014 in tertiary care hospital in Qatar for diagnosis or monitoring purpose were selected. Data of patients > 18 years of age were included in the study. The values of HCV RNA titer and antischistosomal antibody titer were also taken into consideration. Patients were excluded from the study if they had any other concomitant chronic liver disease,including; history of previous antiviral or interferon therapy,immunosuppressive,therapy,chronic hepatitis B infection,human immunodeficiency virus co-infection,autoimmune hepatitis,decompensated liver disease,hepatocellular carcinoma,prior liver transplantation,and if no data about the liver biopsy present. RESULTS: Median age of patients was 46 years. About 7.1% had no fibrosis,whereas 30.4%,37.5%,20.4%,and 4.6% had fibrosis of stage Ⅰ,Ⅱ,Ⅲ,and Ⅳ respectively. In bivariate analysis,APRI score,levels of AST,platelet count and age of patient showed statistically significant association with liver fibrosis(P < 0.0001); whereas antischistosomal antibody titer(P = 0.52) and HCV RNA titer(P = 0.79) failed to show a significant association. The respective AUC values for no fibrosis,significant fibrosis,severe fibrosis and cirrhosis of APRI score were 63%,73.2%,81.1% and 88.9% respectively. This showed good sensitivity and specificity of APRI alone for grading of liver fibrosis. But the inclusion of anti-Schistosoma antibody did not improve the prediction of fibrosis stage. CONCLUSION: The study results suggest that noninvasive biochemical markers like APRI are sensitive and specific in diagnosing the degree of fibrosis and cirrhosis in patient

Metformin alleviates LTA-induced inflammatory response through PPARγ/MAPK/NF-κB signaling pathway in bovine mammary epithelial cells

Mastitis is a common inflammatory cow mammary infection;that causes significant economic loss in dairy *** the interesting connection between metformin’s anti-inflammatory function and mastitis model induced by LTA in pbMECs,our objective was to prove that metformin was beneficial in suppressing proinflammatory response induced by LTA through modulation of mitogen-activated protein kinase(MAPK)and nuclear factor kappa B(NF-κB)signaling pathways and activation of peroxisome proliferator-activated receptor-γ(PPARγ)in *** proliferation of cells and mRNA expression were measured using EdU assay and quantitative reverse transcriptase-polymerase chain reaction(qRT-PCR).Immunoblotting and immunofluorescence analysis were conducted to evaluate the expression of target proteins in inflammatory and anti-inflammatory responses to metformin and ***,pbMECs were allowed to treat with the PPAR antagonist GW9662,and inflammatory markers were detected in the *** results showed that LTA concentration at 100μg/mL significantly stimulated the MAPK14,IL-6 and IL-1βmRNA expressions compared to the control cells(P<0.05)in dose-dependent tests for *** suppressed the phosphorylation expressions of MAPK(ERK1/2,p38,and JNK)in LTA-stimulated *** also reduced the protein expression of NF-κB,interleukin-8(IL-8),interleukin-1β(IL-1β)and interleukin-6(IL-6)in pbMECs pretreated with *** administration activated PPARγphosphorylation by up-regulating the expression of PPARγin LTA-stimulated *** with GW9662 resulted in increased IL-6 expression,which was reversed by *** findings collectively indicated that metformin act to attenuate LTA-stimulated inflammatory response in pbMECs by suppressing MAPK and NF-κB activation via a mechanism partially dependent on PPARγ*** results suggested that metformin could function as an anti-inflammatory drug in the treatment of mastitis.

Streptococcus peumoniae in an Egyptian urban community:incidence of erythromycin-resistance determinants and antibiotic susceptibility profile

Objectives:To determine the incidence of resistance of Streptococcus(Strep).pneumoniae isolated in our locality to erythromycin,to screen for the two resistance determinants erm(B) and mef(A) genes,and to identify the susceptibility profile to commonly used ***:Samples were collected from patients attending the Outpatient Department of Zagazig University Hospital,Zagazig,Egypt,between February 2006 and March *** was identified by conventional *** to erythromycin and 15 antibiotics were identified by disc diffusion method,as outlined by CLSI.E-test was used for MIC determination of ***(B) and mef(A) genes were detected by ***:Eighty-one Strep. pneumoniae strains were ***- one of them(63%) were erythromycin-resistant,and mef(A) gene was the predominant resistance ***,imipenem and gatifloxacin had the best activity against the isolates,whereas tetracycline had the ***-two(51.85%) out of the 81 *** strains were ***:High incidence of resistance to erythromycin and multiple antimicrobials ***(A) was the principal erythromycin-resistance gene.

Cardioprotective and antioxidant effects of oleogum resin "Olibanum" from Boswellia carteri Birdw.(Bursearceae)

One of the leading causes of death worldwide is cardiovascular disease, hence searching for a cure is an important endeavor. The totally safe, edible, and inexpensive Boswellia plant exudate, known as olibanum or frankincense, is considered to possess diverse medicinal values in traditional medicine and from recent biological studies. Investigating the cardioprotective and antioxidant activities of olibanum from a Boswellia species, family Bursearaceae, namely Boswellia carteri Birdw. was the aim of this study. Cardioprotective activity was evaluated using a model of myocardial infarction induced by isoprenaline(ISO), while antioxidant activity was tested adopting nitric oxide scavenging(NOS) and azino-bis-3-ethyl benzthiazoline-6-sulfonic acid(ABTS) assays. The results revealed a mild cardioprotective effect and weak antioxidant activity.

Semen parameters in men with spinal cord injury： changes and aetiology

Aim： To assess the changes in semen parameters in men with spinal cord injury （SCI） and the possible causes of these changes. Methods： The study included 45 subjects with SCI. Semen retrieval was done by masturbation （2）, vigorous prostatic massage （n = 13）, penile vibratory stimulation （n = 13） or electroejaculation （n = 17）. Results： The semen of men with SCI showed normal volume （2.3 ± 1.9 mL） and sperm count （85.0 × 10^6 ± 83.8 × 10^6/mE） with decreased motility （11.6% ± 10.1%）, vitality （18.5 % ± 15.2%） and normal forms （17.5 ± 13.4%）, and pus cells has been increased （6.0 × 10^6 ± 8.2 × 10^6/mL）. Total （13.4 ± 9.9 vs. 7.1 ± 6.8） and progressive （4.4 ± 3.9 vs. 2.2 ± 2.1） motility were significantly higher in subjects with lower scrotal temperatures. There was no statistical significant difference between electroejaculation and penile vibratory stimulation groups as regards any of the semen parameters. Subjects＇ age, infrequent ejaculation, injury duration and hormonal profile showed no significant effect on semen parameters. Conclusion： The defining characteristics of the seminogram in men with SCI are normal volume and count with decreased sperm motility, vitality and normal forms, and the increased number of pus cells. The most acceptable cause of the deterioration of semen is elevated scrotal temperature.

Ghrelin attenuates gastrointestinal epithelial damage induced by doxorubicin

AIM:To examine the influence of ghrelin on the regenerative potential of gastrointestinal(GI)***:Damage to GI epithelium was induced in mice by two intravenous injections of doxorubicin(10 and 6 mg/kg).Some of the doxorubicin-treated mice received a continuous subcutaneous infusion of ghrelin(1.25μg/h)for 10 d via implanted mini-osmotic *** label dividing stem cells in the S-phase of the cell cycle,all mice received a single intraperitoneal injection of 5'-bromo-2'-deoxyuridine(BrdU)one hour before *** stomach along with the duodenum were then removed and processed for histological examination and immunohistochemistry using anti-BrdU ***:The results showed dramatic damage to the GI epithelium 3 d after administration of chemotherapy which began to recover by day *** ghrelintreated mice,attenuation of GI mucosal damage was evident in the tissues examined *** analysis showed an increase in the number of BrdUlabeled cells and an alteration in their distribution along the epithelial lining in response to damage by *** mice treated with both doxorubicin and ghrelin,the number of BrdUlabeled cells was reduced when compared with mice treated with doxorubicin ***:The present study suggests that ghrelin enhances the regenerative potential of the GI epithelium in doxorubicintreated mice,at least in part,by modulating cell proliferation.
