Melanins represent one of the most ancient and important group of natural macromolecular *** have multiple biological roles in almost all organisms across the Phyla,examples being photoprotection,anti-oxidative action,radical scavenger activity,and heavy metal *** the biomedical point of view,melanocytes are involved in the origin of melanoma tumors,and the main therapeutic advances for their treatment have been revised in Part 1 of this *** chemical structure of eumelanin is a biological concern of great importance,and therefore,exploring theoretical molecular models and synthesis mechanisms will be here described,as well as molecular orbital features and supramolecular organization,which are responsible for the key properties that make these biological pigments so important,and so ***,this updated overview is devoted to describe present structural models and physico-chemical characteristics of eumelanin,in order to explain and utilize melanin properties on which new photothermal and ultrasonic protocols for melanoma treatment can be devised and applied.

Numerical Modelling of the Topographic Wetness Index: An Analysis at Different Scales

A variety of landscape properties have been modeled successfully using topographic indices such as topographic wetness index (TWI), defined as ln(a/tanβ), where a is the specific upslope area and β is the surface slope. In this study, 25 m spatial resolution from digital elevation models (DEM) data were used to investigate the scale-dependency of TWI values when converting DEMs to 50 and 100 m. To investigate the impact of different spatial resolution, the two lower resolution DEMs were interpolated to the original 25 m grid size. In addition, to compare different flow-direction algorithms, a second objective was to evaluate differences in spatial patterns. Thus the values of TWI were compared in two different ways: 1) distribution functions and their statistics;and 2) cell by cell comparison of DEMs with the same spatial resolution but different flow- directions. As in previous TWI studies, the computed specific upstream is smaller, on average, at higher resolution. TWI variation decreased with increasing grid size. A cell by cell comparison of the TWI values of the 50 and 100 m DEMs showed a low correlation with the TWI based on the 25 m DEM. The results showed significant differences between different flow-diretction algorithms computed for DEMs with 25, 50 and 100 m spatial resolution.

NATURAL AND ANTHROPOGENIC FACTORS CONTROLLING GULLY EROSION IN THEBASALTIC UPLAND OF SOUTHERN BRAZIL

This paper describes an analysis of natural and anthropogenic factors controlling the evolution of gullies in a rural basin in the basaltic upland in the State of Rio Grande do Sul, Southern Brazil. In this region of deep ferrallitic soils with more than 60% clay, runoff and erosion are of increasing concern. In the Tabo?o drainage basin (100 km2), gully erosion was studied in a field survey that measured rills and gullies. Eighty-four gullies were identified. They had an average length of 136 m, were 10 m wide, and 3 m deep and had a volume of 15.458 m3. Each gully was characterised in terms of factors that included slope, geological structure, presence of piping, drainage, soil use, and the presence of surface and subsurface flow. On average, the main channels had knickpoints varying from 2 m to 7 m, and their evolution in the vertical plane increased until bed-rock basalt material was reached, after which gullies increase in width and length. Gully development was also monitored from 1991 to 2003. Subsurface flow appears to be the principal agent controlling their development. Results show that both natural (slope, surface curvature, geological structure and rainfall) and anthropogenic (soil use, road construction) factors are important in gully development. The change in cultural practices throughout the drainage basin from conventional to direct seeding has led to increased subsurface flow, which was more important than surface runoff in causing erosion. However, the higher rainfall during El Ni?o Southern Oscillation (ENSO) events and the consequently higher subsurface flow were the dominant factors. From 1991 to 2003 a total land loss of 1,013 m3 was observed in one gully, with 236 m3 lost during the 1992 ENSO and 702 m3 during the 1997 ENSO; 95% of the total volume lost occurred during ENSO periods.

The vascular fusariosis is one of the main obstacles in the tomato crop production. Currently, the management of the nutrient solution is presented as a control option for fusariosis, for such reason different nutrient solutions with nitrogen (N), phosphorus (P), potassium (K) and calcium (Ca) modifications;in four saladette tomato varieties Bony Best (BB), Manapal (M), Walter (W) and FLA were evaluated in this research, in order to analyze the damage caused by Fusarium oxysporum ***. lycopersici (FOL) severity. Tomato plants were inoculated with 3 FOL breeds, establishing a completely random design with a factorial arrangement with six repetitions. Disease’s severity was evaluated 30 days after the inoculation using levels of (0 - 4) of the methodology established by Marlatt and Correll (1988). Afterwards, a statistical analysis was done through the Kruskal Wallis test, in which it was observed that treatments 93 (Ca 207 mg∙L-1, R3, variety BB), 95 (207 mg∙L-1 Ca, R3, variety W), 81 (0Ca, R3, BB) showed less disease severity. In the foliar analysis, the best treatments were 22 (193 mg∙L-1 N, R3, M) for N, the greatest P content was treatment 94 (207 mg∙L-1 Ca, R3, M) for K treatment three and 93 (207 mg∙L-1 Ca, R3, variety BB) greatest Ca content.

Objective:To characterize the Entamoeba histolytica(***)antigen(s)recognized by moribound amoebic liver abscess ***:Crude soluble antigen of *** was probed with sera of moribund hamsters in 1D-and 2D-Westem blot *** antigenic protein was then sent for tandem mass spectrometry *** corresponding gene was cloned and expressed in Escherichia coli bl21-AI to produce the recombinant *** ADP-forming acetyl-CoA synthetase(EhACS)protein.A customised ELISA was developed to evaluate the sensitivity and specificity of the recombinant ***:A^75 kDa protein band with a pl value of 5.91-6.5 was found to be antigenic;and not detected by sera of hamsters in the control *** mass spectrometry analysis revealed the protein to be the 77 kDa *** ADP-forming acetyl-CoA synthetase(EhACS).The customised ELISA results revealed 100%sensitivity and 100%specificity when tested against infected(n=31)and control group hamsters(n=5)serum samples,***:This rinding suggested the significant role of EhACS as a biomarker for moribund hamsters with acute amoebic liver abscess(ALA)*** is deemed pertinent that future studies explore the potential roles of EhACS in better understanding the pathogenesis of ALA;and in the development of vaccine and diagnostic tests to control ALA in human populations.

Galectin-3 expression favors metastasis in murine melanoma

Galectin-3 is a member of the lectin family that binds β-galactosides and plays an important role in several types of tumors. Melanoma is an invasive cancer responsible for 80% of deaths associated to skin cancers. There are some evidences that galectin-3 interacts with β-catenin, a molecule involved with Wnt signaling pathway. Here, we evaluate the role of galectin-3 intumor growth and metastasis, as well as its interaction with β-catenin. Murine melanoma cells (B16F10) were injected subcutaneously and intravenously in male C57bl/6 wild-type (WT) and galectin-3 knock-out (KO) mice. Tumor growth and lung melanoma colonies were assessed. The expression of galectin-3 and β-catenin was evaluated by immuno-histochemistry. We observed that tumor growth did not differ between the groups. However, to metastasis, the number of lung colonies in WT mice was significantly increased in comparison to that observed in KO mice. The cytoplasm expression of galectin-3 was observed in subcutaneous and metastatic tumors,w in both groups. We observed its nuclear expression in some of subcutaneous tumors of KO mice. The expression of β-catenin was detected in cell membrane of all subcutaneous tumors analyzed, whereas in the metastatic tumors we observed both cytoplasm and cell membrane staining. Altogether, our data suggest that galectin-3 favors the metastasis of melanoma cells and this process is not associated with β-catenin.

To evaluate the therapeutic effects of bone marrow-derived CD11b+CD14+ monocytes in a murine model of chronic liver *** liver damage was induced in C57bl/6 mice by administration of carbon tetrachloride and ethanol for 6 mo. Bone marrow-derived monocytes isolated by immunomagnetic separation were used for therapy. The cell transplantation effects were evaluated by morphometry, biochemical assessment, immunohistochemistry and enzyme-linked immunosorbent ***11b+CD14+ monocyte therapy significantly reduced liver fibrosis and increased hepatic glutathione levels. Levels of pro-inflammatory cytokines, including tumor necrosis factor-α, interleukin (IL)-6 and IL-1β, in addition to pro-fibrotic factors, such as IL-13, transforming growth factor-β1 and tissue inhibitor of metalloproteinase-1 also decreased, while IL-10 and matrix metalloproteinase-9 increased in the monocyte-treated group. CD11b+CD14+ monocyte transplantation caused significant changes in the hepatic expression of α-smooth muscle actin and *** therapy is capable of bringing about improvement of liver fibrosis by reducing oxidative stress and inflammation, as well as increasing anti-fibrogenic factors.
