Nutritional composition of Ten Persimmon cultivars in the “Ready-to-Eat crisp” Stage. Effect of Deastringency Treatment

Traditionally persimmons have been consumed over-ripened to avoid astringency perception. However, the introduction of new technology that removes astringency while preserving fruit firmness has allowed the commercialization of “ready-to-eat crisp” fruits. Several studies have evaluated the nutritional composition of over-ripened persimmons and have claimed that they are a good source of primary and secondary metabolites that are favourable for human health. Yet very little is known about the nutritional composition of persimmons in the “ready-to-eat crisp” stage. In this context, we determined the main nutritional compounds in ten popular persimmon cultivars, including astringent cultivars (“Rojo Brillante”, “Tone Wase”, “Giboshi”, “Kaki Tipo”, “Aizumishirazu-A”, “Giombo”, “Hachiya”) and non-astringent cultivars (“O’gosho”, “Hana Fuyu” and “Jiro”). To this end, fruits were harvested when their texture was firm, and soluble polyphenols content, total antioxidant capacity and main sugars, organic acids and carotenoids were evaluated. In those astringent cultivars at harvest, the changes in nutritional compounds associated with applying deastringency treatment with high cO2 concentration were determined. Our results revealed the main sugars (glucose, fructose and sucrose), organic acids (citric acid, malic acid and succinic) and carotenoids (β-cryptoxanthin, lutein, violoxanthin, zeaxanthin, and β-carotene) present in the flesh of crisp persimmons. At harvest the content of these metabolites vastly varied among cultivars;astringent cultivars showed higher soluble polyphenols and greater antioxidant capacity, and presented higher contents of sugars and organic acids than non-astringent ones. The deastringency treatment applied to astringent cultivars resulted in a drastic loss of soluble polyphenols and total antioxidant capacity, and induced changes in carotenoids and sugars composition.

Marginal discrepancy and microleakage in crown-copings fabricated by three cAD/cAM systems: An <i>in vitro</i>study

The purpose of this study was to evaluate and compare the microleakage and marginal discrepancy of ceramic crown-copings fabricated by three different cAD/cAM systems. Material and methods: Thirty extracted premolars were prepared for complete all-ceramic crowns with: total convergence 10-degree, 2 mm occlusal reduction, 4.5 mm height, 1.3 mm circular reduction and chamfer marginal preparation. They were divided into three groups and each group (n = 10) received a ceramic crown-coping fabricated by one of three cAD/cAM systems: Procera, KaVo Everest and cerec inLab. The copings were cemented with Panavia F 2.0. After thermal cycling, the teeth were immersed in 2% basic fuchsine dye for 24 hours. Microleakage was scored using a five-point scale and evaluated under a stereomicroscope at ×18. Marginal discrepancies were assessed with scanning electronic microscope at ×150, with measurements at 50 different points. Data were subjected to a chisquare test, Kolmogorov-Smirnov test and using Rho Spearman’s correlation test (P 0.05). No significant differences were found in marginal discrepancies for the Procera (62.58 ± 8.98 μm) and KaVo Everest systems (65.54 ± 18.56 μm), (P = 0.941). Significant differences were found for cerec inLab (132.18 ± 27.75 μm) compared with Procera and KaVo Everest (P = 0.0001). No correlation between the marginal discrepancies and microleakage for the three cAD/cAM crown-copings systems. conclusions: cerec inLab cAD/cAM crown-copings showed significantly higher values for microleakage and marginal discrepancy in comparison with Procera and KaVo Everest cAD/cAM crown-copings.

Lipid peroxidation and iron accumulation are closely associated with neurodegenerative diseases,such as Alzheimer’s,Parkinson’s,and Huntington’s diseases,or neurodegeneration with brain iron accumulation *** dysfunction,lipofuscin accumulation,autophagy disruption,and ferroptosis have been implicated as the critical pathomechanisms of lipid peroxidation and iron accumulation in these ***,the connection between lipid peroxidation and iron accumulation and the initial cause or consequence in neurodegeneration processes is *** this review,we have compiled the known mechanisms by which lipid peroxidation triggers iron accumulation and lipofuscin formation,and the effect of iron overload on lipid peroxidation and cellular *** vicious cycle established between both pathological alterations may lead to the development of ***,the investigation of these mechanisms is essential for exploring therapeutic strategies to restrict *** addition,we discuss the interplay between lipid peroxidation and iron accumulation in neurodegeneration,particularly in PLA2G6-associated neurodegeneration,a rare neurodegenerative disease with autosomal recessive inheritance,which belongs to the group of neurodegeneration with brain iron accumulation disorders.

Enzyme-Linked Immunosorbent Assays Using the Recombinant gp51 and p24 of Bovine Leukemia Virus for Immunodetection of the Disease

Enzyme-linked immunosorbent assay (ELISA) is often used to test bovine leukemia virus (BLV) infection. However, commercially available kits test in South America detect only antibodies against the gp51 protein. With the aim to improve the sensitivity of the test, we developed here a two-step indirect dual ELISA test that included both proteins p24 and gp51, expressed and produced in E. coli and baculovirus expression system respectively. Two hundred ten BLV sera, stated as double positive or double negative by the combination of commercial agar gel immunodiffusion (AGID) assay and a gp51-ELISA test, were tested with our in house dual rp24/rgp51 ELISA. Firstly, we checked the purified, optimized and standardized proteins as antigen by the checkerboard technique, and set up our in house ELISA test. The concordance correlation coefficient (ccc) and coefficient of variation (cV) intraplate repeatability levels were within the values established by the international standards. The statistical analysis demonstrated the value of sera correctly ranked highest (93.48%), and for 0.3 cutoff, the sensitivity was 95.65% and the specificity 91.30%. In conclusion, the rp24/rgp51 ELISA developed and standardized here demonstrated to have good analytical characteristics to be considered for screening of BLV.

Lithraea molleoides (Anacardiaceae) is a tree that grows in South America including Southern Brazil, Southern, and Eastern Bolivia, Southern Paraguay, Northern, and Central Argentina. Infusions, decoctions, or tinctures from its aerial parts (leaves, buds, and young stems) are employed in ethnomedicine mainly against respiratory, and digestive inflammations and illnesses. Antibacterial, antiviral, antioxidant, anti-inflammatory, and antinociceptive activities, among others, have been reported for L. molleoides. Many of its biological activities have been associated with the reported presence of 5-alkenyl resorcinols. Alkyl/alkenyl catechols and alkyl/alkenyl resorcinols are very common in members of the Anacardiaceae family and several activities have been attributed to them. This work describes the isolation and the structural elucidation of three new 5-alkenyl resorcinols isolated from Lithraea molleoides reported in nature for the first time.

Because of the growing worldwide demand for maize grain,new alternatives have been sought for breeding of this cereal,e.g.,development of polyembryonic varieties,which agronomic performance could positively impact the grain yield per unit area,and nutritional *** objectives of this study were to(1)determine the phytochemicals present in the embryo and endosperm of grain from maize families with high,low,and null polyembryony frequency,which were planted at different locations,and(2)state the relationship between these compounds and seed *** extracted phytochemicals from corn were identified by *** results showed that the genotype with the highest presence of phytochemicals was the brachytic population with high polyembryony called“BAP”,which also required less water during the germination *** number of phytochemicals in both embryo and endosperm tissues was not related to the sowing location where they came from or the type of *** number of different phytochemicals depended on the grain tissue from where they were *** chemical compounds found in the different maize tissues were related to the development of the plant,either in roots or nibs because these are mainly associated with the lignin synthesis.

本文对巴拿马碰撞法律法规,尤其是与海上碰撞事故中确定碰撞责任和碰撞损害赔偿相关的法律法规,进行了详细的研究。本文分析了一般情况下巴拿马管辖权对船舶碰撞案件的适用及范围,并明确了在巴拿马法律体系中没有确定船舶碰撞事故责任分担和责任主体的规定也是现存的问题之一。除此之外,本文还对如何通过比较法研究借鉴国外法律体系中确定船舶碰撞案件中责任主体的法律规定进行了阐述,并明确建立一个能够确定碰撞责任并确保责任主体支付到期赔偿的法律制度具有重要意义。本文还探讨了船舶碰撞案件中的民事责任,即对民事注意义务进行了阐述,对过失、损害以及因果关系等具体问题进行了分析。据预测,一般情况下船舶碰撞风险是主要风险之一,这一风险致使司法机关需要对由哪一方承担此次海事事故所造成的损害赔偿责任展开司法调查。本文选题源于修订和完善巴拿马法律以确定碰撞事故中谁是责任主体、谁来赔偿以及如何赔偿的需求。为了满足这一需求,本文对巴拿马现行法律进行分析,并将其与其他国家的法律进行比较研究。本文建议修改《巴拿马商事海商法》,在其中增加在船舶碰撞案件中确定碰撞责任主体以及责任比例的条款。因为这些条款有助于在碰撞发生时确定碰撞过失、责任以及责任主体,并为碰撞事故的裁判提供一个框架。这表明本文对此领域的研究具有重要意义。然而,还需要完善巴拿马船舶碰撞法规,以确定碰撞发生时的责任主体。

Whole-genome duplications are an important source of evolutionary novelties that change the mode and tempo at which genetic elements evolve within a genome. The cucurbita genus experienced a wholegenome duplication around 30 million years ago, although the evolutionary dynamics of the coding and noncoding genes in this genus have not yet been scrutinized. Here, we analyzed the genomes of four cucurbita species, in eluding a newly assembled genome of cucurbita argyrosperma, and compared the gene con tents of these species with those of five other members of the cucurbitaceae family to assess the evolutionary dynamics of protein-coding and long intergenic noncoding RNA (lincRNA) genes after the genome duplication. We report that cucurbita genomes have a higher protein-coding gene birth-death rate compared with the genomes of the other members of the cucurbitaceae family. c. argyrosperma gene families associated with pollination and transmembrane transport had significantly faster evolutionary rates. lincRNA families showed high levels of gene turnover throughout the phylogeny, and 67.7% of the lincRNA families in cucurbita showed evidence of birth from the neofunctionalization of previously existing protein-coding genes. collectively, our results suggest that the whole-genome duplication in cucurbita resulted in faster rates of gene family evolution through the neofunctionalization of duplicated genes.

AIM： To examine the in vivo phenotype associated with hepatic metastatic lymph node 64 （MLN64） overexpression.METHODS： Recombinant-adenovirus-mediated MLN64 gene transfer was used to overexpress MLN64 in the livers of c57BL/6 mice. We measured the effects of MLN64 overexpression on hepatic cholesterol content, bile flow, biliary lipid secretion and apoptosis markers. For in vitro studies cultured cHO cells with transient MLN64 overexpression were utilized and apoptosis by TUNEL assay was measured.RESULTS： Livers from ***64-infected mice exhibited early onset of liver damage and apoptosis. This response correlated with increases in liver cholesterol content and bilian/bile acid concentration, and impaired bile flow. We investigated whether liver MLN64 expression could be modulated in a murine model of hepatic injury. We found increased hepatic MLN64 mRNA and protein levels in mice with chenodeoxycholic acid-induced liver damage. In addition, cultured cliO cells with transient MLN64 overexpression showed increased apoptosis.cONcLUSION： In summary, hepatic MLN64 overexpression induced damage and apoptosis in murine livers and altered cholesterol metabolism. Further studies are required to elucidate the relevance of these findings under physiologic and disease conditions.
