Identification of the Sex of Earlier Embryos from Generic Hybrids of Chicken-Quail by Wpkci

In this study, a protocol was developed to identify the sex of earlier embryos of chicken （♂）-quail （♀） hybrids and successfully tested the sex proportion of each period （66-120 h）. We acquired cross bred eggs by artificial insemination, hatched them in the same batch according to the standard hatching condition of chicken, and collected earlier living embryos at 66, 72, 78, 84, 90, 96, 102, 108, 114, and 120 h randomly. We adopted RT-PCR protocol and multiple PCR, made the known sex quail as the external control, employed fl-actin as the internal control, and used primers that were designed according to conservative area of gene Wpkci of quail to identify the sex of earlier hybrid embryos. The results indicated that the primer of Wpkci can be used to identify the sex of hybrid embryos accurately; there were more male than female in earlier embryos, the sex proportion of earlier embryos compared with academic numerical value was significantly different （P〈0.01）, and there was no difference between different periods （P〉0.05）. In the present study, we concluded that a simple, fast, credible and stable protocol to identify the sex of earlier hybrids embryos had been established by using primer of Wpkci; in earlier embryos, the death rate of female was higher than that of male and there was no fluctuant peak.

We investigated the antiproliferative activity of （-）-gossypol on the human prostate cancer cell line PC3 in vitro and in vivo to elucidate its potential molecular mechanisms. Cell growth and viability were evaluated using the 3-（4,5-dimethylthiazol-2-yl）-2,5-diphenyltetrazolium bromide （MTT） assay, and cell apoptosis was detected by flow cytometry, terminal deoxynucleotidyl transferase dUTP nick end labelling （TUNEL） and electron microscopy. Expression of proliferating cell nuclear antigen （PCNA）, Bcl-2, CD31, caspase-3 and caspase-8 in tumour tissue was determined by immunohistochemistry. The drug concentration that yielded 50% cell inhibition （IC50 value） was 4.74 μg mL-1. In the PC-3 tumour xenograft study, （-）-gossypol （〉 5 mg kg-1） given once a day for 7 days significantly inhibited tumour growth in a dose-dependent manner. Immunohistochemical analysis revealed that （-）-gossypol enhanced caspase-3 and caspase-8 expression and decreased the expression of PCNA, Bcl-2 and CD31 in tumour tissues. It suggested that cell apoptosis and inhibition of angiogenesis might contribute to the anticancer action of （-）-gossypol.

Limited treatment options are available for aggressive prostate cancer. Gossypol has been reported to have a potent anticancer activity in many types of cancer. It can increase the sensitivity of cancer cells to alkylating agents, diminish multidrug resistance and decrease metastasis. Whether or not it can induce autophagy in cancer cells has not yet been determined. Here we investigated the antiproliferative activity of apogossypolone （ApoG2） and （-）-gossypol on the human prostate cancer cell line PC3 and LNCaP in vitro. Exposure of PC-3 and LNCaP cells to ApoG2 resulted in several specific features characteristic of autophagy, including the appearance of membranous vacuoles in the cytoplasm and formation of acidic vesicular organelles. Expression of autophagy-associated LC3-Ⅱ and beclin-1 increased in both cell lines after treatment. Inhibition of autophagy with 3-methyladenine promoted apoptosis of both cell types. Taken together, these data demonstrated that induction of autophagy could represent a defense mechanism against apoptosis in human prostate cancer cells.

This paper investigates the strength and critical state(CS)behaviours of granular materials via DEM simulations of true triaxial drained tests under three different loading modes including constant b(in-termediate stress ratio)tests with constantσ'1(major principal stress),constant p(mean pressure)and constantσ'3(minor principal stress)*** this end,a series of samples are generated with the same particle size distribution,and with the confining stresses ranging from 100 kPa to 900 *** CS is achieved for all *** the macroscopic behaviours and the microscopic behaviours are examined and compared considering different loading modes,confining stresses and intermediate stress ratios(b).The critical state lines(CSLs)are found to be unique and independent of the loading modes,but dependent on the b *** CSLs with b=0 and b=1 form the two boundaries of CSLs respectively beyond which CSLs under all other b tests cannot go *** different strength criteria are examined and compared in terms of both peak and CS *** Mohr-Coulomb strength criterion is found to be only suitable for axisymmetric loading *** Lade-Duncan criterion is only suitable for describing peak strengths,which is dependent on the loading modes and confining *** Satake criterion and Matsuoka-Nakai criterion are the more appropriate strength criteria for describing CS failures,indicating that the CS values of both the Satake parameter and the Matsuoka-Nakai parameter describe an inherent property that characterizes the CS failure for a given type of *** CS mechanical coordination number is fitted by a curved line for a given b value,which is unique regardless of the loading *** peak and CS values of both major and minor principal fabric tensors decrease with increasing b values,while they increase with increasing b values for the intermediate principal fabric tensor.

Accurate measurements of the radon exhalation rate help identify and evaluate radon risk regions in the *** these measurement methods,the closed-loop method is frequently ***,traditional experiments are insufficient or cannot analyze the radon migration and exhalation patterns at the gas–solid interface in the accumulation *** CFD-based technique was applied to predict the radon concentration distribution in a limited space,allowing radon accumulation and exhalation inside the chamber intuitively and *** this study,three radon exhalation rates were defined,and two structural ventilation tubes were designed for the *** consistency of the simulated results with the variation in the radon exhalation rate in a previous experiment or analytical solution was *** effects of the vent tube structure and flow rate on the radon uniformity in the chamber;permeability,insertion depth,and flow rate on the radon exhalation rate and the effective diffusion coefficient on back-diffusion were *** on the results,increasing the inser-tion depth from 1 to 5 cm decreased the effective decay constant by 19.55%,whereas the curve-fitted radon exhalation rate decreased(lower than the initial value)as the deviation from the initial value increased by approximately 7%.Increasing the effective diffusion coefficient from 2.77×10^(-7) to 7.77×10^(-6) m^(2) s^(-1) made the deviation expand from 2.14 to 15.96%.The conclusion is that an increased insertion depth helps reduce leakage in the chamber,subject to notable back-diffusion,and that the closed-loop method is reasonably used for porous media with a low effective diffusion coefficient in view of the back-diffusion *** CFD-based simulation is expected to provide guidance for the optimization of the radon exhalation rate measurement method and,thus,the accurate measurement of the radon exhalation rate.

Long CoVID,also known for post-acute sequelae of CovID-19,describes the people who have the signs and symptoms that continue or develop after the acute coviD-19 *** CovID patients suffer from an inflammation or host responses towards the virus approx-imately 4 weeks after initial infection with the SARS CoV-2 virus and continue for an unchar-acterized *** infected with CovID-19 before could experience long-CcovID conditions,including the patients who were infected with SARS CoV-2 virus confirmed by tests and those who never knew they had an infection *** with long CoviD may experience health problems from different types and combinations of symptoms over time,such as fa-tigue,dyspnea,cognitive impairments,and gastrointestinal(Gl)symptoms(e.g.,nausea,vom-iting,diarrhea,decreased or loss of appetite,abdominal pain,and dysgeusia).The critical role of the microbiome in these Gl symptoms and long CovID were reported in clinical patients and experimental ***,we provide an overall view of the critical role of the Gl tract and microbiome in the development of long COVID,including the clinical Gl symptoms in patients,dysbiosis,viral-microbiome interactions,barrier function,and inflammatory bowel disease patients with long *** highlight the potential mechanisms and possible treatment based on Gl health and ***,we discuss challenges and future direction in the long CoVID clinic and research.

Long runout landslides involve a massive amount of energy and can be extremely hazardous owing to their long movement distance,high mobility and strong destructive *** methods have been widely used to predict the landslide runout but a fundamental problem remained is how to determine the reliable numerical *** study proposes a framework to predict the runout of potential landslides through multi-source data collaboration and numerical analysis of historical landslide ***,for the historical landslide cases,the landslide-induced seismic signal,geophysical surveys,and possible in-situ drone/phone videos(multi-source data collaboration)can validate the numerical results in terms of landslide dynamics and deposit features and help calibrate the numerical(rheological)***,the calibrated numerical parameters can be used to numerically predict the runout of potential landslides in the region with a similar geological setting to the recorded *** of the runout prediction approach to the 2020 Jiashanying landslide in Guizhou,China gives reasonable results in comparison to the field *** numerical parameters are determined from the multi-source data collaboration analysis of a historical case in the region(2019 Shuicheng landslide).The proposed framework for landslide runout prediction can be of great utility for landslide risk assessment and disaster reduction in mountainous regions worldwide.
