Intestinal parameters of oxidative imbalance in celiac adults with extraintestinal manifestations

AIM To evaluate selected intestinal parameters of oxidative stress, and antioxidant capacity in adult celiac disease patients with extraintestinal *** The study involved 85 adult patients divided into the following subgroups:(1) patients with newly diagnosed celiac disease(CD)(n = 7);(2) celiac patients not adhering to a gluten-free diet(GFD)(n = 22);(3) patients with CD on the GFD(n = 31); and(4) patients with functional disorders of the gastrointestinal tract, serving as controls(n = 25). Celiac patients presented with non-classic symptoms or extraintestinal manifestations. Standard blood tests including serum antioxidant levels(uric acid, bilirubin, and vitamin D), celiac antibody levels, and histopathological status of duodenal biopsy specimens have been determined. The expression of m RNA for tumor necrosis factor α(TNF-α), interleukin 1β(IL-1β), interleukin 10(IL-10), superoxide dismutase(SOD), heat-shock protein 70(HSP-70), hypoxia-inducible factor 1(HIF-1α), and BAX in the duodenal mucosa of patients was analyzed by reverse transcriptase-polymerase chain reaction. RESULTS The mean plasma uric acid level in patients with active CD(newly diagnosed and nonadherent patients) and treated celiac patients was significantly higher than in controls(260.17 ± 53.65 vs 190.8 ± 22.98, P < 0.001, and 261.7 ± 51.79 vs 190.8 ± 22.98, P < 0.001, respectively). The mean bilirubin concentration in active and treated celiac patients was significantly lower than in controls(8.23 ± 5.04 vs 10.48 ± 4.08, P < 0.05 and 8.06 ± 3.31 vs 10.48 ± 4.08, P < 0.05, respectively). The mean plasma vitamin D level was significantly lower in active celiac patients than in treated celiac patients and controls(19.37 ± 9.03 vs 25.15 ± 11.2, P < 0.05 and 19.37 ± 9.03 vs 29.67 ± 5.12, P < 0.001, respectively). Mild Reductions in Mitochondrial NAD- Dependent Isocitrate Dehydrogenase Activity Result in Altered Nitrate Assimilation and Piqmentation But Do Not Impact Growth

Transgenic tomato （Solanum lycopersicum） plants were generated expressing a fragment of the mitochon- drial NAD-dependent isocitrate dehydrogenase gene （SIIDH1） in the antisense orientation. The transgenic plants displayed a mild reduction in the activity of the target enzyme in the leaves but essentially no visible alteration in growth from the wild-type. Fruit size and yield were, however, reduced. These plants were characterized by relatively few changes in pho- tosynthetic parameters, but they displayed a minor decrease in maximum photosynthetic efficiency （Fv/Fm）. Furthermore, a clear reduction in flux through the tricarboxylic acid （TCA） cycle was observed in the transformants. Additionally, bio- chemical analyses revealed that the transgenic lines exhibited considerably altered metabolism, being characterized by slight decreases in the levels of amino acids, intermediates of the TCA cycle, photosynthetic pigments, starch, and NAD（P）H levels, but increased levels of nitrate and protein. Results from these studies show that even small changes in mitochon- drial NAD-dependent isocitrate dehydrogenase activity lead to noticeable alterations in nitrate assimilation and suggest the presence of different strategies by which metabolism is reprogrammed to compensate for this deficiency.

COVID-19 Pandemic Waves in Italy: An Epidemiological Overview about Infections, Swabs and Death Rates

Introduction: On March 11, 2020, the World Health Organization (WHO) declared that the entire World was overrun by a pandemic. Science has managed, in a short time, to characterize a new disease, sequence a new viral genome, develop diagnostics, produce treatment protocols and establish the efficacy of drugs and vaccines in randomized controlled trials. In this paper we have dealt with different topics regarding the pandemic linked to COVID-19. Objectives: Aim of this paper is to compare the number of deaths attributable to COVID-19, that occurred in the different Italian macro-areas, in the different pandemic waves;we studied the trend of the curves relating to the proportion of deaths to the number of infected in the Italian macro-areas for the pandemic waves and analysed the mortality data, focusing on the Italian context and comparing it with other foreign countries. We examined the data regarding swabs, hospitalizations, home isolation, admissions to intensive care and deaths registered in pandemic period. Results: Geographically, Italy was differently affected by the pandemic. Northern Italy was the most affected area. In comparison with some foreign countries, Italy is one of the nations that paid the most in terms of deaths, due to: delays in understanding the seriousness of the emergency;the slow management in the tracking systems of contagions;the high number of hospitalizations;a corporate organizational system poorly planned. Conclusion: The years 2020 and 2021 have been dramatic and unprecedented. The year 2021 was the year of redemption, where, despite social, economic and health difficulties, thanks to mass vaccination, we were able to give a real strong response to the pandemic. Trust in science has led to a drastic decrease in mortality throughout the world.

Intrahepatic expression of genes related to metabotropic receptors in chronic hepatitis

AIM： To screen for genes related to metabotropic re- ceptors that might be involved in the development of chronic hepatitis. METHODS： Assessment of 20 genes associated with metabotropic receptors was performed in liver speci- mens obtained by punch biopsy from 12 patients with autoimmune and chronic hepatitis type B and C. For this purpose, a microarray with low integrity grade and with oligonucleotide DNA probes complementary to target transcripts was used. Evaluation of gene expression was performed in relation to transcript level, correlation between samples and grouping of clinical parameters used in chronic hepatitis assessment. Clini- cal markers of chronic hepatitis included alanine and aspartate aminotransferase, ~,-glutamyltranspeptidase, alkaline phosphatase and cholinesterase activity, levels of iron ions, total cholesterol, triglycerides, albumin, glucose, hemoglobin, platelets, histological analysis of inflammatory and necrotic status, fibrosis according to METAVIR score, steatosis, as well as anthropometric body mass index, waist/hip index, percentage of adi- pose tissue and liver size in ultrasound examination. Gender, age, concomitant diseases and drugs were also taken into account. Validation of oligonucleotide microarray gene expression results was done with the use of quantitative real-time polymerase chain reaction （qRT-PCR）. RESULTS： The highest （0.002 〈 P 〈 0.046） expres- sion among genes encoding main components of metabotropic receptor pathways, such as the a subunit of G-coupled protein, phosphoinositol-dependent pro- tein kinase or arrestin was comparable to that of an- giotensinogen synthesized in the liver. Carcinogenesis suppressor genes, such as chemokine ligand 4, tran- scription factor early growth response protein 1 and lysophosphatidic acid receptor, were characterized by the lowest expression （0.002 〈 P 〈 0.046）, while the factor potentially triggering hepatic cancer, transcrip- tion factor JUN-B, had a 20-fold higher expression. Living with high concentrations of urea: They can!

Marine elasmobranchs maintain their body fluid isoosmotic or slightly hyperosmotic to the external medium by the retention of large urea concentrations. This review focuses on the strategies adopted by these fishes to maintain a large outwardly direct concentration gradient of this osmolyte minimizing the loss across the main interfaces between body fluid and the external medium such as the gills, the kidney and the rectal gland, thus reducing the cost of making urea. The high plasma osmolarity, mainly main-tained by urea retention, is a challenge to volume homeostasis when fish move from seawater to water with a low salinity, since the high water permeability of branchial epithelium would cause a net flux of water into the animal. Since the renal regulation of urea retention in habitat with different salinities is crucial for the osmotic homeostasis of these species, the regulation of the activity and/or the expression of urea trans porters in renal tubules will be also discussed. In addition attention will be paid on the urea– methylamine system involved in maintaining the stability and functioning of many proteins since it is known that the high urea concentration found in marine elasmobranch fish, similar only to that found in mammalian kidney, has a destabilizing effect on many macromolecules and inhibits functions such as ligand binding.

Oxidative stress as a crucial factor in liver diseases

Redox state constitutes an important background of numerous liver disorders. The redox state participates in the course of inflammatory, metabolic and proliferative liver diseases. Reactive oxygen species(ROS) are primarily produced in the mitochondria and in the endoplasmic reticulum of hepatocytes via the cytochrome P450 enzymes. Under the proper conditions, cells are equipped with special molecular strategies that control the level of oxidative stress and maintain a balance between oxidant and antioxidant particles. Oxidative stress represents an imbalance between oxidant and antioxidant agents. Hepatocytic proteins, lipids and DNA are among the cellular structures that are primarily affected by ROS and reactive nitrogen species. The process results in structural and functional abnormalities in the liver. Thus, the phenomenon of oxidative stress should be investigated for several reasons. First, it may explain the pathogenesis of various liver disorders. Moreover, monitoring oxidative markers among hepatocytes offers the potential to diagnose the degree of liver damage and ultimately to observe the response to pharmacological therapies. The present report focuses on the role of oxidative stress in selected liver diseases.

A single nucleotide mutation in the dual-oxidase 2(DUOX2) gene causes some of the panda's unique metabolic phenotypes

The giant panda(Ailuropoda melanoleuca) is an iconic bear native to China,famous for eating almost exclusively *** unusual dietary behavior for a carnivore is enabled by several key adaptations including low physical activity,reduced organ sizes and hypothyroidism leading to lowered energy *** adaptive phenotypes have been hypothesized to arise from a panda-unique single-nucleotide mutation in the dual-oxidase 2(DUOX2) gene,involved in thyroid hormone *** test this hypothesis,we created genome-edited mice carrying the same point mutation as the panda and investigated its effect on metabolic *** mice were 27% smaller than heterozygous and wild-type ones,had 13 % lower body mass-adjusted food intake,5 5 % decreased physical activity,lower mass of kidneys(11%) and brain(5%),lower serum thyroxine(T4:36%),decreased absolute(12%) and mass-adjusted(5%) daily energy expenditure,and altered gut *** with T4 reversed the effects of the *** work uses a state-of-the-art genome editing approach to demonstrate the link between a single-nucleotide mutation in a key endocrine-related gene and profound adaptive changes in the metabolic phenotype,with great importance in ecology and evolution.

Predicting Electric Energy Consumption for a Jerky Enterprise

Wholesale and retail markets for electricity and power require consumers to forecast electricity consumption at different time intervals. The study aims to increase economic efficiency of the enterprise through the introduction of algorithm for forecasting electric energy consumption unchanged in technological process. Qualitative forecast allows you to essentially reduce costs of electrical energy, because power cannot be stockpiled. Therefore, when buying excess electrical power, costs can increase either by selling it on the balancing energy market or by maintaining reserve capacity. If the purchased power is insufficient, the costs increase is due to the purchase of additional capacity. This paper illustrates three methods of forecasting electric energy consumption: autoregressive integrated moving average method, artificial neural networks and classification and regression trees. Actual data from consuming of electrical energy was used to make day, week and month ahead prediction. The prediction effect of prediction model was proved in Statistica simulation environment. Analysis of estimation of the economic efficiency of prediction methods demonstrated that the use of the artificial neural networks method for short-term forecast allowed reducing the cost of electricity more efficiently. However, for mid- range predictions, the classification and regression tree was the most efficient method for a Jerky Enterprise. The results indicate that calculation error reduction allows decreases expenses for the purchase of electric energy.

近年来,中波两国的合作势头发展迅猛,要维持这个增长势头就需要保持有效的沟通交流,而培养合格的中-波译员可以帮助两国架起有效沟通的桥梁。然而,迄今为止中-波这一口译语言对在学术界引起的关注极少,作为一位学习口译方向的波兰人,作者希望尝试填补这方面的研究空缺。以吉尔的认知负荷模型与彼德·科德的错误分析理论为分析框架,本文分析了波兰学生在进行中-波同传中可能遇到的语言对特殊性方面的挑战。作者指出了波兰语在词汇、语法及句法层面的若干特点,并假设这些“错误诱因”可能会对中-波同传造成困难。作者对波兰雅盖隆大学的八位波兰籍汉学专业口译课的学生进行了一项中文译入波兰语的同传实验。通过收集和分析学生的译文,找出语言对特殊性如何影响学生的口译产出。数据表明,实验结果与本文最初的假设相符。为解决中-波语言对特有问题,作者提出了初步应对策略,以期打造更好的,以语言对特殊性为导向的课程。这些策略包括:增进对双方语言文化差异了解的前瞻性策略,以及简化,预测等过程性策略。对实验中口译学生所用策略的分析表明,过程性策略在中-波同传中具有一定的适用性。
