We investigated anti-hyperglycemic and anti-obese effects of Panax ginseng berry extract and its major constituent, ginsenoside Re, in obese diabetic C57BL/6J ob/ob mice and their lean littermates. Animals received daily intraperitoneal injections of Panax ginseng berry extract for 12 days. On day 5, 150 mg/kg extract-treated ob/ob mice had significantly lower fasting blood glucose levels compared to vehicle-treated mice (156±9.0 mg/dl vs. 243±15.8 mg/dl, P<0.01). 目的观察扶正化瘀片联合抗病毒药物治疗乙肝肝硬化(Liver cirrhosisi,LC)失代偿期患者的临床疗效及对其肝脏硬度、肝纤维化的影响。方法选取2020年5月—2022年5月期间淮安市第四人民医院收治的乙肝LC失代偿期患者共260例,按随机数字表法分为对照组和观察组,每组各130例。对照组给予恩替卡韦分散片治疗,观察组在对照组基础上加服扶正化瘀片,均治疗24周。观察比较两组患者治疗前后中医证候积分[胁痛、胁下痞块、面色晦暗、口干不欲饮、乏力]、肝功能[总胆红素(Total bilirubin,TBil)、谷草转氨酶(Alanine aminotransferase,AST)、谷丙转氨酶(Aspartate aminotransferase,ALT)、白蛋白(Albumin,ALB)]、血清肝纤四项[层黏连蛋白(Laminin,LN)、Ⅲ型前胶原(Procollagen typeⅢ,PCⅢ)、IV型胶原(IV collegen,CIV)、透明质酸(Hyaluronic acid,HA)]、肝脾影像学[脾静脉内径、门静脉内径、脾脏厚度、肝脏瞬时弹性硬度(Liver stiff-ness measurement,LSM)]及Th1/Th2型细胞因子[白细胞介素-4(Interleukin-4,il-4)、白细胞介素-6(Interleukin-6,il-6)、白细胞介素-18(Interleukin-18,il-18)、干扰素-γ(Inferferon-γ,IFN-γ)]指标变化情况,评估抗病毒效果[乙肝病毒脱氧核糖核酸(Hepatitis B virus-deoxyribonucleic acid,HBV-DNA)载量及转阴率、乙肝e抗原(Hepatitis B e antigens,HBeAg)转阴率]及临床疗效。结果治疗后两组患者胁痛、胁下痞块、面色晦暗、口干不欲饮和乏力证候积分均较治疗前降低,差异有统计学意义(Pil、AST、ALT水平均较治疗前降低,ALB水平均较治疗前升高,差异有统计学意义(Pil、AST、ALT水平均明显低于对照组,ALB水平明显高于对照组,差异有统计学意义(Pil-4、il-6水平较治疗前降低,il-18、IFN-γ水平较治疗前升高,差异有统计学意义(P0.05);且观察组il-4、il-6水平明显低于对照组,il-18、IFN-γ水平明显高于对照组,差异有统计学意义(P<0.05)。治疗后观察组HBV-DNA载量低于对照组。观察组HBV-DNA转阴率59.23%(77/130)和HBeAg转阴率56.15%(73/130)明显高于对照组41.54%(54/130)、38.46%(50/130),差异有统计学意义(P<0.05)。治疗后观察组总有效率93.85%(122/130)明显高于对照组81.54(106/130),差异有统计学意义(P<0.05)。结论扶正化瘀片与抗病毒药物联合治疗能够改善乙肝LC失代偿期患者症状和肝功能,降低肝脏硬度和肝纤维化程度,维持Th1/Th2型细胞因子平衡。

Pain is considered the most common complaint worldwide for which patients seek treatment. Conventional analgesic agents play an important role in modern pain therapy however they cause several adverse effects. Therefore newer and better analgesics continue to be investigated. In this controlled clinical trial, we evaluated the analgesic effects of two herbal medicines, Corydalis yanhusuo and Angelicae dahuricae. We used cold-pressor test;a simple, reliable, and widely used model in humans;for induction of tonic pain.

Hematopoietic stem cells (HSCs) are tissue-specific cells giving rise to all mature blood cell types regulated by a diverse group of hematopoietic cytokines and growth factors that influences the survival & proliferation of early progenitors and differentiation mechanisms by modulating the functional activities of HSCs. In this study, the functional yet distinctive role of three novel combinations of gene pairs RRAGC & PSMC2;CKAP4 & MANF;and CTR9 & CNTNAP2 have been newly identified. These novel combinations of genes were confirmed and expressed in K562 human leukemic cell line in the presence of cytokine combination (il-3, FLT-3 and SCF) using RT-PCR and siRNA-mediated gene knock down strategy. This study signifies the synergistic role of gene pairs in different molecular activities like ubiquitination or proteasomal degradation, calcium mobilization, dopamine signaling.

YBa2Cu3O7-x(YBCO)镀膜导体在电力等能源领域有巨大应用前景.利用倾斜衬底沉积法在无织构的金属衬底上生长了MgO双轴织构的模板层,在这一模板层上实现了双轴织构的YBCO薄膜的外延生长.这些膜的双轴织构用X射线极图分析、φ-扫描作了测定,观测到了MgO[001]方向相对于衬底法线倾斜了31°.研究了不同缓冲层材料对YBCO外延生长的取向和双轴织构的影响,外延生长的高质量的YBCO薄膜的转变温度和临界电流密度分别达到了91K和5.5×105A/cm2的较高的值.

本文研究表明,通过死麻风杆菌刺激单核吞噬细胞和Schwann细胞使其对杀伤细胞(K细胞)攻击的敏感性增强。与此相反,活麻风杆菌对宿主细胞的感染,与加热震荡蛋白质(HSP)诱异物一样,使两细胞对K细胞的敏感性降低。现将实验研究结果报告如下。死麻风杆菌刺激的K562细胞和单核吞噬细胞的裂解:通过与死分枝杆菌共同孵育24小时激活外周血单核细胞,然后采用自细胞介素(il-2)每周2次刺激单核细胞。2～4周后。

目的:基于网络药理学和分子对接探讨银翘散治疗儿童抽动障碍(tic disorder,TD)的潜在药理机制。方法:借助中药系统药理数据库和分析平台(TCMSP)检索筛选银翘散的活性成分,利用TCMSP,SwissTargetPrediction数据库和TargetNet服务器对银翘散活性成分的靶点进行预测,通过比较毒物基因组学数据库(Comparative Toxicogenomics Database,CTD)和人类基因数据库(The Human Gene Database,GeneCards)获取抽动障碍(Tic Disorder,TD)的相关靶点。通过Venny图网站(http://***/webtools/Venn/)取银翘散活性成分靶点与TD相关靶点交集,获取银翘散治疗TD的潜在靶点。利用Cytoscape 3.8.0对有效成分及治疗靶点进行可视化,构建“中药-有效成分-治疗靶点”网络。借助STRING 11.0数据库构建蛋白互作网络,同时借助Cytoscape 3.8.0软件中CytoHubba计算网络中节点的拓扑参数,筛选出网络中的关键靶点。利用R语言的“ClusterProfiler”包对潜在治疗靶点进行基因本体论(Gene Ontology,GO)和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)通路富集分析。最后,借助AutoDock Vina(Vina)平台进行分子对接,预测银翘散主要有效成分与关键靶点的结合度。结果:通过筛选得到银翘散的有效成分173个,预测靶点407个;TD相关靶点307个,银翘散治疗TD的治疗靶点共41个,其中ALB、il-6、AKT1、APP、NR3C1等靶点在银翘散治疗TD的生物网络中具有关键作用,主要涉及神经递质传递信号、炎症、免疫等信号转导通路。分子对接结果显示银翘散活性成分与ALB、il6、AKT1等关键靶点具有较高结合能力。结论:银翘散治疗TD的途径可能是多种活性成分和靶点的相互作用,通过调控免疫、炎症、神经递质等多条通路干预神经信号的传递。
