Although many causes of Alzheimer’s disease(Ad)may exist,both the original amyloid cascade a.d tau hypotheses posit that abnormal misfolding a.d accumulation of amyloid-β(Aβ)a.d tau protein is the central event causing the ***,that conclusion could be only partly true,a.d there is conflicting evidence about the role of both proteins in Ad,being able to precede a.d influence one *** researchers argue that these proteins are mere executors rather than primary causes of ***,there have been continuing refinements of both hypotheses,with alternative explanations proposed.Aβa.d tau proteins may be independently involved in specific neurotoxic pathways;yet there may be other crucial processes going on in early ***,accumulating evidence suggests that Aβa.d tau act synergistically,rather than a.ditively in disease onset(Jeremic et al.,2021,2023a).

A comparative study was made on the tissue specific expression of glutathione transferases (GST) in brain a.d testis after exposure of rat to phenobarbitol (PB) a.d β-methylcholanthrene (MC). Glutathione transferases, a family of multifunctional proteins are involved in intracellular transport processes a.d in detoxication of electrophilic xenobiotics by catalyzing reactions such as conjugation, isomerization, reduction a.d thiolysis. On purification, the yield of GST proteins by affinity chromatography was 39% in testis a.d 32% in brain. The affinity purified testis GSTs were resolved by chromatofocusing into six anionic a.d four cationic isozymes, a.d in brain glutathione transferases were resolved into four anionic a.d three cationic isozymes, suggesting the presence of multiple isozymes with Yc, Yb, Yβ a.d Yδ in both of them. In testis a.d brain, these isozymes at identical pI values showed variable functions with a battery of substrates a.d the cationic isozymes of brain a.d testis showed identical properties in CHP (cumene hydroperoxide) at pH values of above 7.0. Substrate specificity studies a.d immunoblot analysis of testis a.d brain proteins revealed that they play a predominant role in the detoxication of phenobarbitol or β-methylcholanthrene. Expression of the isozymes in testis a.d brain on exposure to PB a.d MC indicated elevated subunit variation. In both testis a.d brain, Yδ of π class was expressed on PB treatment a.d Yc of α class a.d Yβ of μ class was expressed in MC treated testis a.d only Yc was predominantly expressed in MC treated brain. Thus these subunits expression is considered as markers for carcinogenesis a.d specific to chemical toxicity under phenobarbitol a.d β-methylcholanthrene stress.
