Photothermal and photodynamic therapies(PTT/PDT)hold promise for localized tumor treatment,yet their full potential is hampered by limitations such as the hypoxic tumor microenvironment and inadequate systemic immune *** these challenges,we present a novel near-infrared(NIR)-triggered RNS nanoreactor(PBNO-Ce6)to amplify the photodynamic and photothermal therapy efficacy against triple-negative breast cancer(TNBC).The designed PBNOCe6 combines sodium nitroprusside-doped Prussian Blue nanoparticles with Chlorin e6 to enable on-site RNS production through NIR-induced concurrent NO release and ROS *** not only enhances tumor cell eradication but also potentiates local and systemic antitumor immune responses,protecting mice from tumor *** in vivo evaluations revealed that treatment with PBNO-Ce6 leads to a remarkable 2.7-fold increase in cytotoxic T lymphocytes and a 62%decrease in regulatory T cells in comparison to the control PB-Ce6(Prussian Blue nanoparticles loaded with Chlorin e6),marking a substantial improvement over traditional PTT/*** such,the PBNO-Ce6 nanoreactor represents a transformative approach for improving outcomes in TNBC and potentially other malignancies affected by similar barriers.

Arbuscular mycorrhiza and plant growth promoting endophytes facilitates accumulation of saponin under moderate drought stress

Objective: Paris polyphylla var. yunnanensis, one of the important medicinal plant resources in Yunnan,China, usually takes 6–8 years to be harvested. Therefore, it is urgent to find a method that can not only shorten its growth years, but also improve its quality. In this study, we examined the effects of a combination treatment of arbuscular mycorrhizal fungi(AMF) and plant growth-promoting endophytes(PGPE)and drought stress on the accumulation of saponins in ***: P. polyphylla var. yunnanensis was infected with a mixture of AMF and PGPE under drought stress. The content of saponins, as well as morphological, physiological, and biochemical indicators, were all measured. The UGTs gene related to saponin synthesis was obtained from transcriptome data by homologous comparison, which were used for RT-PCR and phylogenetic ***: Regardless of water, AMF treatment could infect the roots of P. polyphylla var. yunnanensis, however double inoculation with AMF and PGPE(AMF + PGPE) would reduce the infection rate of AMF. Plant height, aboveground and underground fresh weight did not differ significantly between the single inoculation AMF and the double inoculation treatment under different water conditions, but the inoculation treatment significantly increased the plant height of P. polyphylla var. yunnanensis compared to the noninoculation treatment. Single inoculation with AMF considerably increased the net photosynthetic rate,stomatal conductance, and transpiration rate of P. polyphylla var. yunnanensis leaves under various water conditions, but double inoculation with AMF + PGPE greatly increased the intercellular CO2concentration and chlorophyll fluorescence parameter(Fv/Fm). Under diverse water treatments, single inoculation AMF had the highest proline content, whereas double inoculation AMF + PGPE may greatly improve the amount of abscisic acid(ABA) and indoleacetic acid(IAA) compared to normal water under moderate drought. Two-photon photodynamic therapy(TP-PDT)has garnered significant attention because of its excellent depth of tissue penetration and high spatiotemporal ***,the limited targeting ability and oxygen dependency of photosensitizers(PSs)significantly hinder the effectiveness of photodynamic therapy in hypoxic tumor ***,we designed and synthesized two lipid droplet(LD)-targeted two-photon PSs(TBPCP and TBCP)by reducing benzene rings to achieve“acceptor planarization”.Notably,acceptor planarization not only enhanced the intramolecular charge transfer but also transferred the photochemical reaction from typeⅡ(TBPCP)to typeⅠ(TBCP).Under the irradiation of 940 nm femtosecond pulsed laser,TBPCP and TBCP showed bright two-photon-excited fluorescence and excellent LD targeting in living *** TBPCP(typeⅡPS),the outstanding TP-PDT efficacy of TBCP(typeⅠPS)under hypoxic conditions could be obtained in both cellular experiments and multicellular tumor spheroids(MCTS)***,both TBPCP and TBCP could induce the lipid peroxidation in the typeⅠor typeⅡPDT due to the location of LD,depleting GSH and inactivating GPX4 to induce nonprogrammed ferroptosis in cells.

Perspective of Calcium Imaging Technology Applied to Acupuncture Research

Acupuncture, a therapeutic treatment defined as the insertion of needles into the body at specific points(ie, acupoints), has growing in popularity world-wide to treat various diseases effectively, especially acute and chronic pain. In parallel, interest in the physiological mechanisms underlying acupuncture analgesia, particularly the neural mechanisms have been increasing. Over the past decades, our understanding of how the central nervous system and peripheral nervous system process signals induced by acupuncture has developed rapidly by using electrophysiological methods. However, with the development of neuroscience, electrophysiology is being challenged by calcium imaging in view field, neuron population and visualization in vivo. Owing to the outstanding spatial resolution, the novel imaging approaches provide opportunities to enrich our knowledge about the neurophysiological mechanisms of acupuncture analgesia at subcellular, cellular, and circuit levels in combination with new labeling, genetic and circuit tracing techniques. Therefore, this review will introduce the principle and the method of calcium imaging applied to acupuncture research. We will also review the current findings in pain research using calcium imaging from in vitro to in vivo experiments and discuss the potential methodological considerations in studying acupuncture analgesia.

Intraplantar Injection of Monosodium Iodoacetate Produces Hyperalgesia in Rats and the Mechanism Underlying the Effect

Objective: To observe the effect of intraplantar injection of monosodium iodoacetate (MIA) on pain perception in rats and to investigate the role of transient receptor potential vanilloid type 1 (TRPV1) in the effect. Methods: Adult male Wistar rats were used in the experiment. 1) MIA was injected subcutaneously into the right hindpaw of rats, the low, medium, and high doses of MIA were 0.11, 0.33, and 1 mg, respectively, then the changes of paw withdrawal thermal latency, paw withdrawal mechanical threshold, and dynamic weight bearing within 4 hours after MIA injection were measured. 2) Capsazepine (TRPV1 antagonist, 30 μg) was injected subcutaneously into the right hindpaw of rats at 2 hours after intraplantar injection of MIA (1 mg), then the changes of paw withdrawal thermal latency, paw withdrawal mechanical threshold, and dynamic weight bearing within 1 hour after capsazepine injection were measured. Results: 1) The paw withdrawal thermal latency, paw withdrawal mechanical threshold, and dynamic weight bearing decreased after intraplantar injection of MIA in rats and the effect lasted for at least 4 hours. 2) The MIA-induced reduction in paw withdrawal thermal latency, paw withdrawal mechanical threshold, and dynamic weight bearing were significantly alleviated after intraplantar injection of capsazepine in rats. Conclusion: Intraplantar injection of MIA can produce thermal pain, mechanical pain, and spontaneous pain for more than 4 hours, which may be due to the TRPV1 activation caused by MIA.
