The goal of this stud. was to d.termine whether mutation of the Mn-bind.ng site of wild.type recombinant Phlebia rad.ata manganese peroxid.se 3 affected.the pH-d.pend.nce kinetic parameters. pH range investigated.was 2.5 – 12.0. The catalytic efficiency of the mutant enzymes at high and.low pH in comparison to the wild.type was investigated.using stand.rd.rPr-MnP3 protocol. Wild.type recombinant Phlebia rad.ata MnP3 enzyme showed.optimal activity with Mn (II) as substrate at pH 5.0 and.remained.mod.rately active (approximately 40%) in the pH range of 6.0 - 9.0. The rPr-MnP3 mutants’ maximum activity ranged.between 5.5 and.8.0. Wild.type and.mutants rPr-MnP3 enzymes exhibited.a similar pH profile with optimum pH of 3.0 for ABTS oxid.tion. Mutation has severely d.creased.the catalytic efficiency for Mn (II) oxid.tion at pH 5.0. The rPr-MnP3 enzymes showed.enhanced.affinity for Mn (II) at alkaline pH and.a more alkaline range for catalysis than ever reported.for any Manganese Peroxid.se. This stud. reveals that at higher pH, rPr-MnP3 can function with alternative ligand. in the Mn (II) site and.d.es not have an absolutely obligate requirement for an all carboxylate ligand.set. These results further strongly confirm that Mn2+ bind.ng site is the only prod.ctive catalytic site for Mn (II) oxid.tion.

A Multigrid.Method.for a Mod.l of the Implicit Immersed.Bound.ry Equations

Explicit time stepping schemes for the immersed.bound.ry method.require very small time steps in ord.r to maintain *** the equations that arise from an implicit d.scretization is ***,several d.fferent approaches have been proposed.but a complete und.rstand.ng of this problem is still emerging.A multigrid.method.is d.veloped.and.explored.for solving the equations in an implicittime d.scretization of a mod.l of the immersed.bound.ry *** mod.l problem consists of a scalar Poisson equation with conformation-d.pend.nt singular forces on an immersed.*** mod.l d.es not includ. the inertial terms or the incompressibility *** method.is more efficient than an explicit method.but the efficiency gain is *** multigrid.method.alone may not be an effective solver,but when used.as a precond.tioner for Krylov method.,the speed.up over the explicit-time method.is *** example,d.pend.ng on the constitutive law for the bound.ry force,with a time step 100 times larger than the explicit method.the implicit method.is about 15-100 times more efficient than the explicit method.A very attractive feature of this method.is that the efficiency of the multigrid.precond.tioned.Krylov solver is shown to be ind.pend.nt of the number of immersed.bound.ry points.

Hyd.od.namic Interaction of Elastic Capsules in Bound.d.Shear Flow

This paper presents a mod.fied.Loop’s subd.vision algorithm for stud.ing the d.formation of a single capsule,the hyd.od.namic interaction between two capsules and.the hyd.od.namic d.ffusion of a suspension of capsules in bound.d.shear flow.A subd.vision thin-shell mod.l is employed.to compute the forces generated.on the surface of the elastic capsule d.ring *** capsule surface is approximated.using the mod.fied.Loop’s subd.vision scheme which guarantees bound.d.curvature and.C1 continuity everywhere on the limit *** present numerical technique has been valid.ted.by stud.ing the d.formation of a spherical capsule in shear *** are also performed.for a biconcave capsule over a wid. range of shear rates and.viscosity ratios to investigate its *** ad.ition,the hyd.od.namic interaction between two elastic capsules in bound.d.shear flow is *** on the wall separation d.stance,the trajectory-bifurcation points that separate reversing and.crossing motions for both spherical and.biconcave capsules can be *** to the spherical capsules,the biconcave capsules exhibit ad.itional types of interaction such as rotation and.head.on *** head.on collision results in a large trajectory shift which contribute to the hyd.od.namic d.ffusion of a suspension.A suspension of a large number of biconcave capsules in shear flow is also simulated.to show the ability of the mod.fied.scheme in running a large-scale simulation over a long period.of time.

Patients with long-stand.ng inflammatory bowel d.sease(IBd.involving at least 1/3 of the colon are at increased.risk for colorectal cancer(CRC).Ad.ancements in CRC screening and.surveillance and.improved.treatment of IBd.has red.ced.CRC incid.nce in patients with ulcerative colitis and.Crohn’s *** cases of CRC are thought to arise from d.splasia,and.recent evid.nce suggests that the majority of d.splastic lesions in patients with IBd.are visible,in part thanks to ad.ancements in high d.finition colonoscopy and.*** practice guid.lines have supported.the use of chromoend.scopy with targeted.biopsies of visible lesions rather than trad.tional rand.m *** are encouraged.to end.scopically resect visible d.splasia and.only recommend.surgery when a complete resection is not *** technologies such as virtual chromoend.scopy are emerging as potential tools in CRC *** with IBd.at increased.risk for d.veloping CRC should.und.rgo surveillance colonoscopy using new approaches and.techniques.

Epigenetic effects of ethanol on liver and.gastrointestinal injury

Alcohol consumption causes cellular injury. Recent d.velopments ind.cate that ethanol ind.ces epigenetic alterations, particularly acetylation, methylation of histones, and.hypo- and.hypermethylation of d.A. This has opened.up a new area of interest in ethanol research and.is provid.ng novel insight into actions of ethanol at the nucleosomal level in relation to gene expression and.patho-physiological consequences. The epigenetic effects are mainly attributable to ethanol metabolic stress (Emess), generated.by the oxid.tive and.non-oxid.tive metabolism of ethanol, and.d.sregulation of methionine metabolism. Epigenetic changes are important in ethanol-ind.ced.hepatic steatosis, fibrosis, carcinoma and.gastrointestinal injury. This ed.torial highlights these new ad.ances and.its future potential.

The steel reinforced.plastic pipe is a new kind.of pressure pipe. It is mad. up with steelwires and.plastic. Because reinforced.skeleton of the steel wire increase the complexityof plastic flow d.ring the extrusion phase, the trad.tional d.sign criteria of extrusiond.e is not suitable. The stud. on extrusion d.e of the kind.of pipe is very importantstep in prod.ce d.velopment. Using finite element (FE) method.in this paper, theflow rule of molten plastic insid. the d.e has been pred.cted.and.a groap of optimalstructural parameters was obtained. These results are helpful for red.cing the d.signcycle and.improve the quality of the final prod.ct.

The process of mixed.rare earth metal (REM) conversion coating on aluminum alloy LY12 (2024) was introd.ced. The corrosion resistance of mixed.REM conversion coating was examined.by using weight loss test and.electrochemical method. The results show that the mixed.REM conversion coating increases the corrosion resistance of the alloy surface. Examination result of scanning electron microscope (SEM) shows that the coating exhibits honeycomb-like feature. X-ray d.ffraction (XRd. analysis reveals that the coating is amorphous. The result of X-ray photoelectron spectroscopy (XPS) ind.cates that mixed.REM conversion coating consists pred.minantly of Ce and.O. The contents of other rare earth elements (La, Pr) are relatively low.
