SERPINH1 promoted the proliferation and metastasis of colorectal cancer by activating PI3K/Akt/mTOR signaling pathway

BACKGROUND Serpin peptidase inhibitor clade H member 1(SERPINH1)was initially recognized as an oncogene implicated in various human malignancies.Nevertheless,the clinical relevance and functional implications of SERPINH1 in colorectal cancer(CRC)remain largely elusive.AIM To investigate the effects of SERPINH1 on CRC cells and its specific mechanism.METHODS Quantitative real-time polymerase chain reaction,western blotting analysis,The Cancer Genome Atlas data mining and immunohistochemistry were employed to examine SERPINH1 expression in CRC cell lines and tissues.A series of in-vitro assays were performed to demonstrate the function of SERPINH1 and its possible mechanisms in CRC.RESULTS SERPINH1 demonstrated elevated expression levels in both CRC cells and tissues,manifested at both mRNA and protein tiers.Elevated SERPINH1 levels correlated closely with advanced T stage,lymph node involvement,and distant metastasis,exhibiting a significant association with poorer overall survival among CRC patients.Subsequent investigations unveiled that SERPINH1 overexpression notably bolstered CRC cell proliferation,invasion,and migration in vitro,while conversely,SERPINH1 knockdown elicited the opposite effects.Gene set enrichment analysis underscored a correlation between SERPINH1 upregulation and genes associated with cell cycle regulation.Our findings underscored the capacity of heightened SERPINH1 levels to expedite G1/S phase cell cycle progression via phosphatidylinositol 3-kinase/AKT/mechanistic target of rapamycin pathway activation,thereby facilitating CRC cell invasion and migration.CONCLUSION These findings imply a crucial involvement of SERPINH1 in the advancement and escalation of CRC,potentially positioning it as a novel candidate for prognostic assessment and therapeutic intervention in CRC management.

The combination of horseradish peroxidase(HRP)and a fluorescence substrate has been attracting great interests in developing sensitive biochemical analysis and immunoassays.10-Acetyl-3,7-dihydroxyphenoxazine(ADHP or Amplex red)is the most sensitive fluorogenic substrate known for HRP in current market,however,it suffers from some drawbacks,such as non-specific reactivity to carboxylesterase and limited fluorescence stability.In the present study,a novel HRP substrate10-cyclopropylcarbonyl-dichloro-dihydroxyphenoxazine(AR-2),has been prepared,which exhibited improved sensitivity than ADHP in sensing HRP.Moreover,the fluorescence of AR-2/HRP demonstrated improved tolerance to physiological relevant p H fluctuation as compared to ADHP/HRP.Successful detection of uric acid/urate oxidase reaction indicated excellent application prospect of AR-2/HRP for monitoring H_(2)O_(2)-generating biochemical reactions.More interestingly,an enzyme-linked immunosorbent assay(ELISA)using AR-2 as the fluorescence reporter has been successfully used in detecting IgG against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)from human serum samples.Overall,AR-2 exhibits improved performances over the commercial ADHP,which will be an ideal alternative to ADHP in HRP-based fluorescence biochemical analysis and immunoassays.

Objective YAP1 plays a dual role as an oncogene and tumor suppressor gene in several tumors;differentiating between these roles may depend on the YAP1 phosphorylation pattern.The specific function of YAP1 in B cell acute lymphoblastic leukemia(B-ALL),however,is currently unclear.Thus,in the present study,the role of YAP1 in B-ALL was investigated using relevant cell lines and patient datasets.Methods The effects of shRNA-mediated knockdown on YAP1 and LATS1 levels in the NALM6 and MOLT-4 cell lines were examined using Western blotting,quantitative real-time polymerase chain reaction,flow cytometry,immunostaining,and nude mouse subcutaneous tumorigenesis experiments.Gene expression levels of Hippo pathway-related molecules before and after verteporfin(VP)treatment were compared using RNA-Seq to identify significant Hippo pathway-related genes in NALM6 cells.Results Patients with ALL showing high YAP1 expression and low YAP1-Ser127 phosphorylation levels had worse prognoses than those with low YAP1 protein expression and high YAP1-Ser127 phosphorylation levels.YAP1-Ser127 phosphorylation levels were lower in NALM6 cells than in MOLT-4 and control cells;YAP1 was distributed in the nuclei in NALM6 cells.Knockdown of YAP1 inhibited MOLT-4 and NALM6 cell proliferation and arrested the NALM6 cell cycle in the G0/G1 phase.Before and after VP treatment,the expression of the upstream gene LATS1 was upregulated;its overexpression promoted YAP1-Ser127 phosphorylation.Further,YAP1 was distributed in the plasma.Conclusion LATS1 may downregulate YAP1-Ser127 phosphorylation and maintain B-ALL cell function;thus,VP,which targets this axis,may serve as a new therapeutic method for improving the outcomes for B-ALL patients.

Soft electrochemical actuators that can generate mechanical motions in response to electrical stimuli can be used as artificial muscles in microscale robots.The electrode materials are the key component of the electrochemical actuator because they limit the ion diffusion and accumulation processes and thus affect the deformation in fast response-time and potential engineering applications.Low-dimensional materials,such as one-dimensional(1D)CNT and two-dimensional(2D)vertically stood WS_(2),are great electrode due to their good electrochemical stability and high surface-to-volume ratios,but show limited performances when being used on their own in the actuators because of low reconstruction or low conductivity.A combination of 1D CNT and 2D WS_(2)could form a heterojunction with both high electrochemical activity and great mechanical stability.Herein,by depositing highly disordered graphene nanosheets(HDGNs)on CNT films and then WS_(2)on the formed low-dimensional nanocarbon film,a hybrid WS_(2)/HDGN/CNT film was synthesized through a multistep CVD method.Vertically grown WS_(2)nanosheets were achieved by taking advantage of HDGNs which limits CNTs sliding.So,the volume expansion caused by the transition from WO_(3)to WS-2can force the vertical growth of WS_(2).This hybrid film shows excellent electrochemical properties,such as high specific capacitance and high rate performance.Soft actuators based on the WS_(2)/HDGN/CNT films show good actuating performance,including stability and reconfigurability.

在明初的乡村治理理念中,乡里制度是以里甲制为中心的综合多功能体,蕴含了丰富的社会管理、教化、治安等功能,也蕴含了巧妙的慈善功能。鳏寡孤独残疾有养、婚姻丧葬有助等慈善目标被融于官方的礼法体系和里甲编排等制度安排中。明中叶以后,社会分化加剧,里甲面临瓦解,部分职能废弛,有识之士在重整乡村秩序的过程中,借重的依然是明初的乡治理念。以乡约为中心的保甲、社仓等制度设计成为明代中后期基层治理和慈善救助的主要内容,保甲、乡约、社仓、社学“四合一”是明中后期理想的乡治模式。由于明代乡村治理中贯穿了精神与物质、慈善与控制相结合的原则,基层社会得以在较长时间中维持相对稳定的秩序。

High serum reproductive hormone levels at mid-pregnancy support Meishan pig prolificacy

Increasing prolificacy is an important aim in the pig industry. Regions associated with litter size have been revealed, but detailed molecular mechanisms are unclear. The Meishan pig is one of the most prolific breeds, with higher prolificacy than the Yorkshire pig, which exhibits high feeding efficiency and lean meat yield. The ovary is the key organ determining reproductive traits during pregnancy by synthesizing and secreting reproductive hormones essential for conceptus maintenance. In this comparative multi-omics study of the ovary transcriptome, proteome, and metabolome on day 49 of pregnancy, we aimed to identify genomic, proteomic, and metabolomic differences between the ovaries of Meishan and Yorkshire pigs to reveal potential molecular mechanisms conferring high prolificacy. Meishan pigs demonstrated general downregulation of steroid biosynthesis and butanoate metabolism in the ovary during mid-pregnancy at both transcriptome and proteome levels but exhibited higher serum cholesterol, estradiol, and progesterone levels than Yorkshire pigs. We also identified several single-nucleotide polymorphisms in the genes of the steroid hormone pathway associated with litter number, average birth weight, and total litter weight. Lower biosynthesis rates but elevated serum levels of reproductive hormones during mid-and late pregnancy are essential for the greater prolificacy of Meishan pigs.

SlGH9-15 regulates tomato fruit cracking with hormonal and abiotic stress responsiveness cis-elements

Fruit cracking occurs easily during the late period of fruit development when plants encounter an unsuitable environment,dramatically affecting fruit production and marketing.This study conducted the bulked segregant RNA-Seq(BSR)to identify the key regulatory gene of fruit cracking in tomatoes.BSR-Seq analysis illustrated that two regions associated with irregularly cracking were located on chromosomes 9 and 11,containing 127 candidate genes.Further,through differentially expression analysis and qRT-PCR in cracking-susceptible and cracking-resistant genotypes,the candidate gene SlGH9-15(Solyc09g010210)with significantly differential expression levels was screened.Bioinformatics analysis of the GH9 gene family revealed that 20 SlGH9 genes were divided into three groups.The phylogenetic analysis showed that SlGH9-15 was closely related to cell wall construction-associated genes AtGH9B1,AtGH9B6,OsGH9B1,and OsGH9B3.The cis-acting elements analysis revealed that Sl GH9-15 was activated by various hormones(ethylene and ABA)and abiotic stresses.The expression pattern indicated that 13 SlGH9 genes,especially SlGH9-15,were highly expressed in the cracking-susceptible genotype.Its expression level gradually increased during fruit development and achieved maximum value at the red ripe stage.Additionally,the cracking-susceptible tomato showed higher cellulase activity and lower cellulose content than the cracking-resistant tomato,particularly at the red ripe stage.This study identified SlGH9-15 as a key gene associated with fruit cracking in tomatoes for the first time and gives new insights for understanding the molecular mechanism and complex regulatory network of fruit cracking.

Sesame(Sesamum indicum L.)is a significantly lucrative cash crop for millions of small-holder farmers.Its seeds are an important source of a highly appreciated vegetable oil globally and two clinically essential antioxidant lignans,sesamin and sesamolin.Accordingly,many countries import millions of tons of sesame seed every year.The demand for lignan-rich sesame seeds has been increasing in recent years due to the continuous discovery of several pharmacological attributes of sesamin and sesamolin.To meet this demand,the sesame breeder’s primary objective is to release sesame cultivars that are enriched in oil and lignans.Thus,it is necessary to summarize the information related to the sesamin and sesamolin contents in sesame in order to promote the joint efforts of specialized research teams on this important oilseed crop.In this article,we present the current knowledge on the sesamin and sesamolin contents in S.indicum L.with respect to the updated biosynthesis pathway,associated markers,governing loci,available variability in sesame germplasm,the in planta potential roles of these compounds in sesame,and the newly discovered pharmacological attributes.In addition,we propose and discuss some required studies that might facilitate genomics-assisted breeding of high lignan content sesame varieties.

Wobbling motion for a triaxial rotor plus a single quasiparticle

Wobbling motion in a system comprising a triaxial rotor and a single quasiparticle is studied employing the particle-rotor model.The energy spectra,wobbling frequencies,electromagnetic transition probabilities,g-factors,angular momentum components,spin coherent state maps,and static quadrupole moments are investigated.These investigations were conducted with regard to the Fermi surface transitioning from the lowest h_(11/2) orbit to the highest one.As the Fermi surface increases,notable transformations occur in the wobbling mode.Initially,the mode exhibits a transverse revolution around the short axis of the nucleus.However,as the Fermi surface continues to increase,the mode gradually shifts to a longitudinal revolution around the intermediate axis.Eventually,it transitions to a transverse revolution around the long axis.Notably,the stability of the long axis transverse mode diminishes relative to its counterpart along the short axis as the total angular momentum increases atγ=20∘.
