Changes of nutrients and microbial communities in recovery process of abandoned rare earth tailings

Rare earth element(REE)mining has wrought severe ecosystem destruction,particularly in the loss of carbon(C),nitrogen(N),and phosphorus(P).Furthermore,its effects on nutrient recovery characteristics and associated microbial abundance are still poorly *** this study,an investigation was conducted to determine C,N,and P recovery patterns and microbial communities in abandoned REE mining tailings with different abandoned durations of 1.5(1.5-Y),7(7-Y),10(10-Y),and 14(14-Y)years and at an unmined site(the control group)in Jiangxi Province,*** tailing delamination at each site was investigated in 0–15(layer 1)and 15–30(layer 2)cm tailing *** the abandoned duration of REE tailings,C and P in layer 1 continued to recover from 1.62 to 8.51 g kg^(-1) and from 0.71 to 1.94 g kg^(-1),respectively;N losses of 0.60 and 0.22 g kg^(-1) occurred at the 1.5-Y and 10-Y sites,while N in layer 1 slightly increased at the 14-Y ***,Methylosinus,Bradyrhizobium,Pseudomonas,and Azospirillum were dominant bacteria at the 14-Y site and in the *** relative abundances of N-fixing bacteria Mesorhizobium(32.94%),Sinorhizobium(0.24%),Frankia(0.71%),and Burkholderia(2.38%)at the 14-Y site were 1.33–3.97 times those in the control(24.19%,0.18%,0.32%and 0.60%,respectively),which was helpful for tailing N *** the 14-Y site,Tylospora(50.24%),Luellia(11.02%),Tomentella(6.94%),and Chaetomium(4.34%)were the dominant fungal genera,while Tylospora(56.93%),Suillus(11.45%),and Penicillium(6.78%)predominated in the *** relative abundance of the P-dissolving fungus Aspergillus at the 14-Y site(0.42%)was 4.2 times that in the control(0.1%),which may lead to the improved tailing P *** the 14-Y site,woody plants such as Pinus massoniana and native pioneering herbs and ferns such as Dicranopteris dichotoma and Nephrolepis auriculata probably invaded from adjacent *** study unraveled the natural restoration of plant and micro

Enhanced adaptability of Sesbania rostrata to Pb/Zn tailings via stem nodulation

Sesbania rostrata is wellknown for its stem nodulation, but the roles of stem nodulation in root nodulation and adaptation of S. rostrata to Pb/Zn-enriched tailings environment has been poorly understood. We investigated the effects of inoculating （with stem nodule treatment） and non-inoculating （without stem nodule treatment） Azorhizobium caulinodans on the growth, root nodulation, and N fixation of S. rostrata grown on three different types of soil substrata： Pb/Zn tailings, garden soil amended tailings, and garden soil. The results showed that plant height, stem basal diameter, biomass, chlorophyll content, nitrogen content and N-accumulation per plant were 2.3%-4.9%, 2.2%-7.7%, 27.8%-72.2%, 17.1%-23.5%, 12.3%-34.2%, and 43.1%-131.2%, respectively, higher in treatments with stem nodule than those without stem nodule for the same soil substrate. With respect to soil substrata, all measurements had consistently higher values in tailings than in amended tailings and garden soil, indicating that the poorer the soil condition, the greater the contribution of stem nodule. In contrast, the number and fresh weight of root nodules on plants without stem nodule were 6.9-11.6 times and 5.8-29.0 times higher than those with stem nodule, respectively, especially with respect to the plants grew on Pb/Zn tailings. In general, stem nodulation favored plant growth and nitrogen fixation of S. rostrata, but suppressed root nodulation. With the ability of stem and root nodulation, S. rostrata can be used as a pioneer plant species for remediation of Pb/Zn tailings.

【目的】液泡膜糖转运蛋白(TST或者TMT)是植物发育过程中发挥重要作用的一种糖转运蛋白。研究旨为探究TST基因家族在葡萄生长发育中的作用,并进一步为阐明TST基因功能提供坚实的基础。【方法】通过同源分析法从葡萄基因组中鉴定出13个TST基因,对基因的结构和编码蛋白质进行生物信息学分析;用qRT-PCR技术分析‘鄞红’葡萄发育过程中不同组织的TST表达水平,并对不同时期葡萄果肉中可溶性糖含量进行相关性分析。【结果】TST基因家族分布在6条染色体上,存在3对片段重复和3对串联重复基因;根据系统发育将其分为3个亚家族,各亚家族成员结构相似;TST基因含有大量与激素、光和胁迫响应相关的顺式作用元件;TST家族蛋白质由α-螺旋和无规则卷曲组成,各亚族蛋白模型相似;VvTST在不同组织中均有表达,并存在时空表达特异性;相关性分析发现,葡萄果肉中4个VvTST基因(VIT_18s0001g12560、VIT_18s0122g00850、VIT_04s0023g01860和VIT_03s0038g03940)的表达水平与其中可溶性糖的积累呈现相似变化趋势。【结论】VvTST可能对葡萄果肉中可溶性糖积累起到至关重要的作用。

Cr-C类石墨复合涂层制备表征及其在硬质合金铣刀上的应用

为了提高硬质合金铣刀的使用寿命,降低摩擦加工时的摩擦因数,本文结合Cr元素对涂层生长的促进作用与类石墨(GLC)涂层的优异性能,采用磁控溅射离子镀的方法,在硬质合金(YG8)上制备Cr-C类石墨复合涂层。通过拉曼光谱对复合涂层成分类石墨进行表征。利用扫描电镜观察涂层截面形貌,结合线扫描与面扫描结果对涂层元素成分分布进行表征。对复合涂层试样进行摩擦磨损试验,测定摩擦因数并计算磨损率,采用硬度测量仪测定复合涂层的硬度,使用自动划痕仪来评定复合涂层结合力。结果表明:Cr-C类石墨复合涂层具有典型的类石墨非晶碳结构特征,复合涂层整体厚度约为0.68μm;Cr元素主要分布在底层与过渡层,少量掺杂在工作层;C元素主要分布在工作层中,各层涂层内部没有裂纹、孔洞等缺陷,涂层的致密性较好。Cr-C类石墨复合涂层摩擦因数约为0.2,由磨痕形貌测得磨损体积为65.6651 mm^(3),磨损率为2.28×10^(-4) mm^(3)/(N·mm),复合涂层摩擦因数低,耐磨性好。复合涂层硬度可达824~986 HRV,与传统类石墨涂层相比,硬度提升28.22%~51.69%,复合涂层与基体之间的结合力为24 N,与同样应用于硬质合金铣刀的涂层相比,具有良好的结合性能。本文研究可为Cr-C类石墨复合涂层在硬质合金铣刀中的应用提供依据。

Identification of cadmium-induced genes in maize seedlings by suppression subtractive hybridization

污秽颗粒在复合绝缘子表面的沉积会影响其电气特性,导致沿面放电甚至污秽闪络的发生,从微观的角度探究其沉积机理,能够为电力系统的稳定运行提供理论依据。文中基于JKR接触力学理论,通过传统牛顿力学法和离散元方法分别对颗粒碰撞过程中的受力情况进行分析,提出综合考虑法向碰撞、颗粒反弹速度的碰撞沉积模型。根据不同碰撞沉积模型进行仿真实验,将所得结果与现有实验结果进行比较。结果表明:对比牛顿力学方法,离散元方法得到的碰撞沉积模型更能准确预测SiO_(2)颗粒在复合绝缘子表面的沉积情况。绝缘子表面颗粒沉积数随粒径的增大呈先增大后减小的趋势,在粒径20μm时达到最大沉积数。改变风向,绝缘子串中压端与高压端NSDD的比值随风向角度的增大而减小,下表面与上表面NSDD的比值随角度的增大而减小。绝缘子表面颗粒沉积数随风速的增大呈先增大后减小的趋势,并在±30º时达到最大。该文模型能够更为准确的描述污秽颗粒在绝缘子表面的沉积过程,研究结果有助于完善复合绝缘子表面污秽沉积机理。

Cadmium and copper uptake and accumulation by Sesbania rostrata seedling, a N-fixing annual plant: implications for the mechanism of heavy metal tolerance

Comparisons in subcellular and biochemical behaviors of cadmium between Iow-Cd and high-Cd accumulation cultivars of pakchoi （Brassica chinensis L.）

Subcellular distributions and chemical forms of cadmium （Cd） in the leaves, stems and roots were investigated in low-Cd accumulation cultivars and high-Cd accumulation cultivars ofpakchoi （Brassica chinensis L.）. Root cell wall played a key role in limiting soil Cd from entering the protoplast, especially in the low-Cd cultivars. The high-Cd cultivars had significantly higher leaf and stem Cd concentrations than the low-Cd cultivars in cell wall fraction, chloroplast/trophoplast fraction, organelle fraction and soluble fraction. In low-Cd cultivars, which were more sensitive and thus had greater physiological needs of Cd detoxification than high-Cd cultivars, leaf vacuole sequestrated higher proportions of Cd. Cd in the form of pectate/protein complexes （extracted by 1 tool. L~ NaC1） played a decisive role in Cd translocation from root to shoot, which might be one of the mechanisms that led to the differences in shoot Cd accumulation between the two types of cultivars. Furthermore, the formation of Cd- phosphate complexes （extracted by 2% HAc） was also involved in Cd detoxification within the roots of pakchoi under high Cd stress, suggesting that the mechanisms of Cd detoxification might be different between low- and high-Cd cultivars.
