Identification of deregulated miRNAs and their targets in hepatitis B virus-associated hepatocellular carcinoma

AIM： TO identify the differentially expressed miRNAs and their targets in hepatitis B virus （HBV）-associated hepatocellular carcinoma （HCC）. METHODS： Six hundred and sixty seven human miRNAs were quantitatively analyzed by Taqman lowdensity miRNA array （TLDA） in HBV-HCC tissues. Gene ontology （GO） and Kyoto Encyclopedia of Genes and Genomes （KEGG） pathway analyses were used to analyze the significant function and pathway of the differentially expressed miRNAs in HBV-HCC. TargetScan software was used to predict the targets of deregulated miRNAs. Western blotting and luciferase assay were performed to verify the targets of these ***： Ten up-regulated miRNAs （miR-217, miR- 518b, miR-517c, miR-520g, miR-519a, miR-522, miR- 518e, miR-525-3p, miR-512-3p, and miR-518a-3p） and 11 down-regulated miRNAs （miR-138, miR-214, miR-214#, miR-199a-5p, miR-433, miR-511, miR-592, miR-483-3p, miR-483-5p, miR-708 and miR-1275） were identified by Taqman miRNAs array and confirmed quantitatively by reverse transcription polymerase chain reaction in HCC and adjacent non-tumor tissues. GO and KEGG pathway analysis revealed that ＂regulation of actin cytoskeleton＂ and ＂pathway in cancer＂ are most likely to play critical roles in HCC tumorigenesis. MiR- 519a and ribosomal protein S6 kinase polypeptide 3 （RPS6KA3） were predicted as the most significant can-didates by miRNA-mRNA network. In addition, cyclin D3 （CCND3） and clathrin heavy chain （CHC）, usually up-regulated in HCC tissues, were validated as the di- rect target of miR-138 and miR-199a-5p, respectively.

Mining Design Pattern Use Scenarios and Related Design Pattern Pairs:A Case Study on Online Posts

In common design pattern collections,e.g.,design pattern books,design patterns are documented with templates that consist of multiple attributes,such as intent,structure,and sample *** adapt to modern developers,the depictions of design patterns,especially some specific attributes,should advance with the current programming technologies,for example,“known uses”,which exemplifies the use scenarios of design patterns in practice,and“related patterns”,which describes the relatedness between a design pattern and the others within a ***,it is not easy to update the contents of these attributes manually due to the diversity of the programming *** address this problem,in this work,we conducted a case study to mine design pattern use scenarios and related design pattern pairs from Stack Overflow posts to enrich the two *** first extracted the question posts relevant to each design pattern by identifying the design pattern ***,the topics of the posts were discovered by applying topic modeling ***,by analyzing the topics specified for each design pattern,we detected 195 design pattern use scenarios and 70 related design pattern pairs,involving 61 design patterns *** findings are associated with a variety of popular software frameworks and programming *** could complement the existing design pattern collections and help developers better acknowledge the usage and relatedness of design patterns in today's programming practice.

The internal kink mode with a real frequency comparable with the bulk ion diamagnetic frequency is investigated in the case of lower hybrid wave *** is found that circulating supra-thermal electrons can directly destabilize the internal kink mode by resonance interaction in the neighborhood of the q =1 rational *** pressure gradient of circulating supra-thermal electrons on the resonance surface is crucial for excitation of the mode.

Exact Solitary-wave Solutions and Periodic Wave Solutions for Generalized Modified Boussinesq Equation and the Effect of Wave Velocity on Wave Shape

By means of the undetermined assumption method, we obtain some new exact solitary-wave solutions with hyperbolic secant function fractional form and periodic wave solutions with cosine function form for the generalized modified Boussinesq equation. We also discuss the boundedness of these solutions. More over, we study the correlative characteristic of the solitary-wave solutions and the periodic wave solutions along with the travelling wave velocity＇s variation.

Low intensity ultrasound-induced apoptosis in human gastric carcinoma cells

AIM： To investigate the low intensity ultrasound （US）- induced apoptosis in human gastric carcinoma cells and its potential mechanism and to suggest a new therapeutic approach to gastric carcinoma. METHODS： Human SGC-7901 gastric carcinoma cells were cultured in vitro and irradiated by low intensity US for 10 min at different intensities with different incubation times after irradiation. Morphologic changes were examined under microscope with trypan blue staining and then the percentage of early apoptotic cells was detected by flow cytometry （FCM） with double staining of fiuorescein isothiocyanate （FITC）- Annexin V/propidium iodide （PI）. Two-dimensional electrophoresis （2DE） was used to get the protein profile and some proteins differently expressed after US irradiation were identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry （MALDI-TOF-MS）. Functional analysis was performed to investigate the mechanism of US-induced cell apoptosis. RESULTS： The percentage of apoptotic cells increased about 10% after US irradiation （12.0 W/cm^2, 12 h culture）, The percentage of early apoptosis and secondary necrosis in the US-irradiated cells increased with the increased US intensity. Moreover, apoptotic cells increased with the increased culture time after US irradiation and reached its maximum at about 12 *** new proteins appeared after US irradiation and were up or down regulated more than 2 times. Some heat shock proteins （HSPs） were found to be associated with the signal process simulating the apoptosis of cells. CONCLUSION： Low intensity US could induce apoptosis in human gastric carcinoma cells. US-induced apoptosis is related to US intensity/culture time. US-induced apoptosis may be caspases-dependent and endoplasmic reticulum （ER） stress-triggered apoptosis may also contribute to it. Proteomic experimental system is useful in finding the protein alteration in carcinoma cells after US irradiation, helping to develop a new cancer

Pectate lyase is a factor in the adaptability for Heterodera glycines infecting tobacco

The soybean cyst nematode, Heterodeara glycines, is a serious pathogen of soybean, and reported to be the host of a wide range of Fabaceae. In the present study, the host specificity and reproductivity of two populations of H. glycines collected from soybean and tobacco were identified and characterized. The comparative identity between β-1,4-endoglucanase, pectate lyase and chorismate mutase of H. glycines parasitizing on soybean and tobacco were 99, 97 and 98%, respectively. The qR T-PCR analysis indicated that the expression of pectate lyase 2 gene was significantly higher in second-stage juveniles of H. glycines Henan population parasitizing on tobacco than that of H. glycines Shanxi population parasitizing on soybean. In addition, the pectic acid content of cell wall was significantly higher(45%) in the roots of tobacco than the roots of soybean. Our results indicate that the changes in transcript parasitism genes may be a result of long-term evolution illustrating how a plant-parasitic nematode adapts to the host environment for optimal infestation and survival.

Protein profile of human hepatocarcinoma cell line SMMC-7721:Identification and functional analysis

AIM： TO investigate the protein profile of human hepatocarcinoma cell line SMMC-7721, to analyze the specific functions of abundant expressed proteins in the processes of hepatocarcinoma genesis, growth and metastasis, to identify the hepatocarcinoma-specific biomarkers for the early prediction in diagnosis, and to explore the new drug targets for liver cancer therapy. METHODS： Total proteins from human hepatocarcinoma cell line SMMC-7721 were separated by two-dimensional electrophoresis （2DE）. The silver-stained gel was analyzed by 2DE software Image Master 2D Elite. Interesting protein spots were identified by peptide mass fingerprinting based on matrix-assisted laser desorption/ ionization time-of-flight mass spectrometry （MALDI-TOF-MS） and database searching. RESULTS： We obtained protein profile of human hepatocarcinoma cell line SMMC-7721. Among the twenty-one successfully identified proteins, mitofilin, endoplasmic reticulum protein ERp29, ubiquinol-cytochrome C reductase complex core protein I, peroxisomal enoyl CoA hydratase, peroxiredoxin-4 and probable 3-oxoacid CoA transferase 1 precursor were the six novel proteins identified in human hepatocarcinoma cells or tissues. Specific functions of the identified heat-shock proteins were analyzed in detail, and the results suggested that these proteins might promote tumorigenesis via inhibiting cell death induced by several cancer-related stresses or via inhibiting apoptosis at multiple points in the apoptotic signal pathway. Other identified chaperones and cancer-related proteins were also analyzed.CONCLUSION： Based on the protein profile of SMMC-7721 cells, functional analysis suggests that the identified chaperones and cancer-related proteins have their own pathways to contribute to the tumorigenesis, tumor growth and metastasis of liver cancer. Furthermore, proteomic analysis is indicated to be feasible in the cancer study.

Time-delay Compensation Method for Networked Control System Based on Time-delay Prediction and Implicit PIGPC

A network time-delay compensation method based on time-delay prediction and implicit proportional-integral-based generalized predictive controller(PIGPC) is proposed. The least squares support vector machine(LSSVM) is used to predict the current time-delay, the parameters of the least squares support vector machine are optimized by particle swarm optimization(PSO) algorithm,and the predicted time-delay is used instead of the actual time-delay as the parameters of the network time-delay compensation controller. In order to improve the compensation effect of implicit generalized predictive controller(GPC), this paper puts forward an implicit generalized predictive control algorithm with proportional-integral-based(PI) structure and designs the controller based on implicit PIGPC. Through the simulation results, the effectiveness of this design in the paper is verified.
