RING-box proteins regulate leaf senescence and stomatal closure via repression of ABA transporter gene ABCG40

Plant hormone abscisic acid(ABA)plays an indispensable role in the control of leaf senescence,during which ABA signaling depends on its biosynthesis.Nevertheless,the role of ABA transport in leaf senescence remains unknown.Here,we identified two novel RING-box protein-encoding genes UBIQUITIN LIGASE of SENESCENCE 1 and2(ULS1 and ULS2)involved in leaf senescence.Lack of ULS1 and ULS2 accelerates leaf senescence,which is specifically promoted by ABA treatment.Furthermore,the expression of senescence-related genes is significantly affected in mature leaves of uls1/uls2 double mutant(versus wild type(WT))in an ABA-dependent manner,and the ABA content is substantially increased.ULS1 and ULS2 are mainly expressed in the guard cells and aging leaves,and the expression is induced by ABA.Further RNA-seq and quantitative proteomics of ubiquitination reveal that ABA transporter ABCG40 is highly expressed in uls1/uls2 mutant versus WT,though it is not the direct target of ULS1/2.Finally,we show that the acceleration of leaf senescence,the increase of leaf ABA content,and the promotion of stomatal closure in uls1/usl2 mutant are suppressed by abcg40 loss-of-function mutation.These results indicate that ULS1 and ULS2 function in feedback inhibition of ABCG40-dependent ABA transport during ABA-induced leaf senescence and stomatal closure.

Flexible Polydimethylsiloxane Composite with Multi-Scale Conductive Network for Ultra-Strong Electromagnetic Interference Protection

Highly conductive polymer composites(CPCs) with excellent mechanical flexibility are ideal materials for designing excellent electromagnetic interference(EMI) shielding materials,which can be used for the electromagnetic interference protection of flexible electronic devices.It is extremely urgent to fabricate ultra-strong EMI shielding CPCs with efficient conductive networks.In this paper,a novel silver-plated polylactide short fiber(Ag@PL ASF,AAF) was fabricated and was integrated with carbon nanotubes(CNT) to construct a multi-scale conductive network in polydimethylsiloxane(PDMS) matrix.The multi-scale conductive network endowed the flexible PDMS/AAF/CNT composite with excellent electrical conductivity of 440 S m-1and ultra-strong EMI shielding effectiveness(EMI SE) of up to 113 dB,containing only 5.0 vol% of AAF and 3.0 vol% of CNT(11.1wt% conductive filler content).Due to its excellent flexibility,the composite still showed 94% and 90% retention rates of EMI SE even after subjected to a simulated aging strategy(60℃ for 7 days) and 10,000 bending-releasing cycles.This strategy provides an important guidance for designing excellent EMI shielding materials to protect the workspace,environment and sensitive circuits against radiation for flexible electronic devices.

Evaluation and structural analysis of the functions of the Tibetan Plateau National Park Cluster

National parks are useful entities for looking at coordinated efforts to improve the Tibetan Plateau’s function as an ecological safety barrier and the region’s green development.Research on the characteristics of the function structures of the Tibetan Plateau’s national parks is vital to promoting their systematic and coordinated development.This paper combines the pressure-state-response model,the rank-size rule and a coupling and coordination model to identify and evaluate the functions of national parks on the Tibetan Plateau and to analyze the categories,hierarchy and structures behind those functions.The results indicate the following:(1)The Tibetan Plateau National Park Cluster needs to maintain internal and external relations.Internally it needs to rationally allocate resources between ecological protection,recreation and community development,and externally it needs to promote its role as an ecological security barrier and promote regional green development by rationally ranking and organizing the individual national parks,so as to handle their co-evolution of functions at multiple scales.(2)Ecological protection,recreation and community development are the most prominent functions of the Tibetan Plateau National Park Cluster,but there is scope to develop their scientific research and education functions.The Zipf index shows that their multi-functional level conforms with the rank-size rule,indicating balanced development.Individual national parks need to strengthen their optimal functions according to their characteristic localization.(3)The degree of coupling between the functions of the Tibetan Plateau National Park Cluster is 0.7809,and the degree of coordination is 0.6227,which indicates a very strong coupling and moderate coordination.The coupling strength and degree of coordination between the multiple functions vary greatly among the individual national parks,which reflects their different function structures.There are four function structure types:fully coordinated,optimally developed,moderately

Plant high-affinity K^(+) transporters(HKTs)mediate Na^(+) and K^(+) uptake,maintain Na^(+)/K^(+) homeostasis,and therefore play crucial roles in plant salt tolerance.In this study,we present cryoelectron microscopy structures of HKTs from two classes,classI HKT1;1 from Arabidopsis thaliana(AtHKT1;1)and class II HKT2;1 from Triticum aestivum(TaHKT2;1),in both Na^(+) -and K^(+) -bound states at 2.6-to 3.0-A resolutions.BothAtHKT1;1and TaHKT2;1function ashomodimers.Each HKT subunit consists of four tan-dem domain units(D1-D4)with a repeated K^(+) -channel-like M-P-M topology.In each subunit,D1-D4 assemble into an ion conduction pore with a pseudo-four-fold symmetry.Although both TaHKT2;1and AtHKT1;1 have only one putative Na^(+) ion bound in the selectivity filter with a similar coordination pattern,the two HKTs display different K^(+) binding modes in the filter.TaHKT2;1 has three K^(+) ions bound in the selec-tivity filter,but AtHKT1;1 has only two K^(+) ions bound in the filter,which has a narrowed external entrance due to the presence of a Ser residue in the first filter motif.These structures,along with compu-tational,mutational,and electrophysiological analyses,enable us to pinpoint key residues that are critical for the ion selectivity of HKTs.The findings provide new insights into the ion selectivity and ion transport mechanisms of plant HKTs and improve our understanding about how HKTs mediate plant salt tolerance and enhance crop growth.

Estimating the Predictability Limit of Tropical Cyclone Tracks over the Western North Pacific Using Observational Data

In this study,the nonlinear local Lyapunov exponent(NLLE)approach was used to quantitatively determine the predictability limit of tropical cyclone(TC)tracks based on observed TC track data obtained from the Joint Typhoon Warning Center.The results show that the predictability limit of all TC tracks over the whole western North Pacific(WNP)basin is about 102 h,and the average lifetime of all TC tracks is about 174 h.The predictability limits of the TC tracks for short-,medium-,and long-lived TCs are approximately 72 h,120 h,and 132 h,respectively.The predictability limit of the TC tracks depends on the TC genesis location,lifetime,and intensity,and further analysis indicated that these three metrics are closely related.The more intense and longer-lived TCs tend to be generated on the eastern side of the WNP(EWNP),whereas the weaker and shorter-lived TCs tend to form in the west of the WNP(WWNP)and the South China Sea(SCS).The relatively stronger and longer-lived TCs,which are generated mainly in the EWNP,have a longer travel time before they curve northeastwards and hence tend to be more predictable than the relatively weaker and shorter-lived TCs that form in the WWNP region and SCS.Furthermore,the results show that the predictability limit of the TC tracks obtained from the best-track data may be underestimated due to the relatively short observational records currently available.Further work is needed,employing a numerical model to assess the predictability of TC tracks.

Ras homolog(Rho)-associated kinases(ROCKs)belong to the serine-threonine kinase family,which plays a pivotal role in regulating the damage,survival,axon guidance,and regeneration of neurons.ROCKs are also involved in the biological effects of immune cells and glial cells,as well as the development of neurodegenerative disorders such as Alzheimer’s disease,Parkinson’s disease,and multiple sclerosis.Previous studies by us and others confirmed that ROCKs inhibitors attenuated the symptoms and progression of experimental models of the abovementioned neurodegenerative diseases by inhibiting neuroinflammation,regulating immune imbalance,repairing the blood-brain barrier,and promoting nerve repair and myelin regeneration.Fasudil,the first ROCKs inhibitor to be used clinically,has a good therapeutic effect on neurodegenerative diseases.Fasudil increases the activity of neural stem cells and mesenchymal stem cells,thus optimizing cell therapy.This review will systematically describe,for the first time,the effects of abnormal activation of ROCKs on T cells,B cells,microglia,astrocytes,oligodendrocytes,and pericytes in neurodegenerative diseases of the central nervous system,summarize the therapeutic potential of fasudil in several experimental models of neurodegenerative diseases,and clarify the possible cellular and molecular mechanisms of ROCKs inhibition.This review also proposes that fasudil is a novel potential treatment,especially in combination with cell-based therapy.Findings from this review add support for further investigation of ROCKs and its inhibitor fasudil for the treatment of neurodegenerative diseases.

Charge effects on quinoline hydrodenitrogenation catalyzed by Ni-Mo-S active sites-A theoretical study by DFT calculation

The charge distribution on Ni-Mo-S active sites can affect hydrodenitrogenation(HDN)activity.In this study,a series of model Ni-Mo-S were developed with various charge distributions.For comparison,the charge distribution effects on quinoline HDN were studied.The results show that a lack of electrons and extra protons can both lower the orbital eigenvalue of the Ni-Mo-S,leading to stronger adsorption of nitrogen-containing compounds and inhibition of ammonia desorption.Electron deficiency will improve the generation of active hydrogen on the active sites but inhibit hydrogen transfer to the nitrogen compounds;extra protons can provide H^(+)to the nitrogen compounds,which will flexibly transfer between the nitrogen compound and active sites,thus improving the cleavage of the C-N bond.

A WRKY transcription factor confers aluminum tolerance via regulation of cell wall modifying genes

Modification of cell wall properties has been considered as one of the determinants that confer aluminum(Al)tolerance in plants,while how cell wall modifying processes are regulated remains elusive.Here,we present a WRKY transcription factor WRKY47 involved in Al tolerance and root growth.Lack of WRKY47 significantly reduces,while overexpression of it increases Al tolerance.We show that lack of WRKY47 substantially affects subcellular Al distribution in the root,with Al content decreased in apoplast and increased in symplast,which is attributed to the reduced cell wall Al-binding capacity conferred by the decreased content of hemicellulose I in the wrky47-1 mutant.Based on microarray,real time-quantitative polymerase chain reaction and chromatin immunoprecipitation assays,we further show that WRKY47 directly regulates the expression of EXTENSIN-LIKE PROTEIN(ELP)and XYLOGLUCAN ENDOTRANSGLUCOSYLASE-HYDROLASES17(XTH17)responsible for cell wall modification.Increasing the expression of ELP and XTH17 rescued Al tolerance as well as root growth in wrky47-1 mutant.In summary,our results demonstrate that WRKY47 is required for root growth under both normal and Al stress conditions via direct regulation of cell wall modification genes,and that the balance of Al distribution between root apoplast and symplast conferred by WRKY47 is important for Al tolerance.

Activation of the NOD-like receptor protein 3(NLRP3)inflammasome signaling pathway is an important mechanism underlying myocardial pyroptosis and plays an important role in inflammatory damage to myocardial tissue in patients with cardiovascular diseases(CVDs),such as diabetic cardiomyopathy,ischemia/reperfusion injury,myocardial infarction,heart failure and hypertension.Noncoding RNAs(nc RNAs)are important regulatory factors.Many Chinese medicine(CM)compounds,including their effective components,can regulate pyroptosis and exert myocardium-protecting effects.The mechanisms underlying this protection include inhibition of inflammasome protein expression,Toll-like receptor 4–NF-κB signal pathway activation,oxidative stress,endoplasmic reticulum stress(ERS),and mixed lineage kinase 3 expression and the regulation of silent information regulator 1.The NLRP3 protein is an important regulatory target for CVD prevention and treatment with CM.Exploring the effects of the interventions mediated by CM and the related mechanisms provides new ideas and perspectives for CVD prevention and treatment.

An eukaryotic translation initiation factor, AteIF_5A-2,affects cadmium accumulation and sensitivity in Arabidopsis

Cadmium (Cd) is one of the most toxic elements and can be accumulated in plants easily; meanwhile, eIF5A is a highly conserved protein in all eukaryotic organisms. The present work tried to investigate whether eIF5A is involved in Cd accumulation and sensitivity in Arabidopsis (Arabidopsis thaliana L.) by comparing the wild-type Columbia-0 (Col-0) with a knockdown mutant of AteIF5A-2, fbr12-3 under Cd stress conditions. The results showed that the mutant fbr12-3 accumulated more Cd in roots and shoots and had significantly lower chlorophyll content, shorter root length, and smaller biomass, suggesting that downregulation of AteIF5A-2 makes the mutant more Cd sensitive. Real-time polymerase chain reaction revealed that the expressions of metal transporters involved in Cd uptake and translocation including IRT1, ZIP1, AtNramp3, and AtHMA4 were significantly increased but the expressions of PCS1 and PCS2 related to Cd detoxification were decreased notably in fbr12-3 compared with Col-0. As a result, an increase in MDA and H2O2 content but decrease in root trolox, glutathione and proline content under Cd stress was observed, indicating that a severer oxidative stress occurs in the mutant. All these results demonstrated for the first time that AteIF5A influences Cd sensitivity by affecting Cd uptake, accumulation, and detoxification in Arabidopsis.
